AbstractThis study aimed to analyze the growth of two types of blood vessel building cells: endothelial cells (ECs) and smooth muscle cells (SMCs) on surfaces with different morphology. Two types of materials, differing in morphology, were produced by the solution blow spinning technique. One-layer materials consisted of one fibrous layer with two fibrous surfaces. Bi-layer materials consisted of one fibrous-solid layer and one fibrous layer, resulting in two different surfaces. Additionally, materials with different average fiber diameters (about 200, 500, and 900 nm) were produced for each group. It has been shown that it is possible to obtain structures with a given morphology by changing the selected process parameters (working distance and polymer solution concentration). Both morphology (solid versus fibrous) and average fiber diameter (submicron fibers versus microfibers) of scaffolds influenced the growth of ECs. However, this effect was only visible after an extended period of culture (6 days). In the case of SMCs, it was proved that the best growth of SMCs is obtained for micron fibers (with an average diameter close to 900 nm) compared to the submicron fibers (with an average diameter below 900 nm).

AbstractSolution blow spinning (SBS) has recently emerged as a novel method that can produce nano- and microfiber structures suitable for tissue engineering. Gelatin is an excellent precursor for SBS as it is derived mainly from collagens that are abundant in natural extracellular matrices. Here we report, for the first time the successful generation of 3D thermally crosslinked preforms by using SBS from porcine gelatin. These SBS mats were shown to have three-dimensional fibrous porous structure similar to that of mammalian tissue extracellular matrix. In pharma industry, there is an urgent need for adequate 3D liver tissue models that could be used in high throughput setting for drug screening and to assess drug induced liver injury. We used SBS mats as culturing substrates for human hepatocytes to create an array of 3D human liver tissue equivalents in 96-well format. The SBS mats were highly cytocompatible, facilitated the induction of hepatocyte specific CYP gene expression in response to common medications, and supported the maintenance of hepatocyte differentiation and polarization status in long term cultures for more than 3 weeks. Together, our results show that SBS-generated gelatin scaffolds are a simple and efficient platform for use in vitro for drug testing applications.

Gelatin (GA) is a natural protein widely used in food packaging, but its fabricated fibrous film has the defects of a high tendency to swell and inferior mechanical properties. In this work, a novel spinning technique, solution blow spinning (SBS), was used for the rapid fabrication of nanofiber materials; meanwhile, nylon 66 (PA66) was used to improve the mechanical properties and the ability to resist dissolution of gelatin films. Morphology observations show that GA/PA66 composite films had nano-diameter from 172.3 to 322.1 nm. Fourier transform infrared spectroscopy and X-ray indicate that GA and PA66 had strong interaction by hydrogen bonding. Mechanical tests show the elongation at break of the composite film increased substantially from 7.98% to 30.36%, and the tensile strength of the composite film increased from 0.03 MPa up to 1.42 MPa, which indicate that the composite films had the highest mechanical strength. Water vapor permeability analysis shows lower water vapor permeability of 9.93 g mm/m2 h kPa, indicates that GA/PA66 film’s water vapor barrier performance was improved. Solvent resistance analysis indicates that PA66 could effectively improve the ability of GA to resist dissolution. This work indicates that SBS has great promise for rapid preparation of nanofibrous film for food packaging, and PA66 can be applied to the modification of gelatin film.

This study attempted to produce polymeric microfibers with low-cost and photocatalytic properties, making it possible to remedy two modern problems, plastic disposal and irregular effluent disposal, for which we used the solution blows pinning (SBS) technique to produce recycled polystyrene (PS) microfibers (recycled waste from transparent barrel pen), the use of the SBS also has good mobility for the benefit of fibers, allowing the fibers to be produced directly under the surface where intend to be used, through the SEM was found the ideal concentration to produce uniform microfibers. With the minor average diameter, FTIR analysis of the fibers showed peak characteristics of PS, demonstrating that most of the transparent barrel pen is composed of PS. The as-prepared fibers of recycled PS were incorporated into their polymer solution with a TiO2 Degussa P25 concentration of 10% (w/w) concerning the polymer mass. For the study of photocatalytic activity, the dye Rhodamine B was used as an indicator. Excellent photocatalytic activity, XRD pattern of PS and PS/TiO2-10% fibers showed PS and TiO2 in two phases, anatase and rutile.
