These study aimed to determine antioxidant activity, total phenolic compound and pigment content of tropical S. platensis, G. arcuata and U. lactuca. S. platensis was extracted with aquadest (SPA), while G. arcuata and U. lactuca were extracted with n-hexane (GAH and ULH), ethyl-acetate (GAE and ULE) and methanol (GAM and ULM). The IC50 value was used to calculate the ability of extract to inhibit free radicals using DPPH (515 nm). The TPC was determined using Folin-Ciocalteu (725 nm). Phycocyanin (620 nm), phycoerythrin (652 nm), allophycocyanin (562 nm), carotenoid (470 nm) and chlorophyll (645 nm and 662 nm) were measured based on color intensity. The results indicated that SPA was able to inhibit free radicals (46.12±2.03%), contains TPC (26.64±0.16 mg/GAE samples) and dominated by phycocyanin pigments (0.301±0.09 mg/g). G. arcuata extracted with ethyl-acetate (GAE) showed the best value with IC50 136.267±0.28 ppm, TPC 303.507±1.155 mg/GAE samples, carotenoid 0.528 ±0.009 μmol/g and chlorophyll a 2.845 ±0.069 mg/g. The U. lactuca extracted with ethyl-acetate also showed the best value with IC50 462.560±2.44 ppm, TPC 210.129±1.387 mg/GAE samples, carotenoid 0.442 ±0.046 μmol/g and chlorophyll a 9.216 ±0.103 mg/g. This results conlcuded that macroalgae extracted with ethyl acetate had better activity which categorized as moderate antioxidant.

Exposure of sunlight lead tropical Sargassum sp. to maintain their growth and moreover to bring up their secondary metabollite for life struggling. Sargassum sp. has bioactive compounds that has a potential antioxidant acitivity such as phenolic compounds as well as chlorophyll and carotenoids. This research was conducted to determine antioxidant activities, phenolic compound and pigmens of Sargassum sp. with different solvent that have different polarities. Sample was macerate with n-heksana, ethyl acetate, methanol and aquadest. All the parameters were done spectrophotometrically. IC50 was used to determine the antioxidant activity by antiradical scavenging activity using DPPH (515 nm). Total phenolic compound were tested by Folin-Ciocalteu solution  and used gallic acid as standard (725 nm). The chlorophylls a content were measured at wavelength 662 nm and 645 nm and carotenoids were measured at wavelength 470 nm. The results showed best IC50 is achieved by aquadest extract (72.95 ±0.22 ppm). The highest Total phenolic compound is achieved by ethyl acetat extract (120.29 ±0,404 mg GAE/g sample). The highest chlorophyll a content is achieved by ethyl acetat extract (18.23 ± 0,049 mg/g sample) and the highest carotenoid content is achieved by ethyl acetat extract extract (60.65 ±0,008  µmol/g sample). It can be concluded that aquadest extract can be categorized as the strong antioxidant and antiradical activity, ethyl acetat as a medium antioxidant activity. The simple and save methods of aquadest extract promising that Sargassum  sp. frorn Indonesia is a good candidate compoud for nutraceutical and cosmeceutical approach.

Pigmen content, total phenolic compound and antioxidant activity Sargassum sp.   Sargassum sp. contains secondary metabolites which potentially act as natural source of antioxidant. The purpose of this study was to determine bioactive contents (pigments, total phenolic compounds), and antioxidant activities of Sargassum sp. The method of this research is descriptive-explorative.  Sample was extracted with methanol, while pigments extraction with aseton 80%.  Chlorophyll a, b and carotenoids were carried out with spectrophotometer method. The total phenolic compounds were analyzed by spectrophotometer method using Follin-Ciocalteu reagent, and antioxidant activities were measured using DPPH (1,1-diphenyl-2- picrylhidrazyl) method. The result of this research showed that methanol extract of Sargassum sp. had the presence of chlorophyll a 2.84 mg/g, chlorophyll b 1.15 mg/g, and carotenoids 2.63 μmol/g sample. The total phenolic compounds of methanol extract were 57.97 mg GAE/g sample. Methanol extract of Sargassum sp. was showing a strong antioxidant activity that could be seen in IC50 value (69,27 ppm) in less than 100 ppm concentration. Sargassum sp. mengandung beberapa metabolit sekunder yang berpotensi sebagai sumber antioksidan alami. Tujuan dari penelitian ini adalah mengkaji komponen bioaktif (pigmen, total fenolik) dan aktivitas antioksidan dari rumput laut Sargassum sp. Metoda penelitian ini adalah diskriptif eksploratif.  Sampel Sargassum sp. basah diekstraksi dengan cara maserasi dengan pelarut  metanol, sedangkan ekstraksi pigmen menggunakan aseton 80%. Penentuan kadar pigmen (klorofil a, b, karotenoid ), total fenolik dan aktivitas antioksidan berdasarkan metode spektrofotometri. Total fenolik diukur berdasarkan uji Follin-Ciocalteu, dan   aktivitas antioksidan  berdasarkan uji DPPH (1,1-diphenyl-2- picrylhidrazyl). Hasil penelitian menunjukan bahwa ekstrak metanol Sargassum sp.  mengandung pigmen klorofil a sebesar 2,84 mg/g, klorofil b sebesar 1,15 mg/g, dan karotenoid sebesar 2,63 μmol/g. Kadar total fenolik ekstrak metanol adalah 57,97 mgGAE/g, sementara aktivitas antioksidannya  (IC50) sebesar 69,27 ppm.  Berdasarkan nilai IC50nya, ekstrak metanol Sargassum sp berpotensi sebagai antioksidan kuat (kurang dari 100 ppm).

