Chapter 6 summarizes the work described in the book. It places Environmental Biodynamics in context of the broader field of general systems theory. It argues that to realize the full potential of Environmental Biodynamics, environmental medicine must refocus the examination of the interaction of environment and health from an emphasis on measuring physiological “moments” (i.e., static measures of environmental factors, infrequent anthropometry, momentary health indicators) to studying dynamic human–environment interfaces, physiological states, and the processes that constrain to those states. To this end, Chapter 6 provides a set of endeavors that must be undertaken to capitalize on and formally test the biodynamic interface paradigm. First, focus scientific inquiry on interfaces that connect biological and environmental systems; second, develop theoretical frameworks that focus on the identification and interpretation of constraints in biological–environmental interfaces; and third, develop laboratory, clinical, and epidemiological methods to relate the complexity characterized at the level of biodynamic interfaces to human health.

Collaborative, One Health approaches support governments to effectively prevent, detect and respond to emerging health challenges, such as zoonotic diseases, that arise at the human-animal-environmental interfaces. To overcome these challenges, operational and outcome-oriented tools that enable animal health and human health services to work specifically on their collaboration are required. While international capacity and assessment frameworks such as the IHR-MEF (International Health Regulations—Monitoring and Evaluation Framework) and the OIE PVS (Performance of Veterinary Services) Pathway exist, a tool and process that could assess and strengthen the interactions between human and animal health sectors was needed. Through a series of six phased pilots, the IHR-PVS National Bridging Workshop (NBW) method was developed and refined. The NBW process gathers human and animal health stakeholders and follows seven sessions, scheduled across three days. The outputs from each session build towards the next one, following a structured process that goes from gap identification to joint planning of corrective measures. The NBW process allows human and animal health sector representatives to jointly identify actions that support collaboration while advancing evaluation goals identified through the IHR-MEF and the OIE PVS Pathway. By integrating sector-specific and collaborative goals, the NBWs help countries in creating a realistic, concrete and practical joint road map for enhanced compliance to international standards as well as strengthened preparedness and response for health security at the human-animal interface.

In this study, I use microbiome datasets from global soil samples and diverse hosts to learn whether soil microbial taxa are found in host microbiomes, and whether these observations fit the narrative that environmental interaction influences human microbiomes. A major motivation for conducting host-associated microbiome research is to contribute towards understanding how the environment may influence host physiology. The microbial molecular network is considered a key vector by which environmental traits may be transmitted to the host. Research on human evolution seeks evidence that can inform about the living experiences of human ancestors. This objective is substantially enhanced by recent work on ancient biomolecules from preserved microbial tissues, such as dental calculus, faecal sediments and whole coprolites. A challenge yet is to distinguish authentic biomolecules from environmental contaminants deposited contemporaneously, primarily from soil. However, we do not have sound expectations about the soil microbial elements arriving to host-associated microbiomes in a modern context. One assumption in human microbiome research is that proximity to the natural environment should affect biodiversity or impart genetic elements. I present evidence supporting the assumption that environmental soil taxa are found among host-associated gut taxa, which can recapitulate the surrounding host habitat ecotype. Soil taxa found in gut microbiomes relate to a set of universal ‘core’ taxa for all soil ecotypes, demonstrating that widespread host organisms may experience a consistent pattern of external environmental cues, perhaps critical for development. Observed differentiation of soil feature diversity, abundance and composition among human communities, great apes and invertebrate hosts also indicates that lifestyle patterns are inferable from an environmental signal that is retrievable from gut microbiome amplicon data. To prevent and mitigate chemical risks in the petrochemical industry, such as fires and spillage, process safety management (PSM), is essential, especially where flammable, corrosive, explosive, toxic, or otherwise dangerous chemicals are used. We investigated process safety (PS) between man–machine (material equipment) and environmental interfaces by using process hazard analysis (PHA) and fault tree analysis (FTA). By analyzing the data obtained through machinery and mechanical integrity (MI), pre-startup safety review (PSSR), current operating modes, and areal locations of hazardous atmospheres (ALOHA) simulations of the disaster’s aftermath, the cause of the styrene plant accident was found to be the fuel furnace (F101) switching process. Although the furnace had been extinguished, fuel continued to enter the furnace, and it was exposed to a high-temperature surface, resulting in the flashing ignition of the C4 fuel. The plan-do-check-act (PDCA) management model can be used to forestall the system from accident, and it is used to improve the proposal and develop countermeasures that would increase PSM performance and substantially lessen the impact of the thermal hazard. Disasters are often attributable to the unsafe state of machinery, equipment, or the environment, dangerous behaviors of the operator, and the lack of a thorough management system. It is anticipated that the investigation and analysis of the accident would not only find the real cause of the disaster but also lead to the establishment of better effective solutions for common safety problems.

Innate lymphoid cells (ILCs) patrol environmental interfaces to defend against infection and protect barrier integrity. Using a genetic tuning model, we demonstrate that the signal-dependent transcription factor (TF) STAT5 is critical for accumulation of all known ILC subsets in mice and reveal a hierarchy of STAT5 dependency for populating lymphoid and nonlymphoid tissues. We apply transcriptome and genomic distribution analyses to define a STAT5 gene signature in natural killer (NK) cells, the prototypical ILC subset, and provide a systems-based molecular rationale for its key functions downstream of IL-15. We also uncover surprising features of STAT5 behavior, most notably the wholesale redistribution that occurs when NK cells shift from tonic signaling to acute cytokine-driven signaling, and genome-wide coordination with T-bet, another key TF in ILC biology. Collectively, our data position STAT5 as a central node in the TF network that instructs ILC development, homeostasis, and function and provide mechanistic insights on how it works at cellular and molecular levels.
