A set of V–(4–8)Cr–(0–4)Ti alloys was fabricated to survey an optimum composition to reduce the radioactivity of V–Cr–Ti alloys. These alloys were subjected to nano-indenter tests before and after 2-MeV He-ion irradiation at 500 °C and 700 °C with 0.5 dpa at peak damage to investigate the effect of Cr and Ti addition and gas impurities for irradiation hardening behavior in V–Cr–Ti alloys. Cr and Ti addition to V–Cr–Ti alloys for solid–solution hardening remains small in the unirradiated V–(4–8)Cr–(0–4)Ti alloys. Irradiation hardening occurred for all V–Cr–Ti alloys. The V–4Cr–1Ti alloy shows the highest irradiation hardening among all V–Cr–Ti alloys and the gas impurity was enhanced to increase the irradiation hardening. These results may arise from the formation of Ti(CON) precipitate that was produced by He-ion irradiation. Irradiation hardening of V–Cr–1Ti did not depend significantly on Cr addition. Consequently, for irradiation hardening and void-swelling suppression, the optimum composition of V–Cr–Ti alloys for structural materials of fusion reactor engineering is proposed to be a highly purified V–(6–8)Cr–2Ti alloy.

AbstractNanocrystalline oxide-dispersion strengthened ferritic alloy formation and its annealing behavior were examined through modern X-ray diffraction pattern analysis and supplemented by microhardness and microscopic measurements. The basic microstructure features, with particular emphasis on evolution of domain size distribution and defect content during mechanical and thermal treatment, were quantified via the whole powder pattern modeling approach. The microstructure of the powdered alloy, formed during mechanical alloying, evolved toward nanocrystalline state consisting of narrow dispersion of very fine crystallites with substantial dislocation density, which exhibited relatively high stability against elevated temperature. It was shown that crystallite size is seriously sustained by the grain-boundary strain, therefore coarsening of grains begins only after the density of dislocations drops below certain level. Obtaining correct results for the annealing-related data at specific temperature range required the incorporation of the “double-phase” model, indicating possible bimodal domain size distribution. The dislocation density and grain size were found not to be remarkably affected after consolidation by hot isostatic pressing.

AbstractThe RNA decay pathway plays key regulatory roles in cell identities and differentiation processes. Although adipogenesis is transcriptionally and epigenetically regulated and has been thoroughly investigated, how RNA metabolism that contributes to the stability of phenotype-shaping transcriptomes participates in differentiation remains elusive. In this study, we investigated Ddx6, an essential component of processing bodies (PBs) that executes RNA decay and translational repression in the cytoplasm and participates in the cellular transition of reprogramming. Upon adipogenic induction, Ddx6 dynamically accumulated to form PBs with a binding partner, 4E-T, at the early phase prior to emergence of intracellular lipid droplets. In contrast, preadipocytes with Ddx6 knockout (KO) or 4E-T knockdown (KD) failed to generate PBs, resulting in significant suppression of adipogenesis. Transcription factors related to preadipocytes and negative regulators of adipogenesis that were not expressed under adipogenic stimulation were maintained in Ddx6-KO and 4E-T-KD preadipocytes under adipogenic induction. Elimination of Dlk1, a major negative regulator of adipogenesis, in 3T3L1 Ddx6-KO cells did not restore adipogenic differentiation capacity to any extent. Similar to murine cells, human primary mesenchymal stem cells, which can differentiate into adipocytes upon stimulation with adipogenic cocktails, required DDX6 to maturate into adipocytes. Therefore, RNA decay of the entire parental transcriptome, rather than removal of a strong negative regulator, could be indispensable for adipogenesis.
