Environmental responsiveness of fibre diameter in grazing fine wool Merino sheep

Fibre diameter, fibre length, and the ratio of fibre length growth to mean fibre diameter (L/D), fibre diameter profile characteristics, and staple strength were examined in 16 fine wool Merino wethers in a 12-month field experiment. Variations in fibre diameter, fibre length, and L/D were shown to be associated with fibre diameter profile characteristics and staple strength. At constant fibre diameter, L/D was significantly positively related to variation in fibre diameter along the staple. A positive correlation between seasonal variation in L/D and variation in diameter between fibres was also observed. Staple length was significantly positively correlated with along-staple variation in fibre diameter and negatively correlated with variation in fibre diameter among fibres. Among-fibre variation in fibre diameter was not significantly correlated with along-staple variation in fibre diameter. Seasonal variation in fibre length growth, fibre diameter, and the ratio of length to diameter throughout the year was associated with increased variation in fibre diameter along the fibre diameter profile and reduced staple strength in grazing sheep. Seasonal variation in fibre diameter was mostly related to mean fibre diameter, L/D, and seasonal variation in fibre length growth rate. Changes in fibre diameter throughout the year were also related to seasonal changes in body weight, fat depth, and skin thickness.

This study examined differences in fibre diameter profiles (FDPs) and midside characteristics of Merino sheep in 2 environments, 4 bloodlines, 3 years, and 44 sire groups. Environment significantly (P < 0.05) influenced all characteristics except one measure of fibre diameter change and staple length. Bloodline also significantly (P < 0.05) influenced all characteristics except staple strength. The maximum fibre diameter, one measure of fibre diameter change, and staple length were significantly different (P < 0.05) between sires. Variation in fibre diameter profile characteristics between bloodlines and sires changed across the environment in which the sheep are maintained. Despite these differences between bloodlines and sires in the FDP, midside mean fibre diameter, fibre diameter variation, and staple length, there were no significant differences between bloodlines in staple strength. The relationships between the FDP and midside characteristics with staple strength were also examined over these bloodlines and environments. Along-staple variation in fibre diameter (r = –0.32 to –0.50), between-fibre fibre diameter variation (r = –0.25 to –0.48), rate of fibre diameter change (r = –0.16 to –0.38), and midside variation in fibre diameter (r = –0.25 to –0.51) were all negatively correlated with staple strength. Maximum fibre diameter (r = 0.08 to 0.18), minimum fibre diameter (r = 0.25 to 0.49), and midside mean fibre diameter (r = 0.09 to 0.35) were positively associated with staple strength. FDP characteristics explained 5–30% more variation in staple strength than could be explained using the standard midside characteristics of mean fibre diameter, fibre diameter variation, and staple length alone. These relationships were also different between environments and bloodlines. The inclusion of the FDP characteristics as explanatory variables provided an alternative interpretation for how absolute fibre diameter and fibre diameter variation combine to explain staple strength. These results suggested that animals and sires might be able to be selected on FDP characteristics to improve staple strength. More detailed genetic studies are required before these selection strategies can be recommended.

The associations between fibre growth characteristics and wool staple strength were investigated in groups (n = 10) of Merino wethers with either low or high staple strength. Sheep grazed together on pastures based on subterranean clover and annual rye grass for about 13 months. The sheep were weighed and injected intradermally with [35 S]-cysteine at about 14-day intervals. Mid-side patches were harvested and dye bands placed in the wool at about 28-day intervals. Patch clean wool growth, pasture digestible dry matter/ha and pasture crude protein/ha had similar seasonal amplitudes of production (287, 286 and 267% of respective minimum). These were significantly higher than the seasonal amplitude in liveweight (24.5%). The seasonal amplitude in fibre diameter was significantly greater than that for rate of fibre elongation (71.4 and 41.4% respectively). This seasonality in fibre length and diameter resulted in statistically significant seasonal fluctuations in the ratio of fibre length growth to fibre diameter. Fortnightly variability in fibre diameter was not significantly related to variability in fibre length growth rate between sheep for individual time periods. However, for the pooled data over the experimental period a statistically significant relationship (R2 = 0.13, P<0.01) was improved with the addition of parameters for sampling time and staple strength group. Staple strengths for the low and high staple strength groups were 25.6 and 32.8 N/ktex respectively (P = 0.057). There were no significant differences between the staple strength groups in seasonal change in liveweight, wool production or fibre parameters measured in this study but the low staple strength group had longer fibres. Staple strength was most highly correlated with mid-side fibre diameter coefficient of variation (R2 = 0.50) followed by seasonal amplitude in liveweight.

