Research has been carried out to determine the cell structure of the epidermis and stomata in some mangrove plants in the species Aegiceras corniculatum and Rhizophora apiculata. Descriptive method is used to describe the cell structure of the epidermis and stomata of Aegiceras corniculatum and Rhizophora apiculata and quantitative leaves to calculate the number of stomata, number of epidermis and stomata index based on nail polish on the cross section of epidermal cells on the lower underside of the leaf using a light microscope, while the incision longitudinal to determine leaf thickness between the upper epidermis and the lower epidermis. The results showed that the two mangrove species that grow in the mouth of the Poka and Leahari villages namely Aegiceras corniculatum and Rhizophora apiculata were found to have the same anatomical structure and leaf anatomical characteristics in terms of the shape of epidermal cells, rectangular, octagonal, elongated, and irregular. Aegiceras corniculatum and Rhizophora apiculata have anomositic stomata type because neighboring cells surround the stomata and have the same shape as epidermal cells. Mangrove species in the river estuary of Poka Village have higher number of stomata and smaller epidermal size and lower stomata index than mangrove species in Leahari Village due to the influence of the shade. Aim: To investigate the taxonomic value of leaf anatomical characteristics and to determine the anatomical features that enabled the species to adapt to different environmental conditions. Methodology: Three methods were applied for leaf anatomy observation: sectioning using a sliding microtome, leaf clearing and epidermal peels. Results: The findings showed some common anatomical characteristics shared in all species studied, such as incomplete leaf venation, the presence of trichome and schlerenchyma cells, and the presence of mucilaginous idioblast cells in the petiole and midrib. Straight-to-sinuous anticlinal cell walls, amphistomatic leaves, and three types of stomata: parasitic, anomocytic and staurocytic were on the abaxial side of leaf lamina in this study. Peltate and capitate glandular trichome, as well as simple trichome, were all present in the petiole, midrib and lamina. However, peltate glandular trichome were only found in the root of I. triloba. Druses were observed in all species, except I. batatas. Interpretation: Leaf anatomical characteristics possess taxonomic value, especially in species differentiation and identification, that can be subsequently useful in explaining plant's adaptation to their environment.

ABSTRACT: The objective of the present study was to evaluate the effects of different culture media on the in vitro germination and initial development of Cattleya nobilior, and to acclimatize this species using soybean straw as a substrate component, aiming the production of plants at large scale to reintroduce it in natural environments. Anatomical characteristics associated with development during acclimatization are also depicted. The influence of Murashige and Skoog, Knudson, and Vacin and Went culture media on the in vitro germination and protocorm development were assessed. Acclimatization was accomplished using different proportions of soybean straw (SS) and Bioplant (BP) as substrates. Anatomical studies were conducted on the leaves of plants during acclimatization. KC was the most suitable culture medium for both germination and initial development of C. nobilior. Regarding acclimatization, it is recommended that a substrate composed of 60% SR and 40% BP be initially used. Upon transfer to shade-house conditions, plants should be grown in a mix of 40% SR: and 60% BP. The leaf anatomical characteristics observed are typical of epiphytic orchids adapted to water-poor environments, which indicates that SR did not negatively affect the development of the species. The in vitro propagation of C. nobilior as described herein is efficient for its multiplication for commercial and conservation purposes and SS can be used as an alternative component of the substrate for its acclimatization. Comparative leaf anatomy study were conducted in three taxa of Acanthaceae from peninsular Malaysia. Three chosen taxa were Acanthus ebracreatus (Vahl), Andrographis paniculata (Burm.f.) Wall. ex Nees and Chroesthes longifolia (Wight) B. Hansen which is commonly used as traditional medicine especially in peninsular Malaysia. The main objective is to identify the leaf anatomical characteristics that can be used in plant identification and also for supportive data in plant classification. The procedures involved such as cross section using sliding microtome on the petiole, lamina, midribs and marginal, leaf clearing and observation under light microscope. Results have shown the similarities and variations in leaf anatomical characteristics. The anatomical characteristics observed include petiole and midrib outlines, patterns of petiole and midrib vascular bundles, presence of cystolith cells, presence of hypodermis layers in lamina and presence and types of trichomes. In conclusion, results showed that anatomical characteristics have taxonomic significance that can be used in classification especially at species level.