Free radicals are chemical species with unpaired electrons in their outer orbital that can attack other molecules, causing cell oxidative damage and degenerative diseases. Free radicals can be prevented by antioxidant. An antioxidant can be found in nature as secondary metabolites in plants, such as carrot (Daucus carota  L.). This research was conducted to study the antioxidant activities of roots, leaves, and stems of carrot extracts using DPPH and FRAP methods, determine total phenolic content (TPC) and total flavonoid content (TFC), analyse the correlation between TPC and TFC with AAI DPPH and FRAP, and the correlation between two methods. The sample was extracted by reflux using n-hexane, ethyl acetate, and ethanol. Determination of TPC, TFC, AAI DPPH and FRAP was performed using UV-visible spectrophotometry. Correlation of TPC and TFC with AAI DPPH and FRAP and also the correlation between the two methods were conducted using Pearson's method. Ethyl acetate carrot leaves extract showed the highest TPC and TFC (8.88 ± 0.44 g GAE/100 g and 9.00 ± 0.31 g QE/100 g). AAI DPPH of carrot extract in the range of 0.16 – 1.42, meanwhile AAI FRAP 1.89 – 5.45. TPC and TFC of carrot roots extract showed a significantly positive correlation with AAI DPPH and FRAP. AAI DPPH and FRAP of carrot roots extract gave a significantly positive correlation. Ethyl acetate and ethanol carrot leave extracts were strong to very strong antioxidant by DPPH and FRAP methods. TPC and TFC in carrot roots extract contributed to antioxidant activities by DPPH and FRAP. DPPH and FRAP presented linear results in antioxidant activities of carrot roots extract.

Extraction of petai peels as natural antioxidants source by domestic microwave macerationPetai (Parkia speciosa) contains antioxidants that can prevent free radicals. Research also proves that petai is able to prevent and even overcome diseases because its seeds contain polyphenols and flavonoids which are known as antioxidant compounds. Flavonoids may serve as a catcher of superoxide anion and lipid superoxide radicals, and also may reduce the activity of superoxide free radicals. The objective of this research was to study the effects of solid to solvent ratio, ethanol concentration, and radiation time on the yield and Total Phenolic Compound (TPC) of extracts. Petai peels was firstly crushed without the addition of water. Petai peels was then used for extraction process in a domestic microwave. Solid to solvent ratio, ethanol concentration, and radiation time were varied. Following extraction, solvent was separated from the solid and then evaporated. The resulting extract powders were analyzed for yield and TPC. Antioxidant activity of extract was also studied as IC50. As result, petai peels extracted at solid to solvent ratio of 1:5, ethanol concentration of 96%, and extraction time of 150 second gave the highest TPC value of 41,43 g GAE /100 g petai peel extracts with IC50 of 364 µg/mL.Keywords: petai peels, domestic microwave, antioxidants, phenolics AbstrakPenelitian membuktikan bahwa petai (Parkia speciosa) mengandung antioksidan berupa polifenol dan flavonoid yang bisa menangkal radikal bebas dan mampu mencegah bahkan mengatasi beberapa macam penyakit serta dapat berfungsi sebagai penangkap anion superoksida dan lipid superoksida radikal, serta mengurangi aktivitas radikal bebas superoksida. Tujuan dari penelitian ini adalah untuk mempelajari pengaruh perbandingan padatan dan pelarut, konsentrasi etanol, dan waktu radiasi terhadap perolehan dan kadar senyawa fenolik atau Total Phenolic Compound (TPC) ekstrak kulit petai yang diperoleh dengan metode ekstraksi domestic microwave maceration. Mula-mula kulit petai diblender tanpa penambahan air. Proses ekstraksi kulit petai dengan pelarut etanol dilakukan dengan bantuan microwave pada variasi perbandingan padatan dan pelarut, konsentrasi etanol dan waktu radiasi. Setelah itu, padatan dipisahkan dengan pelarutnya. Pelarut diuapkan untuk mendapatkan ekstrak serbuk kulit petai. Ekstrak serbuk kulit petai dianalisis untuk mengetahui perolehan dan TPC. Aktivitas antioksidan ekstrak juga dianalisis dan dinyatakan dalam nilai Inhibisi 50% (IC50). Proses ekstraksi kulit petai yang menghasilkan TPC tertinggi adalah pada perbandingan padatan dan pelarut 1:5, konsentrasi etanol 96%, dan waktu radiasi 150 detik dengan nilai TPC yang diperoleh sebesar 41,43 g GAE (ekivalen asam galat) /100 g ekstrak kulit petai dan IC50 sebesar 364 µg/mL.Kata kunci: kulit petai, domestic microwave, antioksidan, fenolik

