Aqueous extract of Moringa oleifera Lam. induced mitodepression and chromosomal aberration in Allium cepa, and reproductive genotoxicity in male mice

The objective of the study is to evaluate the antihelminthic property of Moringa oleifera aqueous leaf extract by studying the paralysis and mortality time of Tubiflex tubiflex (blood worm). The aqueous extract at 200mg/ml exhibited paralysis time of 1 minute and death time of 2 minutes which was on par with synthetic drug Albendazole at 20mg/ml. Crude condensed tannin paralysed T.tubiflex at 60 seconds with mortality time of 9 minutes at 1mg/ml. T.tubiflex was selected for the study since they share a similar anatomical and physiological aspects with the intestinal round worm. The study indicated that T.tubiflex was inhibited effectively by condensed tannin in Moringa oleifera aqueous leaf extract and perhaps might be more effective on the round worms as well, since it is consumed as vegetable for its nutritive and medicinal values in tropical and subtropical regions.

Moringa oleifera Lam. is a highly valued medicinal plant in India, especially Kerala. In the present study, antioxidant activity of aqueous extract of leaves of M. oleifera was determined both in-vitro and in-vivo. Male Wistar rats of 3 age groups- 6, 12, and 18 months old were used for in-vivo analysis. In vitro anti-proliferative effect of the extract was carried out in Dalton’s Lymphoma Ascites (DLA) Cells. LCMS-QTOF analysis of the extract was also done to determine the bioactive components present in the extract. Antioxidant activity of M. oleifera leaf showed an IC 50 value of 10.47 ?g/ml and whereas for standard drug, ascorbic acid, it was 19.52 ?g/ml. In-vivo analysis of lipid peroxidation showed a significant reduction of lipid peroxidation in the brains of 12 and 18-months old treated groups. Up to 75% mortality of DLA cancerous cells was observed in-vitro in different concentrations of M. oleifera leaf water extract in a dose-dependent manner, demonstrating its anti-proliferative property. LCMS-QTOF analysis revealed the presence of emodin-8-glucoside in the extract. Molecular docking analysis (Auto Dock Vina) of emodin-8-glucoside with six cancer related proteins showed highest binding affinity with AKT-1 with a binding score of -10.4 kcal/mol, also showed good affinity with NF-kB (p65), Stat-3, Bcl-2, Bcl-xl and c-FLIP. This study helps to choose healthy diet practices to overcome free radical onslaught and cancerous cell proliferation especially in the later stages of life. This can also pave way for the emergence of diet based therapeutic cure for cancer.

Actualmente el uso corriente de plantas medicinales para el tratamiento de las diferentes afecciones se ha incrementado y en la mayoría de los casos se comercializan sin previos estudios de toxicidad. M. oleifera es un árbol distribuido en zonas tropicales y subtropicales, muy utilizado en la medicina tradicional para tratar múltiples afecciones. En el marco del Proyecto de Investigación incentivado 16Q626 “Evaluación de la Genotoxicidad de Extractivos de Especies Medicinales de uso corriente para las afecciones más frecuentes en la Ciudad de Posadas Provincia de Misiones Argentina”, en esta instancia se propuso analizar la genotoxicidad de extractos acuosos de M. oleifera por medio del test de Allium cepa. El experimento consistió en dos etapas, donde además de determinar la concentración que detiene el crecimiento de las raíces un 50% en comparación con los controles y las anomalías macroscópicas, se buscaron los biomarcadores predeterminados de genotoxicidad (cromosomas rezagados, c-mitosis, anafase puente, etc) a concentraciones iguales o superiores a la concentración 50. Las concentraciones más elevadas probadas mostraron mayor inhibición de la mitosis con respecto a los controles, pero no se encontraron efectos genotóxicos, aunque las concentraciones utilizadas en el experimento, no son usuales de M. oleífera en la medicina tradicional.

