Hematological parameters of pure and reciprocal crosses of Pangasianodon hypophthalmus (Sauvage, 1878) and Clarias gariepinus (Burchell, 1822)

SummaryBreeding and larval performance of novel hybrids from reciprocal crosses of Asian catfishPangasianodon hypophthalmus(Sauvage, 1878) and African catfishClarias gariepinus(Burchell, 1822) were investigated in this study. Spawning was by hormonal injection of brood fish, artificial fertilization, and incubation in triplicate aquarium tanks (0.5 × 0.5 × 0.5 m3) with continuous aeration. Reciprocal crosses (♀C. gariepinus× ♂P. hypophthalmusand ♀P. hypophthalmus× ♂C. gariepinus) had lower hatchability (≤50%) than their pure siblings (≥75%). Fish from all crosses survived until the juvenile stage but survival at 35 days post hatching (dph) was higher for pureC. gariepinussib. ♀C. gariepinus× ♂P. hypophthalmuswas observed to be less resistant to degradation of water quality than the other crosses, however it had higher body weight compared with the other crosses that showed similar performance. Morphological comparison of surviving juvenile at 35 dph, showed that all ♀P. hypophthalmus× ♂C. gariepinusand 13% of the ♀C. gariepinus× ♂P. hypophthalmusexhibited the very same morphology as that of their maternal parent species, while the other portion of the ♀C. gariepinus× ♂P. hypophthalmuscross exhibited morphological traits that were intermediate between those of both parent species. This study been the first successful attempt to hybridize both species and therefore, laid the groundwork for further studies on the aquaculture potentials of the novel hybrids.

Hematological parameters have been recognized as valuable tools for monitoring fish health. The aim of the present study was to obtain a basic knowledge of the hematological responses of different groups of fresh water teleosts during acclimation period. Haematological analysis was carried out in three different groups of teleosts namely Group I: Heteropneustes fossilis and Clarias gariepinus (Siluriformes), Group II: Anabas testudineus and Channa striatus (Perciformes), Group III: Labeo rohita and Cyprinus carpio (Cypriniformes). In all groups of teleosts, RBC and Hb were significantly (p<0.05) decreased after 7 days of captivity in response to stress factors raised in capture, handling and sampling procedure. WBC was found to be significantly (p<0.05) increased in group I (Siluriformes), group II (Perciformes) and group III (Cypriniformes) of teleosts after 7 days of captivity due to enhanced production of leucocytes in the hematopoietic tissue of the kidney and the spleen. RBC count and WBC count were found to be higher in H. fossilis and C. gariepinus than other groups of teleosts due to their active predacious nature. Hb values were high in 2nd group (Perciformes) consisting of A. testudineus and C. striatus by virtue of their possessions of accessory respiratory organs. PCV, MCV and MCHC were significantly (p<0.05) increased in three groups of teleosts during acclimation and values were high in second group of consisting of A. testudineus and C. striatus. MCH was higher in 3rd group of fishes consisting of L. rohita and C. carpio.

Probiotics have been widely known to have the ability to improve the immune system of livestock and aquatic animal. The present study was carried out to evaluate the effect of dietary supplementation of two probiotic isolates of Enterococcus faecium on hematological parameters of juvenile African catfish, Clarias gariepinus during pre- and post-challenge with aquatic pathogen, Aeromonas hydrophila. The probiotics were previously isolated from vegetable wastes (mung bean sprouts, Vigna radiate and cucumber, Cucumis sativus) which have been fermented for 7 days. The experimental fish (270 tails) with an average weight of 5.13 ± 1.03 g were distributed and divided randomly  into  i) control (30 tails), fed with commercial diet ii) E1 (30 tails), fed with  diets  supplemented  with  108 CFU/ml  of  E. faecium isolated from fermented cucumber, iii) E2 (30 tails), fed with 108 CFU/ml  of  E. faecium isolated from fermented mung bean sprouts. The feeding trial was conducted for 50 days. All experimental groups were then challenged with A. hydrophila (1.5 × 106 CFU/mL) via intraperitoneal injection on day 51. Prior to challenge, blood samples were collected from five fish randomly selected from each group on the day 51 (pre-challenge).  After 72 hours of post-challenge, blood samples were again collected from five fish from each groups. The hematological parameters such as total erythrocyte count  (RBC),  total  leucocyte  count  (WBC),  packed cell volume (PCV),  hemoglobin  (Hb), the derived blood indices of mean corpuscular volume (MCV) and corpuscular hemoglobin concentration (MCHC) were examined.  Hematological profiles of pre- and post-challenge infected juvenile catfish were compared with the control groups. The RBC, Hb, WBC, PCV, MCV and MCHC of fish fed with probiotics showed higher significant difference (P<0.05) as compared to control groups during pre- and post-challenge of pathogen. The high level of RBC and WBC during pre- and post-challenge showed the capability of the probiotics to improve the immune response of juvenile African catfish and thus increased the fish disease resistance against A. hydrophila infection. The result suggested that E. faecium could be used effectively as a probiotic in aquaculture.
