Comparison of the protective resistance induced by60Co-irradiated cercariae and schistosomula of the WFFS and NMRI strains ofSchistosoma mansoni

ABSTRACTMice, CBA/HT6T6 and C57BL/10, were vaccinated with 1 × 350 or 1 × 500Schistosoma mansonicercariae or schistosomula attenuated with 20 or 56 krad60Co irradiation and challenged with 200 cercariae. Protective resistance against homologous strain challenge was compared using the Winches Farm Field Station (WFFS) and Naval Medical Research Institute (NMRI) strains ofS. mansoni. Maximal resistance to challenge was obtained in both strains of mice with cercariae or schistosomula of either WFFS or NMRI strain attenuated with 20 krad. Protection using organisms attenuated with 56 krad was significantly lower. Since previous studies with the two parasite strains have shown that the biological effects of irradiation are similar, the difference in the immunogenicity of the 56 krad-irradiated NMRI strain in this study from earlier studies must be due either to different local conditions for irradiation or to adaptation of the NMRI strain to a new laboratory environment. This finding may have important implications for vaccination studies and investigations of the mechanisms of immunity where radiation-attenuated parasites are used.

Several biological effects of chalcones have been reported to be associated with their thiol reactivity. In vivo, the reactions can result in the formation of small-molecule or protein thiol adducts. Both types of reactions can play a role in the biological effects of this class of compounds. Progress of the reaction of 4-methyl- and 4-methoxychalcone with glutathione and N-acetylcysteine was studied by the HPLC-UV-VIS method. The reactions were conducted under three different pH conditions. HPLC-MS measurements confirmed the structure of the formed adducts. The chalcones reacted with both thiols under all incubation conditions. The initial rate and composition of the equilibrium mixtures depended on the ratio of the deprotonated form of the thiols. In the reaction of 4-methoxychalcone with N-acetylcysteine under strongly basic conditions, transformation of the kinetic adduct into the thermodynamically more stable one was observed. Addition of S-protonated N-acetylcysteine onto the polar double bonds of the chalcones showed different degrees of diastereoselectivity. Both chalcones showed a Michael-type addition reaction with the ionized and non-ionized forms of the investigated thiols. The initial reactivity of the chalcones and the equilibrium composition of the incubates showed a positive correlation with the degree of ionization of the thiols. Conversions showed systematic differences under each set of conditions. The observed differences can hint at the difference in reported biological actions of 4-methyl- and 4-methoxy-substituted chalcones.

The effect of anesthetic drugs on the localization of adult worms in albino mice was compared. The animals with 56 days of infection were anesthetized with pentobarbital sodium, ether or chlorophorm. Perfusion was carried out immediately after, recovering the worms and classifying them in relation to their localization on the liver or portal vein and the mesenteric veins. Our results showed that pentobarbital sodium produced a greater displacement of the worms to the liver (89%) than ether (76%) and chlorophorm (34%) did, when compared to the control group (22%). The difference between pentobarbital sodium and ether was significant (p < 0.05). We suggest that anesthetic drugs may not be used in studies on the distribution of adult worms in several hosts.

SUMMARYAfter a single subcutaneous injection into baboons the MRC-4 strain of trachoma/inclusion conjunctivitis (TRIC) agent underwent limited multiplication at the injection site, but was then eliminated rapidly from the skin and regional lymph nodes. Forty-eight hours after a single intravenous injection, but not thereafter, it appeared in the peripheral lymph nodes and spleen. The single parenteral injections failed to immunize baboons against conjunctival challenge with the homologous strain. These findings contrasted with those previously reported for the more virulent mutant, MRC-4 f, which multiplied readily in the skin, lymph nodes and spleen, persisted in these tissues up to 3 weeks after injection, and conferred good immunity to conjunctival challenge with MRC-4. The difference in behaviour of MRC-4 and MRC-4 f might be accounted for, at least in part, by the use of a smaller inoculum of live MRC-4; but similar findings in guinea-pigs, reported elsewhere, suggest that the differences observed are real. In conjunction with previous work, the present study suggests that the immunogenicity of TRIC agent is closely related to the mass of antigen that can be administered to or propagated within the recipient.We are grateful to Mr D.