Objective: To determine the anti-diabetic activity of combined aqueous extracts (1:1mixture) of dry leaves of Psidium guajava linn and Moringa oleifera lam as well as to compare the anti-diabetic activity of these plants by in vitro methods. Methods: In vitro alpha amylase inhibitory assay was performed on porcine alpha amylase and the absorbance was measured at 540 nm using a microplate reader and glucose diffusion inhibitory assay using dialysis membrane. Acarbose was used as the standard in the above mentioned methods. Results: The mixture (1:1) of aqueous plant extracts (at a concentration of 100µg/ml) of Psidium guajava linn and Moringa oleifera lam exhibited 72.08333% inhibition with IC50 value of 10.9µg/ml. The leaf extracts of Psidium guajava (at a concentration 100µg/ml) exhibited 71.23288% of a α amylase inhibitory activity with an IC50 values 19.883µg/ml whereas the leaf extracts of Moringa oleifera (at a concentration of 100µg/ml) exhibited 70.58824% of α amylase inhibitory activity with an IC50 value of 27.974 µg/ml. The Acarbose (standard drug) at a concentration of 100µg/ml showed 72.09302% inhibitory effect on the α amylase activity with an IC50 value 8.9µg/ml. In glucose diffusion inhibition assay the mixture of plant extracts exhibited 76.57% inhibition at 150 min which produces more effects than the two plants. The aqueous extract of Psidium guajava leaves exhibited maximum glucose diffusion inhibition (75.32%) at 150 min as well as Moringa oleifera leaf extract showed the maximum inhibition of 73.70% at the same time interval. For acarbose the percentage was 82.74 at 150 min. The interpretation of the results was done by one-way anova method. Conclusion: The combined extract of the leaves of the 2 plants was found to be more effective than individual plant extracts against diabetes. On comparison of two plants Psidium guajava was found to be more active against diabetes than Moringa oleifera. Also the potentiation effect shown by the combination of extract may be due to synergistic effect of the phytochemical constituents. As the 1:1 mixture of the aqueous extract is found to be more active, the combination of the two plants can be used to formulate drugs for treating diabetes.

Abstract Objective Among the psychiatric co-morbidities in epilepsy, anxiety disorders are highly frequent and have profound influence on the quality of life of epilepsy patients. Moringa oleifera Lam. (Moringaceae) is used in traditional medicine to treat various ailments including anxiety and epilepsy. However, no scientific evidence exists to support its use. We studied antiepileptic and anxiolytic activities of aqueous extract of Moringa oleifera Lam. leaves (AEMO). Methods Antiepileptic activity was evaluated using pentylenetetrazole (PTZ) induced seizure and maximum electroshock (MES) induced seizure test and anxiolytic activity was evaluated using elevated plus maze, light/dark box and hole board test. Results In present study, AEMO (250, 375 and 500 mg/kg, i.p.) demonstrated significant antiepileptic and anxiolytic effects. To study involvement of GABA in anxiolytic and antiepileptic activity of AEMO, we also evaluated effect of AEMO on Baclofen induced catatonia, a GABA mediated behavior, wherein AEMO significantly potentiated (preponed) baclofen induced catatonia, which is suggestive of its GABA mimetic action. Conclusion Thus, it may be concluded that aqueous extract of M. oleifera possess anxiolytic and antiepileptic effects possibly mediated via of GABA mimetic action and these findings authenticate the traditional claims about use of Moringa oleifera in treatment of epilepsy and anxiety.

Aims: This research is essentially on the Molluscicidal assessment of Aqueous extract of Moringa oleifera Lam seed on Bulinus Snail for the control of Schistosomiasis. Study Design: This is a controlled study where a total of 810 Bulinus snails were collected from three different streams with each stream representing a community from each of the three senatorial zones of Anambra state where schistosomiasis was endemic. Aqueous extract of Moringa oleifera (Lam) seed at different concentrations were used on the Bulinus snails to determine their molluscidal properties. Place and Duration of Study: Two hundred and Seventy (270) Bulinus snails were collected from streams representing each of the three senatorial zones of Anambra state viz; Obutu Lake, Omogho town, Orumba North Local Government Area (Anambra South), Agulu Lake, Agulu town, Aniocha Local Government Area (Anambra Central) and Omambra River, Omor town, Ayamelum Local Government Area (Anambra North) This research was carried out between November 2018 to April 2019. Methodology: Aqueous dilutions of the grinded Moringa oleifera (Lam) seed were exposed to the Bulinus snails from the various streams for 24 hours, after which the snails were removed from the experimental test solution and washed thoroughly with dechlorinated tap water and transferred to containers with fresh dechlorinated tap water for another 24 hours of recovery. The snails were incubated at 28 ± 5°C and fed with lettuce leaves. Results: Aqueous Moringa oleifera Lam seed extract was toxic to Bulinus adult snails in a dose dependent manner and the total Lethal Concentration (LC50 and LC90) values determined after 24 hours exposure from the whole streams were 468 ppm and 813 ppm respectively. Conclusion: Aqueous seed extract of Moringa oleifera Lam was toxic to Bulinus snail even at low concentrations.
