Abstract We hypothesized that bone may secrete hormones that regulate energy metabolism and reproduction. Testing this hypothesis revealed that the osteoblast-specific secreted protein osteocalcin is a hormone regulating glucose homeostasis and male fertility by signaling through a GPCR, Gprc6a, expressed in pancreatic β bells and Leydig cells of the testes. The systematic exploration of osteocalcin biology, revealed that it regulates an unexpectedly large spectrum of physiological functions in the brain and peripheral organs and that it has most features of an antigeromic molecule. As will be presented at the meeting, this body of work suggests that harnessing osteocalcin for therapeutic purposes may be beneficial in the treatment of age-related diseases such as depression, age-related memory loss and the decline in muscle function seen in sarcopenia.

Objective. Neurological dysfunction commonly occurs in the upper limb contralateral to the hemisphere of the brain in which stroke occurs; however, the impact of stroke on function of the ipsilesional upper limb is not well understood. This study aims to systematically review the literature relating to the function of the ipsilesional upper limb following stroke and answer the following research question: Is the ipsilesional upper limb affected by stroke?Data Source. A systematic review was carried out in Medline, Embase, and PubMed.Review Methods. All studies investigating the ipsilesional upper limb following stroke were included and analysed for important characteristics. Outcomes were extracted and summarised.Results. This review captured 27 articles that met the inclusion criteria. All studies provided evidence that the ipsilesional upper limb can be affected following stroke.Conclusion. These findings demonstrate that clinicians should consider ipsilesional upper limb deficits in rehabilitation and address this reduced functional capacity. Furthermore, the ipsilesional upper limb should not be used as a “control” measure of recovery for the contralateral upper limb.

Objective: Recent improvements in life expectancy globally require intensified focus on noncommunicable diseases and age-related conditions. The purpose of this article is to inform the development of age-specific prevention guidelines for adults aged 50 and above, which are currently lacking. Data Source: PubMed, Cochrane database, and Google Scholar and explicit outreach to experts in the field. Study Inclusion and Exclusion Criteria: Meta-analyses, intervention-based, and prospective cohort studies that reported all-cause mortality, disease-specific mortality, or morbidity in adults were included. Data Extraction: A systematic review was undertaken in 2015 using search terms of a combination of <risk factor> and “intervention,” “mortality,” “reduction,” “improvement,” “death,” and “morbidity.” Data Synthesis: Interventions were categorized according to the Center for Evidence-Based Medicine Level of Evidence framework. Results: A summary table reports for each intervention the impact, strength of evidence, initiation, duration, and details of the intervention. Age-decade-specific preventive recommendations have been proposed relating to physical activity, diet, tobacco and alcohol use, medication adherence, screening and vaccination, and mental and cognitive health. Conclusion: Clear recommendations have been made according to the existing evidence base, but further research investment is needed to fill the many gaps. Further, personalized approaches to healthy aging complemented by population-wide approaches and broader cross-sector partnerships will help to ensure greater longevity is an opportunity, rather than a burden, for society.

AbstractThe hospitalization and symptomatic rates of Severe Acute Respiratory Syndrome Coronavirus Disease 2019 (SARS-CoV-2) are key epidemiological parameters affecting risk analyses conducted for the Canadian Armed Forces (CAF) during the Coronavirus Disease-19 (COVID-19) pandemic. As one of the criteria of a variant of concern (VOC) is that it affects disease severity, the authors sought to understand whether the Alpha and Gamma VOCs are significantly different in these two parameters than the original wildtype of SARS-CoV-2, the most prevalent in relevant areas of Canada as of study initiation. Searches for studies were conducted in Scopus and PubMed, and located through following citations and receiving studies from daily literature scans. For the hospitalization outcome, effect ratios relative to original wildtype were included. For the symptomatic ratio, the ratio itself for each variant was used. Analysis of age-related effects was of particular value, as CAF members are primarily adults under the age of 60. The firmest conclusion of this review is that the Alpha VOC comes with a higher relative risk of hospitalization compared to the original wildtype, most likely above 1.4, while unlikely to be above 2, with the balance of evidence being that the relative risk is not significantly modified by age. The evidence for Gamma is more limited, but the odds ratio may be above 2, and potentially much greater than that, especially for those 20-39 years of age. For both VOCs reports on symptomatic ratio differed on whether there was an effect, as well as its potential direction.

Over the past two decades, many neuroimaging studies have attempted uncover the brain regions and networks involved in path integration and identify the underlying neurocognitive mechanisms. Although these studies made inroads into the neural basis of path integration, they have yet to offer a full disclosure of the functional specialization of the brain regions supporting path integration. In this paper, I reviewed notable neuroscientific studies on visual path integration in humans, identified the commonalities and discrepancies in their findings, and incorporated fresh insights from recent path integration studies. Specifically, this paper presented neuroscientific studies performed with virtual renditions of the triangle/path completion task and addressed whether or not the hippocampus is necessary for human path integration. Based on studies that showed evidence supporting and negating the involvement of the hippocampal formation in path integration, this paper introduces the proposal that the use of different path integration strategies may determine the extent to which the hippocampus and entorhinal cortex are engaged during path integration. To this end, recent studies that investigated the impact of different path integration strategies on behavioral performance and functional brain activity were discussed. Methodological concerns were raised with feasible recommendations for improving the experimental design of future strategy-related path integration studies, which can cover cognitive neuroscience research on age-related differences in the role of the hippocampal formation in path integration and Bayesian modelling of the interaction between landmark and self-motion cues. The practical value of investigating different path integration strategies was also discussed briefly from a biomedical perspective.

Second language acquisition (SLA) can contribute to the changes in the brain. The paper having a holistic perspective towards the relation between brain and language asserts that the impact of SLA on the brain change is poorly studied. Moreover, claiming that the brain change is dynamic implicates the assumption that the plasticity of the brain is not merely determined by age-related factors. In this regard, experience, in general, and SLA, in particular, has a tremendous effect on the brain change. Thus, resting on the claim that SLA is respected as software and contributes to the function of brain, the paper directs the attention towards agrammatism which attracts much attention from the researchers in neurolinguists. The article also tends to cast lights upon our perceptions towards the notion of change in the brain from the neurolinguistic perspective.
