Abstract The Sertoli cell (Sc) has been described as a quiescent cell once the animal has reached sexual maturity. Syrian hamster is an animal that displays testicular regression due to short photoperiod, during which process germ cells and Sc are removed through apoptosis. The aim of this work was to investigate histochemically whether the spontaneous testicular recrudescence processes after exposure to a short photoperiod lead to an increase in Sc proliferative activity in order to restore the normal population. Three spontaneous recrudescence groups were established: initial (IR), advanced (AR), and total (TR) recrudescence, which were compared with animal undergoing the regression process (mild: MRg, strong: SRg, and total: TRg) and animals in long photoperiod (Controls). Histological sections were submitted to histochemical techniques for detecting apoptotic and proliferative Sc with bright-field and fluorescence microscopy. For each group, the proliferative Sc index (PScI) and apoptotic Sc index (AScI), and the total number of Sc were obtained. The results revealed the existence of Vimentin+/TUNEL+ as well as Vimentin+/PCNA+ cells. The PScI was significantly higher in TRg and IR than in the other groups. The AScI was only significantly higher in MRg and SRg with respect to the other groups. The total number of Sc increased among TRg, IR, and AR, reaching values similar to those of the Controls. In conclusion, the increase in Sc proliferation from final regression and recrudescence, accompanied by a similar rate of apoptosis to the Control group, is the cause of the restoration of the Sc population during spontaneous recrudescence.

Myotis nigricans presents few and controversial reproductive data, which indicate geographical variation in reproduction. Thus, this study aimed to evaluate the seasonal modifications in testicular and epididymal morphologies in a tropical environment, submitting these organs to morphometric and immunohistochemical analysis. The observations revealed that this species presents two peaks of spermatogenic activity followed by two periods of total testicular regression (a quiescent pre-pubertal-like morphology, where only Sertoli cells and spermatogonia could be observed), in the same annual reproductive cycle, which seem to be only indirectly influenced by abiotic factors. This testicular behaviour seems to be synchronised with the caput and corpus epididymidis, but not with the cauda epididymidis, which presents aspects of sperm storage in May–June. The control of this variation seems to be directly linked to the expression of the androgen receptor, since, throughout the year, it is high in periods of testicular recrudescence and low in periods of deactivation. It is not thought to be directly linked to apoptosis, which is more pronounced in periods of recrudescence than in periods of regression.

Daily injections of L-dihydroxyphenylalanine (L-DOPA, dopamine precursor) given 4 h after 5-hydroxytryptophan (5-HTP, serotonin precursor) induced inhibitory responses in recrudescing gonad (in the first week of December) of Indian palm squirrel, a seasonally breeding subtropical animal. Other temporal relations (L-DOPA given at 0, 8, 12, 16, and 20 h after 5-HTP administration) did not show any effect on the recrudescing gonad. This inhibitory effect of 4 h was evident under short day length (6 : 18) group but was masked by the increasing day length of nature (NDL, late December onwards) and increased photoperiod of long day group (16 : 8). It is apparent that seasonal testicular recrudescence of Indian palm squirrel during short day length by 4 h relation of 5-HTP and L-DOPA is not a pharmacological effect but actually is an alteration of seasonality in this annually breeding mammal. It seems that endogenous mechanism controlling seasonal testicular recrudescence of Indian palm squirrel is reset by timed daily injections of these neurotransmitter drugs. It is suggested that in spite of different environmental factors (photoperiod, humidity, etc.) used by different species to time their annual reproduction, basic mechanism of seasonality appears to be the same, that is, the temporal synergism of neurotransmitter activity.

Our objective was to test the hypothesis that prolactin (PRL) acts at both the pituitary and testis levels to regulate testosterone secretion in the adult ram. The focus was on the mid-regression to mid-redevelopment stages of a photoperiod-condensed ‘seasonal’ testicular cycle. DLS rams (six per group) were given daily s.c. injections of bromocriptine (4 mg) or vehicle during the entire period. Serum PRL concentration in control rams peaked at 103.4±22.1 ng/ml in late regression and then steadily declined (P<0.01) to 19.5±4.3 ng/ml, whereas PRL in treated rams was always ≤4.0 ng/ml. Suppression of PRL tended (P<0.10) to increase the amplitude of natural LH pulses (transition stages) or reduce the number of LH receptors in the testis (regressed stage), although neither change disturbed testosterone levels in peripheral blood. These subtle changes were accompanied by significant (P<0.05) alterations in the capability of the pituitary to release LH (85% more) and of the testes to secrete testosterone (20% less). These effects of PRL were unmasked when rams were given highly stimulative i.v. injections of GNRH (single 3 μg dose) and NIH-oLH-S24 (three 5 μg doses given 20 min apart) respectively. PRL insufficiency also appeared to slow down the ‘seasonal’ rise in FSH secretion and slightly delayed (2 weeks) the times when the testes began to grow and were first significantly (P<0.05) enlarged from the regressed state. We conclude that PRL is an important part of the intricate regulation of the pituitary–gonadal system in moderately seasonal DLS rams.
