Despite a growing number of studies, the role of pollinators as a selection agent for nectar traits remains unclear. Moreover, the lack of data from some biogeographic regions prohibits us from determining their general importance and global patterns. We analyzed nectar carbohydrate traits and determined the main pollinators of 66 plant species in the tropical forests of Mount Cameroon (tropical West Africa). The measured nectar traits included total sugar amounts and proportions of sucrose and hexoses (i.e., glucose and fructose). We report the nectar properties for plants visited by five pollinator groups (bees, butterflies, moths, hoverflies, and specialized birds). Our results indicate that, rather than specific evolution in each of the five plant groups, there was a unique nectar-trait evolution in plants pollinated by specialized birds. The ornithophilous plants had a higher proportion of sucrose and produced larger sugar amounts than the plants pollinated by insects. We also demonstrated a significant phylogenetic signal in the nectar properties in some lineages of the studied plants.

AbstractAmmonia has a cytotoxic effect and can therefore be used as a selection agent for enrichment of zone I hepatocytes. However, it has not yet been determined whether ammonia-treated hepatocyte-like cells are able to proliferate in vitro. In this study, we employed an ammonia selection strategy to purify hepatocyte-like cells that were differentiated from human embryonic stem cells (ESCs) and from induced pluripotent stem cells (iPSCs). The resistance to cytotoxicity or cell death by ammonia is likely attributable to the metabolism of ammonia in the cells. In addition to ammonia metabolism-related genes, ammonia-selected hepatocytes showed increased expression of the cytochrome P450 genes. Additionally, the ammonia-selected cells achieved immortality or at least an equivalent life span to human pluripotent stem cells without affecting expression of the liver-associated genes. Ammonia treatment in combination with in vitro propagation is useful for obtaining large quantities of hepatocytes.

In monocots, however, it does not seem to be effective as it has no or minimal effect on the normal growth of non-transformed plants. Kanamycin was previously demonstrated to bleach the pigments of the non-transgenic plants. Abstract Biocontainment systems are needed to neutralize genetically modified organisms (GMOs) that pose ecological threats outside of controlled environments. In contrast, benign selection markers complement GMOs with reduced fitness. Benign selection agents serve as alternatives to antibiotics, which are costly and risk spread of antibiotic resistance. Here, we present a yeast biocontainment strategy leveraging engineered fluoride sensitivity and DNA vectors enabling use of fluoride as a selection agent. The biocontainment system addresses the scarcity of platforms available for yeast despite their prevalent use in industry and academia. In the absence of fluoride, the biocontainment strain exhibits phenotypes nearly identical to those of the wildtype strain. Low fluoride concentrations severely inhibit biocontainment strain growth, which is restored upon introduction of fluoride-based vectors. The biocontainment strategy is stringent, easily implemented, and applicable to several eukaryotes. Further, the DNA vectors enable genetic engineering at reduced costs and eliminate risks of propagating antibiotic resistance.

Streptococcus pneumoniae is the causative agent of a multitude of diseases, and further study into its pathogenies is vital. The pneumococcus is genetically malleable, and several tools are available to manipulate this pathogen. In this study, we attempted to utilize one such tool, the Sweet Janus cassette, to replace the capsule locus with other capsule loci in our strain background and found that the efficiency of allelic replacement was low and the number of revertant false-positive colonies was high. We determined that the capacity to recombine capsule varied by the initial isolated colony, suggesting that frequency of reversion is dependent on the bacterial clone. Alternative selection markers may further expand the application of Sweet Janus. We created novel cassettes that utilized chlorinated phenylalanine as an alternative counter-selection agent in conjunction with the Janus or Sweet Janus cassette, providing a new dual or triple selection marker. Moreover, we created cassettes that do not require engineered resistance in the background strain, including both single and dual selection markers. We were able to utilize all constructs in allelic replacement of the capsule loci. These novel constructs provide a new means for generating gene deletions in S. pneumoniae that expand experimental applications.

Climate change has enhanced the occurrence of rural fires, since changes in the hydrological cycle have led to the occurrence of increasingly long and frequent periods of drought. This recurrence of rural fires in Portugal, in turn, has led to the successive elimination of vast areas traditionally occupied by native species or species of economic interest, which are being successively replaced by new species with invasive behavior. Among these, Acacia dealbata stands out for its dispersion capacity and for the area it has already occupied. In the present work, which reviews the evolution of forest cover over the last 18,000 years in the Serra da Estrela Natural Park, we intend to demonstrate that fire acts as a species selection agent and that it enhances the development of heliophile and pyrophyte species. For this purpose, an area of the municipality of Seia was selected, more specifically Casal do Rei, where the development of Acacia dealbata forests is monitored. In the end, it was concluded that, in fact, by analyzing the ages of the specimens present in these populations, fire acts as a selection agent by freeing up the space previously occupied by other species, opening the way for the growth of heliophiles and pyrophytes invasive species while enhancing their germination.
