Selective nanomolar binding of biological polyamines to a novel glycoluril-derived molecular tweezer is reported. The high selectivity over their biosynthetic precursor may be of interest, as elevated polyamine levels are linked to several diseases.

The possible removal of copper(II), nickel(II), or lead(II) by an insoluble crosslinked poly(acrylic acid) was investigated in dilute aqueous solution. The binding properties of the polymer were examined at pH = 6.0 or 4.0 with an ionic strength of the medium µ = 0.1 or 1.0 M (NaNO3) using differential pulse polarography as an investigation means. The highest complexing capacity of the polyacid was obtained with lead(II) at pH = 6.0 with µ = 0.1 M, 4.8 mmol Pb(II)/g polymer. The conditional stability constants of the complex species formed were determined using the method proposed by Ruzic assuming that only the 1:1 complex species was formed; for lead(II) at pH = 6.0 and µ = 0.1 M, log K' = 5.3 ± 0.2. It appeared that the binding properties of the polymer increased, depending on the metal ion, in the following order: Ni(II) < Cu(II) < Pb(II). The complexing capacity and log K' values decreased with the pH or with an increase of the ionic strength. These results were in agreement with the conclusions of our previous studies of the hydrosoluble linear analogues. Finally, with the insoluble polymer, the log K' values were comparable to those previously obtained with the linear analogue whereas the complexing capacity values expressed in mmol g-1 were slightly lower.Key words: insoluble crosslinked poly(acrylic acid), copper(II), nickel(II), and lead(II) complexation.

In this paper we report the binding properties, by combined 1H NMR, optical absorption, and fluorescence studies, of a molecular tweezer composed of two Zn(salen)-type Schiff-base units connected by a flexible spacer, towards a series of ditopic diamines having a strong Lewis basicity, with different chain length and rigidity. Except for the 1,2-diaminoethane, in all other cases the formation of stable 1:1 Lewis acid-base adducts with large binding constants is demonstrated. For α,ω-aliphatic diamines, binding constants progressively increase with the increasing length of the alkyl chain, thanks to the flexible nature of the spacer and the parallel decreased conformational strain upon binding. Stable adducts are also found even for short diamines with rigid molecular structures. Given their preorganized structure, these latter species are not subjected to loss of degrees of freedom. The binding characteristics of the tweezer have been exploited for the colorimetric and fluorometric selective and sensitive detection of piperazine.

Rhodamine B thiohydrazide (RBS) was firstly employed as turn-on fluorescent probe for hypochlorite in aqueous solution and living cells. It exhibits a stable response to hypochlorite from 1.0×10-6to 1.0×10-5M with a detection limit of 3.3×10-7M. The response of this probe to hypochlorite is fast and highly selective compared with other reactive oxygen species (such as.OH,1O2, H2O2) and other common anions (such as X-, ClO2-, ClO4-, NO3-, NO2-, OH-, Ac-, CO32-, SO42-).

Synopsis: A well-known tryptophan metabolite, xanthurenic acid, a natural non-fluorescent intermediate siderophore, showed a very selective turn-on response to K+ over other competing metal ions and the detection limit of this natural ionophore was found to be 53 nM at physiological pH.

Molecular tweezers are simple synthetic receptors that are generally composed of two binding domains connected by a spacer group. The non-covalent interactions that occur between the tweezer and its substrate are usually reversible, which facilitates the release of the bound substrate at a target site when triggered by a stimulus such as light, temperature, pH,] or change in chemical potential. In the field of cancer research, one strategy for targeting drug delivery relies on the pH drop in cancerous tissues compared to healthy tissues. We recently showed, for the first time, that it is possible to use pH to tune the binding affinity of molecular tweezers for substrates such as the cancer drug MitoxantroneTM. The molecular tweezer switches conformation from a closed (binding) state to an open (release) state upon acidification. As a result, the targeted delivery of MitoxantroneTM is achieved. This proof of concept shows that molecular tweezers are promising tools for selective drug delivery.
