Abstract Background Freshly mixed root canal sealers when proximate the periapical tissues, trigger varying degrees of cytotoxicity/inflammatory reactions. Simvastatin, a class of the drug statin, is a widely used cholesterol-lowering agent with additional anti-inflammatory activities. This study assessed the effects of simvastatin on cytotoxicity and the release of IL-6 (Interleukin-6) production when incorporated in zinc oxide eugenol and methacrylate resin-based sealers. Methods Experimental groups consisted of conventional zinc oxide eugenol and methacrylate based-EndoREZ sealers (ZE & ER respectively) and 0.5 mg/mL simvastatin incorporated sealers (ZES & ERS). L929 mouse fibroblast cells were exposed to freshly mixed experimental sealers and evaluated for cytotoxicity (MTT assay) and inflammation levels (inflammatory marker IL-6 for ELISA) at various time intervals (0h, 24h and 7th day). The values were compared to the cell control (CC; L929 cells alone) and solvent control (SC; L929 cells + DMSO) groups. All the experiments were conducted in triplicates and subjected to statistical analysis using IBM SPSS Statistics software. Non parametric tests were conducted using Kruskal-Wallis and Friedman tests for inter-group and intra-group comparisons respectively. Pairwise comparison was conducted by post hoc Dunn test followed by Bonferroni correction. P values < 0.05 were considered statistically significant. Results All the experimental groups (ZE, ER, ZES, ERS) exhibited varying degree of cytotoxicity and IL-6 expression compared to the control groups CC and SC. The cell viability for ZE and ER decreased on day 7 as compared to 24 h. ZES and ERS had higher viable cells (75.93% & 79.90%) compared to ZE and ER (54.39% & 57.84%) at all time periods. Increased expression of IL-6 was observed in ZE & ER (25.49 pg/mL & 23.14 pg/mL) when compared to simvastatin incorporated ZE & ER (ZES-12.70 pg/mL & ERS-14.68 pg/mL) at all time periods. Highest level of cytotoxicity and inflammation was observed in ZE compared to all the other groups on day 7. Conclusions Addition of 0.5 mg/mL of simvastatin to the sealers (ZES and ERS) decreased the cytotoxicity in the freshly mixed state and reduces their inflammatory effect.

Dry socket is a common postoperative complication secondary to any tooth extraction but is more commonly associated with mandibular third molars. Dry socket has been treated with various treatment modalities with different success rates. This study aimed to evaluate the effect of using honey and zinc oxide eugenol as an intra-socket medicament for the treatment of dry sockets. Ninety patients were divided into three groups as follows: Group 1 “honey”: 30 patients; Group 2 “zinc oxide eugenol”: 30 patients; and Group 3 “control”: 30 patients. Pre-medication and post-medication pain scores at different time intervals were measured on a verbal rating scale. The mean ages of the patients in each of the groups were as follows: Group 1: 44.26 ± 14.14; Group 2: 45.30 ± 18.08; and Group 3: 51.93 ± 9.75. As regards verbal rating scale pain scores, a significant difference was noted in patients that belonged to Groups 1 and 2, with an immediate reduction in post-medication pain scores. However, patients belonging to Group 3 reported pain and discomfort for over a week. The use of honey and zinc oxide eugenol resulted in significant immediate post-medication pain scores in patients as compared to the control group. Therefore, both honey and zinc oxide eugenol can be used as treatment options for dry socket patients.

ln the techniques for obtaining impressions of edentulous mouths with zinc oxide - eugenol impression materials it is common procedure to add newly mixed material to the already hardened impression in order to improve or "correct" the impression. It was our purpose to find out about the retentive or "adhesive" qualities of the above mentioned "additions" to the hardened material. To obtain this information weused a tensile strenght test in which the "added" material remained in the geometrical center of the test samples. As a comparison, test samples were also used, without added material. Five of the most widely used zinc-oxide/eugenol impression materials in Brazil were utilized throughout the experiments. The results, presented through Graphs and Tables, were statistically analysed. The addition of newly mixed material to already hardened material caused a reduction in tensile strenght values in all live impression pastes. This reduction, however, was not significant in paste "C"; it was significant (.05) in pasle "A” and highly significant (.01) in pastes,"B", "D"and "E”.

Thirty patients were examined, selected and individual impression made. After the impressions made, the casts were mounted in a special apparathus the Gnathothensiometer to measure the differences among three different centric relation techniques. Technique 1 - A bite rim, refined with zinc-oxide-eugenol. Technique 2 - A bite rim, refined with zinc-oxide-eugenol plus a jig. Technique 3 - A bite frame with gauze and zinc-oxide-eugenol. According to the measurements, the technique more accurate, for register the centric relations, seem to be the one using the frame of Jones (Technique nº 3).

