The study of cell division processes is important for the understanding of how genetic information is transmitted from cell to cell and to descendants. To observe the phases of mitosis in meristematic cells of the onion root (Allium cepa), it is necessary to induce the rooting of this bulb that can be influenced by some factors like the type of water and the preparation form of the bulb of the onion for the success of rhizogenesis. In view of the above, the objective was to evaluate the influence of the water source and the opening of holes in the stem of the propagule to induce rooting in a hydroponic system. Two experiments were installed. In the first, one of the treatments was used distilled water and treated water. In the other experiment, three treatments were evaluated (without holes, few holes and a lot of holes in the stem of the bulb). Five days after installation of the first experiment, data were collected on the number of bulbs that gave off roots and this variable was analyzed using the X2 test. In the other test, also at five days, root number and fresh root mass data were obtained and the data were submitted for analysis of variance and the means were compared by the Scott-Knott test. For the induction of rooting in onion bulbs (Allium cepa) in a hydroponic system, water should be used, preferably treated, cleaned and decontaminated. The stem of the onion bulb should receive many perforations to facilitate the absorption of water with the essential nutrients for rooting.

At the ultrastructural level, some of the chromatin-containing lacunar spaces of the interphase nucleolus in root meristematic cells of Allium cepa are seen to be walled off, on one side, by dense-fibrillar material and to be contiguous, on the other side, to electron-transparent areas, of variable sizes and shapes, bordered by dense-granular material continuous with and indistinguishable from the dense-granular component of the nucleolar mass. These electron-transparent areas associated with the lacunar spaces are equated with nucleolar vacuoles since they contain scattered preribosomal-like granules and fibrils and are rimmed by dense-granular material. The relevant observational evidence would be consistent with the view that loops of transcriptionnally active chromatin emanating from the nucleolar organizing region project radially into either only the dense-fibrillar or both the dense-fibrillar and the interior of the electron-transparent vacuolar areas seen to be contiguous to the lacunar spaces in question. In relation to this problem, it is of interest to note that the vacuolar spaces of the interphase nucleolus in Allium cepa occasionally display within their confines discrete masses of fibrillar material, possibly chromatinic in character, and in various states of condensation and configuration.

ABSTRACT Water availability in the Brazilian semiarid is restricted and often the only water source available has high salt concentrations. Hydroponics allows using these waters for production of various crops, including vegetables, however, the water salinity can cause nutritional disorders. Thus, two experiments were conducted in a greenhouse at the Department of Agricultural Engineering of the Federal Rural University of Pernambuco, to evaluate the effects of salinity on the mineral nutrition of crisphead lettuce, cultivar Taina, in a hydroponic system (Nutrient Film Technique), using brackish water in the nutrient solution, which was prepared by adding NaCl to the local water (0.2 dS m-1). A randomized blocks experimental design was used in both experiments. The treatments consisted of water of different salinity levels (0.2, 1.2, 2.2, 3.2, 4.2 and 5.2 dS m-1) with four replications, totaling 24 plots for each experiment. The water added to compensate for the water - depth loss due to evapotranspiration (WCET) was the brackish water of each treatment in Experiment I and the local water without modifications in Experiment II. The increase in the salinity of the water used for the nutrient solution preparation reduced the foliar phosphorus and potassium contents and increased the chloride and sodium contents, regardless of the WCET. Foliar nitrogen, calcium, magnesium and sulfur contents were not affected by increasing the water salinity used for the nutrient solution preparation.

Numerous vesicles were observed among the microtubules of the “preprophase” band in prophase cells from root tips of Allium cepa. The content of these vesicles looks similar to the matrix of adjacent cell walls, and these vesicles often appear to be involved in exocytosis. In addition, the cell walls perpendicular to the plane of (beneath) the preprophase band are often differentially thickened compared to the walls lying parallel to the plane of the band. Our interpretation of these observations is that the preprophase band may direct or channel vesicles containing precursors of the cell wall to localized regions of wall synthesis. The incorporation of constituents of the cell wall into a narrow region defined by the position of the preprophase band may be a mechanism that ensures unidirecitonal growth of meristematic cells.