A study on the tissue proportions and fibre dimensions of plantation-grown Acacia mangium was carried out. Ten selected trees from two age groups (4- and 8-year-old) were obtained from plantation forests in Selangor. Disks were taken from four sampling heights. Acacia mangium wood is diffuse-porous with mostly solitary vessels. The rays are uniseriate. The average percentage of fibres, vessels and rays of 4-year-old and 8-year-old samples are 85.8%, 9.1%, 5.2% and 84.8%, 9.8%, 5.3%, respectively. Acacia is a shortfibred tropical species. The average fibre length, fibre diameter, fibre lumen diameter and fibre wall thickness are 934, 24, 17 and 3.3 µm for 4-year-old samples and 1017,20, 12 and 4.3 µm for 8-year-old sampies, respectively. The sampies near the pith have the shortest fibres and the length increases toward the bark. Fibre length also tends to decrease with height. The vessel percentage decreases with increasing height.

AbstractPolyamines (putrescine, spermidine and spermine) are required for optimal growth in all cells, and are essential for cell proliferation and growth of cultured wool follicles, with an optimal concentration of spermidine required for the fibre elongation. The effects of a local supply of exogenous spermidine on the rate of cell division in the wool follicles, the length growth rate and diameter of fibres were therefore examined in Merino lambs. Three groups of eight lambs (40 kg) were given food at 1·2 ✕ maintenance. Spermidine was injected intradermally into a small patch (3 ✕ 3 cm) on the left flank three times per day for 7 days at one of three concentrations: 1·38, 2·75 or 4·58 џmol in 0·8 ml volume. The same volume of saline was injected into the contralateral side as a control. The concentration of spermidine in the skin patch 3 h after injection on day 7 increased by proportionately 018, 0·33 or 0·41 (P < 0001) respectively. The rates of cell division in the follicle bulb 3 h after the spermidine injection were proportionately 0104, 0184 and 0·283 higher compared with the contralateral side (P = 0078 overall) for the low, medium and high doses of spermidine respectively and differed between the three doses (P < 005). The fibre length growth rate, as measured using autoradiography, was proportionately 0099, 0117 and 0156 higher than that of the contralateral side (P < 0001 overall) for the low, medium and high doses of spermidine respectively, but differences between doses were not significant (P > 005). Spermidine injection did not result in a significant change in fibre diameter during the treatment period. The ratio of fibre length growth rate to fibre diameter was increased by the injection of spermidine (P < 0001). The results suggest that injecting extra spermidine into the skin altered spermidine homeostasis in the skin, stimulated cell proliferation and resulted in increased fibre growth.

The economically important characteristics of the adult fleece of Merino sheep, such as increases in clean fleece weight, fibre length, fibre diameter and crimp characteristics are determined during critical phases of fetal development of the skin and its appendages. Genetics plays a major role in the development of traits, but the maternal uterine environment could also influence development. Treatment of pregnant ewes with cortisol and its analogues has previously been shown to produce changes in wool follicle morphology. The aim of this study was to determine the effect of transient manipulation of maternal cortisol status during critical phases of wool follicle initiation and development in utero. From Days 55–65 post-conception, singleton-bearing Merino ewes were treated with metyrapone (cortisol inhibitor) or betamethasone (cortisol analogue). Lambs exposed to metyrapone in utero were born with hairier birthcoats than the control or betamethasone treatment groups (P < 0.05), displayed a 10% increase in staple length and a reduction in crimp frequency for the first three shearings (P < 0.05). Co-expression network analysis of microarray data revealed up-regulation of members of the transforming growth factor-β and chemokine receptor superfamilies, gene families known to influence hair and skin development. These experiments demonstrate that presumptive transient manipulation of maternal cortisol status coinciding with the initiation of fetal wool follicle development results in long-term alteration in fleece characteristics, namely fibre length and fibre crimp frequency. These results indicate it is possible to alter the lifetime wool production of Merino sheep with therapeutics targeted to gene expression during key windows of development in utero.

