Marine animals such as the southern elephant seal (Mirounga leonina) rely on a productive marine environment and are vulnerable to oceanic changes that can affect their reproduction and survival rates. Davis Base, Antarctica, acts as a moulting site for southern elephant seals that forage in Prydz Bay, but the genetic diversity and natal source populations of these seals has not been characterized. Determining the genetic diversity of moulting populations like this one provides essential information on seal dispersal, inter-population mixing, and foraging behaviours. In this study, we combined genetic and animal tracking data on these moulting seals to identify levels of genetic diversity, natal source population, and movement behaviours during foraging and haul-out periods. Using mitochondrial sequence data, we identified two major breeding lineages of seals at Davis Base. We found that the majority of the seals originated from breeding stocks within the South Atlantic Ocean and South Indian Ocean. One seal was grouped with the Macquarie Island breeding stock (South Pacific Ocean). The Macquarie Island population, unlike the other two stocks, is decreasing in size. Tracking data revealed long-distance foraging activity of the Macquarie Island seal around Crozet Islands. We speculate that changes to the Antarctic marine environment have resulted in a shift in foraging and dispersal strategies, which subsequently affects seal population growth rates. These findings have implications for conservation management plans aimed at improving the population status of the southern elephant seal.

Many remote islands present barriers to effective wildlife monitoring in terms of challenging terrain and frequency of visits. The sub-Antarctic islands of South Georgia and the South Sandwich Islands are home to globally significant populations of seabirds and marine mammals. South Georgia hosts the largest breeding populations of Antarctic fur seals, southern elephant seals and king penguins as well as significant populations of wandering, black-browed and grey-headed albatross. The island also holds important populations of macaroni and gentoo penguins. The South Sandwich Islands host the world’s largest colony of chinstrap penguins in addition to major populations of Adélie and macaroni penguins. A marine protected area was created around these islands in 2012 but monitoring populations of marine predators remains a challenge, particularly as these species breed over large areas in remote and often inaccessible locations. During the 2019/20 austral summer, we trialled the use of an unoccupied aerial vehicle (UAV; drone) to monitor populations of seals, penguins and albatross and here we report our initial findings, including considerations about the advantages and limitations of the methodology. Three extensive southern elephant seal breeding sites were surveyed with complete counts made around the peak pupping date, two of these sites were last surveyed 24 years ago. A total of nine islands, historically recorded as breeding sites for wandering albatross, were surveyed with 144 fledglings and 48 adults identified from the aerial imagery. The UAV was effective at surveying populations of penguins that nest on flat, open terrain, such as Adélie and chinstrap penguin colonies at the South Sandwich Islands, and an extensive king penguin colony on South Georgia, but proved ineffective for monitoring macaroni penguins nesting in tussock habitat on South Georgia as individuals were obscured or hidden by vegetation. Overall, we show that UAV surveys can allow regular and accurate monitoring of these important wildlife populations.

Alphaviruses infect a diverse range of host organisms including mosquitoes, mammals, and birds. The enigmatic alphavirus non-structural protein 3 (nsP3) has an intrinsically disordered, C-terminal hypervariable domain (HVD) that can interact with a variety of host proteins associated with stress granules (SGs). The HVD displays the highest variability across the more than 30 known alphaviruses, yet it also contains several motifs that are conserved amongst different subgroups of alphaviruses. For some alphaviruses, specific nsP3–SG protein interactions are essential for virus replication. However, it remains difficult to attribute general roles to these virus-host interactions, as multiple amino acid motifs in the HDV display a degree of redundancy and previous studies were performed with a limited number of alphaviruses. To better understand nsP3-host protein interactions we conducted comprehensive co-localization experiments with the nsP3s of 20 diverse alphaviruses: chikungunya, Semliki Forest, Sindbis, Bebaru, Barmah Forest, Getah, Mayaro, Middelburg, O'nyong-nyong, Ross River QML and T48, Una, Whataroa, Southern Elephant Seal, Eilat, Tai Forest (TAFV), Venezuelan/Eastern/Western equine encephalitis (V/E/WEEV) and the aquatic Salmonid alphavirus (SAV), with three different SG proteins (G3BP and its insect homolog Rasputin, FMRP) and BIN1 in mammalian and mosquito cell lines. Despite that all terrestrial alphavirus nsP3s contained at least one BIN1-binding motif (PxPxPR), not all nsP3s co-localized with BIN1. Further, all alphaviruses except SAV, TAFV and VEEV displayed co-localization with G3BP. Although viruses lacking FGxF-like motifs contained Agenet-like domain binding motifs to facilitate interaction with FMRP, cytoplasmic nsP3 granules of all tested alphaviruses co-localized with FMRP. Crispr-Cas9 knockout of G3BP in mammalian cells abolished nsP3-FMRP co-localization for all alphaviruses except V/E/WEEV nsP3s that bind FMRP directly. G3BP knockout also changed nsP3 subcellular localization of Bebaru, Barmah Forest, Getah, and Sindbis viruses. Taken together this study paints a more detailed picture of the diverse interactions between alphavirus nsP3 and SG-associated host proteins. The interaction between nsP3 and G3BP clearly plays a central role and results in recruitment of additional host proteins such as FMRP. However, direct binding of FMRP can make the interaction with G3BP redundant which exemplifies the alternate evolutionary paths of alphavirus subgroups.

AbstractAn unusual sighting of a juvenile southern elephant seal (Mirounga leonina), outside its typical range, was recorded off the southern coast of Sri Lanka between 20th November and 20th December 2019. The individual, a male as confirmed by the presence of a prepuce, underwent a catastrophic moult during this 1 month period, following which no further observations were recorded and so it was assumed it returned to sea. This represents the first confirmed sighting of a southern elephant seal, and indeed a phocid, in Sri Lankan waters.

AbstractWe present estimates of the seasonal and spatial occupation by pinnipeds of the Wildlife Refuge of Ilha dos Lobos (WRIL), based on aerial photographic censuses. Twenty aerial photographic censuses were analysed between July 2010 and November 2018. To assess monthly differences in the numbers of pinnipeds in the WRIL we used a Generalized Linear Mixed Model. Spatial analysis was carried out using Kernel density analysis of the pinnipeds on a grid plotted along the WRIL. Subadult male South American sea lions (Otaria flavescens) were the most abundant pinniped in the WRIL. Potential females of this species were also recorded during half of the census. The maximum number of pinnipeds observed in the WRIL was 304 in September 2018, including an unexpected individual southern elephant seal (Mirounga leonina), and a high number of South American fur seal yearlings (Arctocephalus australis). However, there was no statistically significant difference in counts between months. In all months analysed, pinnipeds were most often found concentrated in the northern portion of the island, with the highest abundances reported in September. This study confirms the importance of the WRIL as a haulout site for pinnipeds in Brazil, recommends that land research and recreational activities occur in months when no pinnipeds are present, and encourages a regulated marine mammal-based tourism during winter and spring months.
