Heat-stable antifungal factor (HSAF) is produced by the fermentation of Lysobacter enzymogenes, which is known for its broad-spectrum antifungal activity and novel mode of action. However, studies on the separation of HSAF have rarely been reported. Herein, alteramide B (the main byproduct) was removed firstly from the fermentation broth by photodegradation to improve the purity of HSAF. Then, the separation of HSAF via adsorption by macroporous adsorption resins (MARs) was evaluated and NKA resin showed highest static adsorption and desorption performances. After optimizing the static and dynamic adsorption characteristics, the content of HSAF in the purified product increased from 8.67 ± 0.32% (ethyl acetate extraction) to 31.07 ± 1.12% by 3.58-fold. These results suggest that the developed strategy via photodegradation and macroporous resin adsorption is an effective process for the separation of HSAF, and it is also a promising method for the large-scale preparation of HSAF for agricultural applications.

Flavonoids are the main constituents of Goji berries and have good biological and pharmacological activities. The mixed-mode macroporous adsorption resins (MARs) for purification of flavonoids from Goji berries through computer-assisted calculation of the molecular size of flavonoids and the precise matching of MAR physical and chemical properties was firstly developed in the present study. Ten varieties of MARs with suitable molecular dimensions and polarities were used for investigating the adsorption/desorption behaviors of the flavonoids. Both AUKJ-1 and BWKX-1 showed higher separation efficiency than other MARs and then were mixed in different ratios to constitute a mixed-mode macroporous adsorption resin to obtain the optimal adsorption phase. Under optimal conditions, total flavonoid content of purified flavonoid (p-FLA) extract increased from 0.97% to 36.88% after one purification. The p-FLA extract from Goji berries significantly improved the expression of six genes with anti-aging effects and played an important role in aging-related Alzheimer’s disease by down-regulating Aβ expression.

Purpose The purpose of this study is to focus on the preparation of macroporous adsorption resins (MARs) functionalized with carbazole and N-methylimidazole, and adsorption behaviors of (–)-epigallocatechin gallate (EGCG) and caffeine (CAF) on the functionalized MARs. Design/methodology/approach Based on the Friedel–Crafts and amination reactions, novel MARs functionalized with carbazole and N-methylimidazole were synthesized and characterized by the BET method. Accordingly, adsorption behaviors and structure-activity relationships for EGCG and CAF were studied in detail. Findings The results showed that pseudosecond-order kinetic model was provided with a better correlation for the adsorption of EGCG and CAF onto L-1 and L-2, and pseudofirst-order kinetic model was the most suitable model to illustrate the adsorption process for EGCG and CAF on L-3. The result indicated that Langmuir, Freundlich, Temkin–Pyzhev and Dubinin–Radushkevich isotherms all could better illustrate the adsorption processes of EGCG and CAF on L-1, L-2 and L-3. Practical implications This study provides theoretical guidance and technical support for the efficient separation and purification of EGCG and CAF from waste tea leaves by MARs on a large scale. In addition, the results showed that this novel MARs would provide useful help and be used in large-scale production of active ingredients from natural products in the industry and other fields. Originality/value Adsorption kinetic models such as pseudofirst-order, pseudosecond-order and intra-particle diffusion kinetic models, and adsorption isotherm models such as Langmuir, Freundlich, Temkin–Pyzhev and Dubinin–Radushkevich isotherms models were adopted to illustrate the adsorption mechanisms of EGCG and CAF. The main driving forces for MARs with no functional groups were pore sieving effects, p–p conjugation effects and hydrophobic interactions, and the other significant driving forces for MARs functionalized with carbazole and N-methylimidazole were electrostatic interactions, ion-dipole and hydrogen bonding interactions. This study might provide scientific references and useful help for large-scale separating and enriching active ingredients in natural products using the technology of MARs with special functional groups.

Stephania cepharantha Hayata is a traditional Chinese herbal medicine used to treat lung cancer, and its alkaloids, especially cepharanthine (CEP), were reported to be its effective ingredients. Therefore, the extraction of potential antitumor ingredients from the plant was of interest. We first explored the optimized solvent extraction of antitumor agents from S. cepharantha Hayata guided by an in vitro antitumor activity assay. The solvent for extraction and its concentration, the liquid to material ratio, extraction duration, particle size, macerating time, and the frequency of extraction were investigated using a single-factor experiment. An orthogonal design (L9, 34) was constructed to determine the suitable extraction conditions. The crude extract was then purified sequentially by macroporous adsorption resins (MR) for the enrichment of CEP. Under these optimal conditions, the yield of total alkaloids in the herbs was 3.4%, whereas the CEP content was 2.9%. Total alkaloids exhibited significant anti-proliferative activities in the A549 cell line. Our study provides means for the further development and use of the antitumor components from S. cepharantha, which has potential for application in the pharmaceutical industry.