Objective: Achyranthes aspera, commonly called Sangketan in Indonesian, is a wild plant used as traditional medicine. The roots of Sangketan can beused as a wound healer, which is attributed to the involvement of arginine and its metabolites, nitric oxide, which directly influence the wound healingprocess. The aim of the present study was to determine the potential of Sangketan root extracts to inhibit arginase activity.Methods: The roots were extracted using the multistage ultrasound-assisted extraction method using n-hexane, ethyl acetate, and methanol solvents.Each of the extract associated with the three solvents was tested for arginase inhibition activity using a microplate-based colorimetric method,followed by the determination of total phenolic compound and total flavonoid concentrations.Results: The results of the inhibition test of arginase activity based on n-hexane, ethyl acetate, and methanolic extracts were 9.56, 17.58, and 29.77%,respectively, at concentrations of 100 μg/mL; the total phenolic compound concentrations were 3.91, 4.83, and 11.18 mg GAE/g of sample, respectively,and the total flavonoid concentrations were 0.29, 0.80, and 0.88 mg QE/g of sample, respectively.Conclusion: According to the results, Sangketan root extracts had low potency with regard to arginase inhibitory activity.

Rhizopora mucronata merupakan salah satu jenis mangrove yang berpotensi  sebagai sumber antioksidan alami. Daun tanaman ini mengandung senyawa metabolit sekunder seperti tanin, fenolat, klorofil, karotenoid dan alkaloid. Penelitian ini bertujuan menentukan aktivitas antioksidan ekstrak daun R. mucronata. Sampel diambil dari kawasan mangrove Tugurejo, Semarang dan diekstraksi  secara bertingkat berturut-turut dengan pelarut n-heksana, etil asetat dan metanol. Aktivitas antioksidan diukur dengan metode DPPH (1,1-diphenyl-2-picrylhidrazyl) dan nilainya ditentukan berdasarkan nilai Inhibitory Concentration (IC50)pada panjang gelombang 516,5 nm. Kadar senyawa fenolat total ditentukan secara spektrofotometri pada panjang gelombang 725 nm dengan metode Folin-Ciocalteu, kadar klorofil a dan b ditentukan dengan metode spektrofotometri pada panjang gelombang 663 nm dan 645 nm dan kadar karotenoid diukur pada 480 nm. Hasil penelitian menunjukkan ekstrak metanol memiliki nilai IC50terkecil (113,41  ppm), diikuti ekstrak n-heksana 151,13 ppm dan ekstrak etil asetat 184,78 ppm. Kandungan total fenolat tertinggi terdapat pada ekstrak metanol yaitu 21,06 mg GAE/g sampel, ekstrak n-heksana 13,27 mg GAE/g sampel dan ekstrak etil asetat 2,08 mg GAE/g sampel. Kandungan klorofil a tertinggi terdapat pada ekstrak metanol yaitu 2,304  mg/g, diikuti ekstrak n-heksana 0,705 mg/g dan ekstrak etil asetat 0,64 mg/g. Kandungan klorofil b tertinggi dicapai ekstrak metanol yaitu 0,97 mg/g, ekstrak n-heksana 0,50 mgg dan ekstrak etil asetat 0,13 mg/g. Kandungan karotenoid tertinggi dicapai pada ekstrak metanol yaitu 6,49 mg GAE/g, diikuti ekstrak etil asetat (0.54 mg GAE/g) dan ekstrak n-heksana (1,37 mg GAE/g). Ekstrak metanol memiliki aktivitas antioksidan tertinggi dan termasuk dalam antioksidan kategori sedang, sedangkan ekstrak etil asetat dan  n-heksana termasuk dalam  antioksidan kategori lemah.   Rhizopora mucronata is one type of mangrove that has the potential as a source of natural antioxidants. Total phenolic compound concentration was determined spectrophotometrically at 725 nm wavelength with Folin-Ciocalteu method. The levels of chlorophyll a and b was determined by spectrofotometry method at 663 nm and 645 nm wavelength and carotenoid levels were measured at 480 nm. The results showed that methanol extract had the smallest IC50 value (113,41 ppm), followed by n-hexane extract 151,13 ppm and ethyl acetate extract 184,78 ppm. The highest total phenolic content was found in methanol extract, 21.06 mg GAE / g sample, n-hexane extract 13,27 mg GAE / g sample and ethyl acetate extract 2.08 mg GAE / g sample. The highest content of chlorophyll a contained in methanol extract is 2,304 mg / g, followed by n-hexane extract 0,705 mg / g and ethyl acetate extract 0,64 mg / g. The highest content of chlorophyll b was methanol extract of 0.97 mg / g, n-hexane extract 0,50 mgg and ethyl acetate extract 0,13 mg / g. The highest carotenoid content was achieved in the methanol extract of 6.49 mg GAE / g, followed by ethyl acetate extract (0.54 mg GAE / g) and n-hexane extract (1.37 mg GAE / g). Methanol extract has the highest antioxidant activity and is included in medium category antioxidants, while ethyl acetate and n-hexane extracts are included in weak category antioxidants.

