Background: Trichophyton spp., as pathogenic species to humans and animals, cause different forms of dermatophytosis through the production of particular enzymes, playing an essential role in tissue invasion. Among these, herein, keratinase was investigated, for the specific case of Trichophyton mentagrophytes, as a target of the effects of Allium hirtifolium and Allium jesdianum extracts, thus pharmacological potential of these plants was studied against keratinase activity. Methodology: Sampling was carried out on 20 bald patients from medical diagnostic laboratories and mycology centers, with suspected dermatophytosis of scalp. For confirming the presence of Trichophyton mentagrophytes in the specimens, different laboratory procedures were applied. Trichophyton mentagrophytes isolates were cultured on a screening medium containing keratin to verify production of the keratinase enzyme. The best enzyme-producing isolate was selected by measuring diameter of transparent halo around colony to be used in subsequent stages. Afterwards, the optimized conditions maximizing enzyme production and activity were determined. Finally, the inhibitory effect of different dilutions of aqueous and alcoholic extracts of Allium jesdianum and Allium hirtifolium on extracellular keratinase activity was studied. Results : Sixteen out of 20 fungal isolates were identified as the Trichophyton mentagrophytes. The most desirable reduction on keratinase activity was reported for dilution values of 50 and 100 mg/ml of both aqueous and ethanolic extracts of A.jesdianum, though much more significant decrease belonged to the latter, and for dilution values of 25 and 100 mg/ ml of both aqueous and ethanolic extracts of A.hirtifolium. Conclusion : Concerning our results, it is suggested that paying special attention to these natural compounds for the treatment of dermatophytosis could be remarkably effective, considering significant production of keratinase observed in T. mentagrophytes, and they are beneficial, as they have no side effects and offer an alternative to currently available medications, which are under the restriction of drug resistance.

Background: Allium hirtifolium and Satureja khuzestanica are the Iranian endemic plants and proper candidates for antioxidant studies. This study investigated the antioxidant, anti-hemolytic properties, and phytochemical composition in different extracts of A. hirtifolium and S. khuzestanica. Methods: Hydroalcoholic, methanolic, and n-hexane extracts of A. hirtifolium and S. khuzestanica plants were prepared using soaking and ultrasonic methods. Different plant extracts were evaluated for the presence of secondary metabolites using standard methods based on colorimetric analysis. The antioxidant properties of the compounds were measured by the ferric reducing ability of power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods. Finally, the anti-hemolytic effects of the extracts were investigated using the 2,2’-Azobis(2-amidinopropane) dihydrochloride (AAPH) methods. Results: Phytochemical analysis indicated the presence of flavonoids, phenolic acids, glycosides, steroids, and terpenoids in S. khuzestanica, as well as amino acid compounds, flavonoids, saponins, glycosides, and phenols in A. hirtifolium. FRAP assay showed that the methanolic extract of S. khuzestanica and the n-hexane extract of A. hirtifolium have the highest total antioxidant activity. The results of the DPPH assay also demonstrated that the minimum IC50 was related to the hydroalcoholic extract of S. khuzestanica (18.58 µg/mL) and the n-hexane extract of A. hirtifolium (87.95 µg/mL). In general, the extracts of both plants could reduce the percentage of AAPH-induced hemolysis while being significant only in some concentrations (P<0.001). Conclusion: Overall, A. hirtifolium and S. khuzestanica can be used as a herbal supplement in the human diet due to their anti-hemolytic effects.

AbstractAllium is one of the well-known genera of the Amaryllidaceae family, which contains over 780 species. Onions, garlic, leeks, and shallots are the most important species of this genus. Allium hirtifolium (shallot) is a rich source of proteins, carbohydrates, lipids, amino acids, and bioactive compounds such as organic sulfur compounds with an expansive range of biological activities and medicinal attributes. To identify the putative compounds and genes involved in the organic sulfur pathway, we applied GC–MS and RNA-seq techniques for the bulb, stem, and flower tissues of A. hirtifolium. The essential oil analysis revealed the maximum amount of sulfur compounds in stem against flower and bulb tissues. Transcriptome profiling showed 6155, 6494, and 4259 DEGs for bulb vs. flower, bulb vs. stem, and flower vs. stem, respectively. Overall, more genes were identified as being up-regulated rather than down-regulated in flower tissue compared to the stem and bulb tissues. Our findings in accordance with other results from different papers, suggest that carbohydrates are vital to bulb formation and development because a high number of identified DEGs (586 genes) were mapped to carbohydrate metabolism. This study has detected the genes in the organic sulfur pathway and indicated that the alliinase gene shows a high variability among different tissues. In general, this study formed a useful genomic resource data to explore tissue-specific sulfur pathway in A. hirtifolium, which is helpful for functional breeding.

Background and Objectives. Allium hirtifolium Boiss (Persian shallot), as an edible vegetable, has several pharmacological properties including antimicrobial, anti-inflammatory, and antioxidative effects, while its protective effects in liver cells are controversial. In this study, we examined the effect of A. hirtifolium extract on cadmium- (Cd-) induced hepatotoxicity in rats. Materials and Methods. Thirty-six male Wistar rats were divided into six groups: groups 1, 2, and 3 received vehicle, Cd (100 mg/L/day by drinking water), and A. hirtifolium extract (200 mg/kg/day; orally), respectively. Groups 4, 5, and 6 were Cd groups which were treated with A. hirtifolium extract (50, 100, and 200 mg/kg/day, respectively). After 2 weeks, liver enzymes such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) and also oxidative stress biomarkers including lipid peroxidation (LPO), total antioxidant capacity (TAC), total thiol molecule (TTM), and the histopathological changes were determined using standard procedure. Results. The findings showed that Cd caused a remarkable rise in levels of serum hepatic enzymes such as ALT (P<0.001), AST (P<0.01) and ALP (P<0.001) compared with the control group. In addition, Cd led to the decreasing of the levels of TTM (P<0.001) and TAC (P<0.001) and increasing of LPO (P<0.001) in liver tissue in comparison with the control group. In this regard, remarkable vascular congestion, hepatocellular degeneration, and vacuolization were observed in hepatic tissue of Cd-treated rats. Following the administration of A. hirtifolium extract, a significant improvement was observed in the functional and oxidative stress indices of hepatic tissue alongside histopathologic changes. Conclusion. The current study indicated that the A. hirtifolium extract might prevent hepatic oxidative injury by improving oxidant/antioxidant balance in rats exposed to Cd.

ABSTRACT Two different potato chip coatings—aqueous extracts including Zataria multiflora and Allium hirtifolium at concentrations of 1, 3, 5, and 7% and hydrocolloids individually or in combination—were used to decrease acrylamide content, and their effects on the characteristics of the product were then investigated. According to the results, the incorporation of hydrocolloids as the coating was more efficient in the reduction of acrylamide production than with the extracts. Also, the application of each extract and hydrocolloids individually can be considered a more efficient technique for acrylamide reduction than their mixture. In this regard, the economic aspects of the application of hydrocolloids in the coating of fried potato crisps can be evaluated. HIGHLIGHTS
