Differential transcriptome analysis of the common shrimp Crangon crangon: Special focus on the nuclear receptors and RNAi-related genes

Abstract Background The common littoral shrimp Palaemon serratus is an economically important decapod resource in some European communities. Aquaculture practices prevent the genetic deterioration of wild stocks caused by overfishing and at the same time enhance the production. The biotechnological manipulation of sex-related genes has the proved potential to improve the aquaculture production but the scarcity of genomic data about P. serratus hinders these applications. RNA-Seq analysis has been performed on ovary and testis samples to generate a reference gonadal transcriptome. Differential expression analyses were conducted between three ovary and three testis samples sequenced by Illumina HiSeq 4000 PE100 to reveal sex-related genes with sex-biased or sex-specific expression patterns. Results A total of 224.5 and 281.1 million paired-end reads were produced from ovary and testis samples, respectively. De novo assembly of ovary and testis trimmed reads yielded a transcriptome with 39,186 transcripts. The 29.57% of the transcriptome retrieved at least one annotation and 11,087 differentially expressed genes (DEGs) were detected between ovary and testis replicates. Six thousand two hundred seven genes were up-regulated in ovaries meanwhile 4880 genes were up-regulated in testes. Candidate genes to be involved in sexual development and gonadal development processes were retrieved from the transcriptome. These sex-related genes were discussed taking into account whether they were up-regulated in ovary, up-regulated in testis or not differentially expressed between gonads and in the framework of previous findings in other crustacean species. Conclusions This is the first transcriptome analysis of P. serratus gonads using RNA-Seq technology. Interesting findings about sex-related genes from an evolutionary perspective (such as Dmrt1) and for putative future aquaculture applications (Iag or vitellogenesis genes) are reported here. We provide a valuable dataset that will facilitate further research into the reproductive biology of this shrimp.

The first edition of the ECA European Crystallography School (ECS1), to be held in Pavia (Italy) during the course of IYCr2014, has found good balance between two apparently diverging goals: (i) to help students and young researchers to find their way in modern science, while keeping a special focus on the molecular and crystalline structure to interpret properties and functionality of materials; (ii) to raise the social and academic awareness of the great advances that crystallography has allowed and will allow to many branches of sciences. Students may choose between two formulae, i.e. a 6-day course with lectures and hands-on sessions held by renowned scientists, covering the state-of-the-art of crystallographic methods, theories and applications, at the same time indicating their future perspectives and cutting-edge aspects, or a 10-day Erasmus Intensive Programme, including a 3-day preparatory course and granting 3 ECTS credits. IYCr2014 is a unique opportunity to stimulate and ignite widespread interest in crystallography; therefore, some frontier seminars will be open to University faculty members, students, and to the general public. Efforts will be made to create a nice and friendly environment, with the goal to provide chances for future collaborations. Students will be invited to bring a poster showing their research results, projects or scientific interests. This will allow students to discuss their ideas with experienced crystallographers and favour aggregation. The programme received good support from scientific institutions and vendors, and a great response from the students: more than 110 pre-registrations from 33 countries were already received at the time this abstract was prepared, showing that there is a real need for both fundamental and advanced teaching in crystallography. We hope that this format will be continued and improved so as to provide a stable, periodic rendezvous for students and researchers under the common theme of crystallography.

Earthworms and leeches are sentinel animals that represent the annelid phylum within terrestrial and freshwater ecosystems, respectively. One early stress signal in these organisms is related to innate immunity, but how nanomaterials affect it is poorly characterized. In this survey, we compare the latest literature on earthworm and leeches with examples of their molecular/cellular responses to inorganic (silver nanoparticles) and organic (carbon nanotubes) nanomaterials. A special focus is placed on the role of annelid immunocytes in the evolutionarily conserved antioxidant and immune mechanisms and protein corona formation and probable endocytosis pathways involved in nanomaterial uptake. Our summary helps to realize why these environmental sentinels are beneficial to study the potential detrimental effects of nanomaterials.

Abstract Diabetes mellitus is one of the most common chronic diseases but also one of the most heterogeneous. Apart the common phenotypes of type 1 and type 2 diabetes, around 1-2% of all cases arise from a single gene mutation and are known as monogenic diabetes. Diabetes diagnosed within the first 6 months of life is known as neonatal diabetes and has been extensively studied during the last two decades. Unraveling the genetic cause and molecular mechanism of this rare diabetes phenotype led to a dramatic change in the treatment of these children who often can be switched from insulin to sulphonylurea treatment. The aim of this paper is to review the known genetic causes of neonatal diabetes and to highlight the most recent aspects of the disease caused by mutations in the KATP and insulin genes, with a special focus on the individualized treatment of these cases

The literary interest in Greenland has been growing steadily in recent years. Both the question of the countryʼs secession from Denmark and the handling of imperialism's historical heritage have occupied the public. The author Kim Leine diligently contributes to the debate with his works on the cultural encounters between Danes and Greenlanders. Following an overview of Leine’s description of the coexistence of colonizers and colonized and the political and religious influence of the former on the latter in the beginning of the 18. century this paper discusses how Leine tries to debunk the common perception of Danes as soft colonizers. The main purpose of the article is to investigate how the difference between the two cultures manifests itself in the father-son relations, with a special focus on the interpretation of the act of sacrifice in the biblical story of Abraham and Isaac. The approach is inspired by postcolonial theory.