Abstract The snap bean (Phaseolus vulgaris L.) is an economically important legume worldwide due to its good nutritional quality, being considered as a source of protein, fibre, vitamins and minerals. Furthermore, snap beans present high phenolic compound levels and antioxidant activity, factors that contribute in the prevention of the oxidation effects by free radicals. Hence, the present study characterized five snap bean genotypes (UEL 1, UEL 2, UEL 405, UEL 415 and Alessa) with respect to their amino acid concentrations, total phenolic compound contents and antioxidant activities. The amino acid analysis indicated that UEL 415 showed the highest glutamic acid content (12.9 g / 100 g) and UEL 2 the highest histidine content (2.27 g / 100 g). However, only the UEL 1 genotype showed the highest total phenolic compound content (0.492 mg GAE / 100 g) and highest antioxidant activity according to DPPH• radical scavenging (64.71%). The UEL 1, UEL 2 and UEL 415 genotypes presented desirable chemical characteristics for genetic improvement programmes and their commercial exploitation by food industries.

Free radicals are unstable molecules with unpaired electrons in their outer orbitals. An antioxidant is a compound that can be scavenged free radicals. Coffee is one of the natural antioxidants. This research aimed to study the antioxidant activity of medium roasted beans of liberica coffee (Coffea liberica) from three different regions by DPPH and CUPRAC methods. To determine total phenolic content (TPC) and total flavonoid content (TFC), analyze the correlation between TPC and TFC with AAI DPPH and CUPRAC and the correlation between two methods in sample extracts. The sample was extracted by reflux using n-hexane, ethyl acetate, and ethanol. AAI DPPH in the range of 0.397- 18.536, while CUPRAC 0.532-4.674. The highest TPC in ethanol extract of liberica coffee from Aceh (22.585 ± 1.610 g GAE/100 g) and the highest TFC in ethyl acetate extract of liberica coffee from Aceh (4.927 ± 0.355 g QE/100 g). TPC of all samples had a positive and significant correlation with AAI DPPH and CUPRAC. AAI DPPH and CUPRAC value gave a significant and positive correlation. TPC of liberica coffee from three different regions contributed to antioxidant activity by DPPH dan CUPRAC methods. The two methods showed linear results in antioxidant activities of liberica coffee from three regions.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

The antioxidant activities of extracts (n-hexane, dichloromethane, ethyl acetate and methanol) from the leaves and stem barks of Artocarpus scortechinii were evaluated using various biochemical assays. The quantification of the Total Antioxidant Capacity was measured using ferric reducing antioxidant potential (FRAP) and 2,2'-azino-bis(3-ethyl-benzothiazoline-6-sulphonic acid) (ABTS) assays. While, the qualitative of The Total Phenolic Content (TPC) was determined via standard gallic acid calibration graph which was expressed as mg gallic acid equivalent (GAE)/g of dry weight (dw) using Folin Ciocalteau’s reagent. Among all the extracts tested, the methanolic extract of the stem barks showed the highest phenolic content with TPC value of 136.84 mg GAE/g dry weight (dw). FRAP results were expressed as mM equivalent to FeSO4.7H2O by calculating from the standard FeSO4.7H2O calibration graph. The ethyl acetate extract of the stem barks showed the most significant reducing potential in the range between 0.27-2.47 mM FRAP. ABTS+˙ radical scavenging capacity showed that the ethyl acetate extract of the stem barks had the highest scavenging capacity at concentration 1.0 mM with percentage of 90.9%.
