This study intends to elucidate MiRNA-335’s role in hepatoma cell lines (HCC). Real-time PCR was used to detect MiRNA-335 expression in HCC, flow cytometry and MTT were used to detect apoptosis and proliferation. Luciferase reporting system analyzed the targeting relationship between Foxo3a and MiRNA-335. HCC (SMMC7721 cell) exhibited significantly reduced MiRNA-335 compared to normal hepatocyte cell (HL7702). MiRNA-335 mimic inhibited HCC proliferation and enhanced apoptosis, which were reversed by MiRNA-335 inhibitor. Luciferase reporter gene system showed that MiRNA-335 significantly inhibited the fluorescent activity of Foxo3a 3′-UTR, indicating that MiRNA-335 could target Foxo3a RNA. In conclusion, the decrease of MiRNA-335 can promote the proliferation of hepatoma cells and inhibit apoptosis possibly through regulating Foxo3a, which provides a new direction for the treatment of liver cancer.

Abstract Catches reports by set longlines fishing vessels in the 718-fisheries management area (FMA) have been entered into the logbook reporting system since 2012. These vessels targeted reef and demersal fish. Set longlines is considered to be very efficient and productive for catching reef fish. This study aims to see the performance of set longlines vessels. The indicators used are CPUE and fuel efficiency. The results of the study show that the performance of set longlines vessels in the area is getting better and more efficient. Keywords: set longlines; reef fish; CPUE; fuel; efficient   Abstrak Laporan hasil tangkapan oleh kapal penangkap rawai dasar di pengelolaan perikanan (WPP) 718 telah dimasukkan ke dalam sistem pelaporan logbook sejak 2012. Kapal-kapal rawai tesebut menargetkan ikan karang dan ikan demersal. Rawai dasar dianggap sebagai alat tangkap yang sangat efisien dan produktif untuk menangkap ikan karang. Penelitian ini bertujuan untuk melihat kinerja kapal rawai dasar. Indikator yang digunakan adalah CPUE dan efisiensi bahan bakar. Hasil penelitian menunjukkan bahwa kinerja kapal longline set di daerah tersebut semakin baik dan efisien. Kata kunci: rawai dasar; ikan karang; CPUE; bahan bakar; efisien

Abstract Introduction: Japan recently experienced two major heavy rain disasters: the West Japan heavy rain disaster in July 2018 and the Kumamoto heavy rain disaster in July 2020. Between the occurrences of these two disasters, Japan began experiencing the wave of the coronavirus disease 2019 (COVID-19) pandemic, providing a unique opportunity to compare the incidence of acute respiratory infection (ARI) between the two disaster responses under distinct conditions. Sources for Information: The data were collected by using the standard disaster medical reporting system used in Japan, so-called the Japan-Surveillance in Post-Extreme Emergencies and Disasters (J-SPEED), which reports number and types of patients treated by Emergency Medical Teams (EMTs). Data for ARI were extracted from daily aggregated data on the J-SPEED form and the frequency of ARI in two disasters was compared. Observation: Acute respiratory infection in the West Japan heavy rain that occurred in the absence of COVID-19 and in the Kumamoto heavy rain that occurred in the presence of COVID-19 were responsible for 5.4% and 1.2% of the total consultation, respectively (P <.001). Analysis of Observation and Conclusion: Between the occurrence of these two disasters, Japan implemented COVID-19 preventive measures on a personal and organizational level, such as wearing masks, disinfecting hands, maintaining social distance, improving room ventilation, and screening people who entered evacuation centers by using hygiene management checklists. By following the basic prevention measures stated above, ARI can be significantly reduced during a disaster.

Protein fluorescence reporting systems are of crucial importance to in-depth life science research, providing systematic labeling tools for visualization of microscopic biological activities in vivo and revolutionizing basic research. Cotton somatic cell regeneration efficiency is low, causing difficulty in cotton transformation. It is conducive to screening transgenic somatic embryo using the fluorescence reporting system. However, available fluorescence labeling systems in cotton are currently limited. To optimize the fluorescence reporting system of cotton with an expanded range of available fluorescent proteins, we selected 11 fluorescent proteins covering red, green, yellow, and cyan fluorescence colors and expressed them in cotton. Besides mRuby2 and G3GFP, the other nine fluorescent proteins (mCherry, tdTomato, sfGFP, Clover, EYFP, YPet, mVenus, mCerulean, and ECFP) were stably and intensely expressed in transgenic callus and embryo, and inherited in different cotton organs derive from the screened embryo. In addition, transgenic cotton expressing tdTomato appears pink under white light, not only for callus and embryo tissues but also various organs of mature plants, providing a visual marker in the cotton genetic transformation process, accelerating the evaluation of transgenic events. Further, we constructed transgenic cotton expressing mCherry-labeled organelle markers in vivo that cover seven specific subcellular compartments: plasma membrane, endoplasmic reticulum, tonoplast, mitochondrion, plastid, Golgi apparatus, and peroxisome. We also provide a simple and highly efficient strategy to quickly determine the subcellular localization of uncharacterized proteins in cotton cells using organelle markers. Lastly, we built the first cotton stomatal fluorescence reporting system using stomata-specific expression promoters (ProKST1, ProGbSLSP, and ProGC1) to drive Clover expression. The optimized fluorescence labeling system for transgenic somatic embryo screening and functional gene labeling in this study offers the potential to accelerating somatic cell regeneration efficiency and the in vivo monitoring of diverse cellular processes in cotton.

This study compared NIBRS arrest data in a statewide sample with arrest and summons data on the same cases collected directly from law enforcement agencies (LEAs). NIBRS matched LEA data in 84.1% of cases. However, 5.8% of LEA arrests and 52.9% of LEA summons were false negatives, that is, they were incorrectly represented as not cleared by arrest in NIBRS. False negatives were more likely when more than 1 day elapsed between incident and arrest and when the crimes were sexual assault or intimidation. False negatives were less likely in small LEAs (for summons) Recommendations are presented for improving accuracy.
