Here, we developed a rapid, visual and double-checked Logic Gate detection platform for detection of pathogenic microorganisms by aggregation-induced emission luminogens (AIEgens) in combination with Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)/CRISPR associated (Cas). DNA light-up AIEgens (1,1,2,2-tetrakis[4-(2-bromo-ethoxy) phenyl]ethene, TTAPE) was non-emissive but the emission was turned on in the presence of large amount of DNA produced by recombinase polymerase amplification (RPA). When CRISPR/Cas12a was added, all long-stranded DNA were cut leading to the emission quenched. Thus, a method that can directly observe the emission changes with the naked eye has been successfully constructed. The detection is speedy within only 20 min, and has strong specificity to the target. The result can be judged by Logic Gate. Only when the output signal is (1,0), does it represent the presence of pathogenic microorganisms in the test object. Finally, the method was applied to the detect pathogenic microorganisms in environmental water samples, which proved that this method has high selectivity, specificity and applicability for the detection of pathogenic microorganisms in environmental water samples.

Most medical images are low in contrast because adequate details that may prove vital decisions are not visible to the naked eye. Also, due to the low-contrast nature of the image, it is not easily segmented because there is no significant change between the pixel values, which makes the gradient very small Hence, the contour cannot converge on the edges of the object. In this work, we have proposed an ensembled spatial method for image enhancement. In this ensembled approach, we first employed the Laplacian filter, which highlights the areas of fast intensity variation. This filter can determine the sufficient details of an image. The Laplacian filter will also improve those features having shrill disjointedness. Then, the gradient of the image has been determined, which utilizes the surrounding pixels for the weighted convolution operation for noise diminishing. However, in the gradient filter, there is one negative integer in the weighting. The intensity value of the middle pixel might be deducted from the surrounding pixels, to enlarge the difference between the head-to-head pixels for calculating the gradients. This is one of the reasons due to which the gradient filter is not entirely optimistic, which may be calculated in eight directions. Therefore, the averaging filter has been utilized, which is an effective filter for image enhancement. This approach does not rely on the values that are completely diverse from distinctive values in the surrounding due to which it recollects the details of the image. The proposed approach significantly showed the best performance on various images collected in dynamic environments.

Disease control is crucial to minimize potential losses in agriculture and thereby maintain high crop yield. However, for its effectiveness, the pathogen must be detected early and correctly in the production fields. Different methods of diagnosis can be used, from those based on symptoms to molecular tests. Loop-mediated isothermal amplification (LAMP) is a molecular technique that has been widely used in several biological fields, due to the ease with which it can be applied. The reaction can be carried out in a single thermal condition, due to the use of Bst DNA polymerase, isolated from the bacterium Bacillus stearothermophilus, which has high displacement activity. LAMP is a highly exponential amplification method that produces the target DNA in amounts 109 -1010 times between 45 and 60 minutes at 60-65°C. Its advantages are the visualization of results directly with the naked eye and the fact that it does not need sophisticated equipment for its application. In phytopathology, the technique has been gaining prominence in the detection of fungi, viruses, bacteria, nematodes and phytoplasmas, as well as in the monitoring of fungicide-resistant fungi. LAMP can benefit agriculture so that early, accurate and sensitive diagnostics can be carried out in the fields of cultivation and minimize losses caused by diseases. In this review, we present and discuss LAMP tests, developed for plant pathogens detection, which can be useful for researchers who wish to use the technique in their research area

The process of learning online makes the education has the current condition concerning. The problems occur at the Elementary School students in grades 1 and 2 is in the reading introduction stage because not all elementary school students attend kindergarten, but they have to take online schools that cannot meet their teachers in person. The students should be guided by the teacher to learn spelling and counting, could not be realized. The impact for the elementary school students in grades 1 and 2 is cannot read, even they have not memorized the alphabet completely. This is a small thing that is not visible to the naked eye but becomes a big problem in the field of education. The training and guidance for elementary school students can achieve the goal of the concepts of the 2021 KKM program, and  they are not only good at reading, but they are familiar with digital literacy which has developed very rapidly in the globalization era.