The long-term effect of Zinc oxide zinc sulfate (Coltosol®) dressing material on pulpotomy success and tooth survival has not yet been studied. This study compared the success rates of Zinc oxide zinc sulfate and zinc oxide eugenol as coronal dressing materials post radicular pulp amputation in primary teeth pulpotomies. This study included healthy two- to ten-year-old children who had pulpotomies on primary molars between 2012 and 2018 at the Pediatric Dentistry Clinic of the School of Dental Medicine. Data were analyzed at several follow-ups of up to 60 months. Kaplan-Meier survival curves were used to estimate survival probabilities of Zinc oxide zinc sulfate versus zinc oxide eugenol. In the 107 children included in this study, 54 teeth were filled with Zinc oxide zinc sulfate and 53 were filled with zinc oxide eugenol. Follow-up ranged from 12.2 to 73.3 months. Overall survival of Coltosol® vs. IRM filled teeth was 87.1% and 79.3%, respectively. Overall survival probabilities for Coltosol®-filled teeth at 15.5, 24 and 45 months were 95%, 89.8% and 79.7%, respectively, while for IRM they were 93.7%, 83% and 67.7%, respectively. Treatment failure rates and type of treated teeth did not differ between boys and girls (p-value = 0.77 and 0.87, respectively). Zinc oxide zinc sulfate and zinc oxide eugenol exhibited comparable high long-term success rates of up to five years (p = 0.16).

Background. The choice of sealant is an important dentist’s decision with a long-term influence on treatment. Knowledge of the properties and characters of each material is key to the optimal endosealer selection on individual basis.Objectives. A comparison of endodontic sealants based on epoxy resins, calcium hydroxide, zinc oxide eugenol and bioceramics.Methods. Publications were mined in the PubMed and Google Scholar electronic databases to cover the four sealer groups (epoxy, calcium hydroxide, zinc oxide eugenol and bioceramics) over years 2014-2021, including selected relevant sources within 2002-2013; 73 articles were considered for review, regardless of the study design or language. Content and descriptive analyses were used as research tools. Meta-analysis was not used due to a high evidence heterogeneity.Results. The dental market currently offers a variety of sealant groups. Endodontic sealants based on epoxy resins, zinc oxide eugenol, calcium hydroxide and bioceramics are the most common. The review identifies no ideal material for the root canal filling.Conclusion. Manifold studies demonstrate the pros and cons in each endodontic sealant group with respect to variant criteria. Depending on clinical situation, the practitioner can opt for the material property to identify the endosealer.

BACKGROUND Zinc oxide eugenol (ZOE) impression paste is commonly used for secondary impression. Water acts as an accelerator to zinc oxide eugenol paste. Study was carried out to determine its flow property and the effect of addition of water on flow properties for different commercially available zinc oxide eugenol impression pastes. We wanted to compare and evaluate the flow properties of ZOE impression paste without water and after addition of water as an accelerator for three different commercially available zinc oxide eugenol impression pastes. METHODS Three commercially available zinc oxide eugenol impression pastes used were - DPI (Dental Product of India), Neogenate (Septodont), IMAGE (Prime Dental Pro.Ltd). A total of 15 discs were made without accelerator and with addition of 1, 2 and 3 drops of water for the three brands of the zinc oxide eugenol paste. Then the flow of each was tested. Data of flow was analysed using one way- ANOVA and post hoc test. RESULTS It was found that the mean flow of zinc oxide eugenol impression paste without the use of accelerator for DPI, Septodont, Image was 4.72 ± 0.56, 4.20 ± 0.75, and 3.50 ± 0.57 respectively. The mean flow of zinc oxide eugenol impression paste with the use of one drop of accelerator for DPI, Septodont, Image was 4.98 ± 0.3493, 4.54 ± 0.6025, and 3.940 ± 0.4722 respectively. With the use of two and three drops of accelerator for DPI was 4.18 ± 0.7259 and 2.68 ± 0.3701, for Septodont was 3.880 ± 0.6907 and 2.32 ± 0.2775 and for Image flow was 3.20 ± 0.5745 and 1.80 ± 0.4000. CONCLUSIONS DPI had the maximum flow as compared to other brands of zinc oxide eugenol impression paste. KEY WORDS Zinc Oxide Eugenol, Impression Material, Flow, and Accelerator