Even though the Lavandula species may be propagated by seeds, it should not be the preferred propagation method because it causes a great lack of uniformity. On the other hand, asexually propagated lavender crops would provide more homogeneous crops, and clones from high quality plant material would increase the odds for obtaining a higher quality essential oil. However, problems such as poor rooting and restrict market availability for superior clones have been a problem in vegetative propagation of the Lavandula species. The objective of this work was to define which type and size of cutting is more adequate for cutting propagation of L. dentata, a very productive Lavandula species. Cuttings with 5, 8, 10 or 13 cm and from the apical or basal parts of stems cut from L. dentata stock plants were placed in Plantmax HT® filled polystyrene foam trays and kept under intermittent mist system for two months. Averages of root number, length of the longest root, fresh and dry root weight, and percentage of rooting were evaluated. Apical cuttings combined 97.9% rooting with an average of 13.2 roots per cutting and basal cuttings 93.7% rooting with 2.98 roots per cutting. Apical cuttings with at least 10 cm in length were considered the most adequate for cutting propagation of L. dentata.

We investigated the influence of extract from <em>Taxus baccata</em> var. Elegantissima (TbE) shoots in 1:8 dilution, containing paclitaxel in concentration of 81,6 µg/g fresh mass on ultrastructure and tubulin cytoskeleton of meristematic cells of <em>Allium cepa</em> L. root tips. Incubation time 3, 6, 12 and 24 h was followed with postincubation in water for 12 and 24 h. During shorter incubation (till 12 h) the surface of the cell nuclei decreased and chromatin became condensed (in comparison to control) but after 24 h the average surface increased and chromatin condensation decreased. In the course of incubation the average size of plastids and vacuoles increased. Moreover, after treatment mitochondria and plastids showed degradation of ultrastructure, which was reversed after 12 h of postincubation. Immunocytochemical assays demonstrated that in the course of incubation in the ThE extract, the tubulin cytoskeleton became partially disorganised. In most interphase cells, cortical microtubules (MTs) lost their oval transverse orientation. The preprophase band (PPB) position in the cell was often asymmetrical. The MTs array of the karyokinetic spindle and phragmoplast was also disturbed. These alterations were completely reversed during postincubation.

The effect of pure sodium cacodylate on dividing cells was studied. The root meristematic cells of <em>Allium cepa</em> L. (the roots were squashed in acetoorcein) and endosperm cells of <em>Haemanthus katherinae</em> Bak. (<em>in vitro</em> observations) were used. Serious disturbances in karyokinesis and cytokinesis were found that led most often to the formation of polyploid or multinucleate (<em>A. cepa</em>) cells. These results point to damage of the mitotic spindle and phragmoplast. Careful use of cacodylate buffer in ultrastructural studies of microtubules is advised.

Purpose: research and development of design solutions for drainage and watering systems using linear and frontal-circular movement sprinkling machines for the effective use of system water resources when draining and moistening reclaimed lands in non-sloping (low-slope) areas and with a slight slope. Materials and methods. During the research, the well-known design solutions of drainage and watering, irrigation and water circulation systems of various scientists were analyzed. In the course of analysis of constructive solutions and methods of movement, analytical, comparative and logical methods were used. When developing (improving) design solutions, the design method was used. Results. During the research, it was found that in the humid zone, due to the decrease in irrigation norms, there is a possibility of increasing the seasonal load of sprinklers. Based on the results of studies of the principles of moving sprinkler machines and currently used design solutions in order to reduce energy consumption and increase the land use coefficient, three design solutions have been proposed for reclaimed lands with different slopes. It is proposed to use frontal and frontal-circular sprinklers in drainage and watering systems. Humidification in these solutions is carried out by accumulated drainage runoff with the possibility of discharging its excess water into a water intake-water source, as well as, additional withdrawal from it in case of necessity. Conclusions. As a result of the research, the constructive solutions for drainage and watering systems have been proposed using frontal-circular and frontal sprinklers on non-slope (low-slope) reclaimed lands, as well as those with a slope, which make it possible to reduce the number of sprinklers due to reduced irrigation norms and the location of the combined drainage-watering and watering networks.