Young Merino sheep (7-8 months of age, weighing about 40.5 kg) were given weekly supplements of 1050 gfsheep of lupins, lupins coated with potassium sulfate (25 g/kg lupins), lupins and access to a mineral lick (offered at 175 g/sheep.week), or lupins coated with potassium sulfate and access to the lick. The sheep grazed an annual pasture at a stocking rate of 15 sheepha and were given the supplements between 14 December and 5 April. The amount of pasture on offer in mid December exceeded 3000 kg DMha but declined to 1500 kg DMha in late March.There were significant amounts of green feed present in December and March (>20% of pasture DM), a small amount in January, and no green feed in February. Despite this the percentage of sheep licking the minerals was over 70% in January , February; and March, with average intake being about 10 g/sheep.day. No significant differences occurred between treatments for liveweights of sheep during the feeding period or at shearing. Over the feeding period sheep lost weight at about 30 g/day. Neither the additional sulfur nor the mineral lick had any significant effects on fleece weight, mean fibre diameter of the fleece, staple length, or staple strength. Wool growth rates were not different between treatments during the feeding period. These results indicate that under good pasture conditions with some green feed available, and when lupins are supplied, responses to mineral supplements are unlikely.

Anthocephalus cadamba is an important tree species for pulp and paper industry. Anatomical studies were conducted between trees of different age viz., 2, 4, 6, 8 and 10 were selected from different plantation. One plus tree from each age was felled for the study. The transverse discs of 10 cm thickness were collected from three different heights of tree representing base, middle and top positions. Anatomical properties like Fibre Length, Fibre Diameter, Fibre Lumen Width, Fibre Wall Thickness, Flexibility, Slenderness ratio, Runkel ratio, Vessel Length, Vessel Diameter, Vessel Area, Vessel Frequency, Ray Height, Ray Width and Ray Frequency were studied. Present study revealed that the average Runkel ratio of Anthocephalus cadamba was 0.48; hence it is suitable for pulp and paper production. Fibre length at the ages of 6, 8 and 10 were at par. Fibre wall thickness were not significantly different at 2, 4, 6 and 8 age but 10 years of age fibre wall thickness differed significantly from other five different ages. Fibre diameter, fibre lumen width and slenderness were at par for all the five ages. Flexibility was at par in the ages of 2, 4, 6, and 8. Best practice of harvesting Anthocephalus cadamba would be at the age of six. Ray and vessel morphology did not show any specific trend from two to ten years old tree.

Genetic, phenotypic and environmental correlations were estimated between 23 quantitative and qualitative wool and body traits of South Australian Merino sheep. Clean fleece weight was significantly correlated genetically with greasy fleece weight (0.62), yield (0.47), staple length (0.41), crimps per inch (-0.34), secondary/primary follicle ratio (0.37) and skin thickness (0.39). No significant half-sib genetic correlation was found between clean fleece weight and fibre diameter and the dam-offspring correlation was quite low, although significant (0.16, P < 0.05). This indicates that selection for clean fleece weight should not lead to any great increase in fibre diameter. Skin thickness is discussed as a possible early indicator of clean fleece weight.

Visually assessed traits for wool quality and ease of management are important to Merino sheep breeders and need to be considered in breeding programs. The present study reports on the predicted correlated responses in visual traits from selection using a range of standard MERINOSELECT™ indices available for use in Australian Merino breeding programs. The effect of including a visual trait in the index is also reported. The visual scored traits were classer grade, wool quality (handle, character and colour), conformation (neck and body wrinkle, front and back leg structure) and fleece rot. The indices covered a range of micron premiums of varying emphasis on increasing fleece weight and reducing fibre diameter. The indices were for Merino wool enterprises (Merino 14%, Merino 7% and Merino 3.5%) and Merino dual purpose wool and meat enterprises (Dual Purpose 7% and Dual Purpose 3.5%). The breeding objectives for the indices included fleece weight, fibre diameter, coefficient of variation of fibre diameter, staple strength, bodyweight and reproduction. Selection over 10 years using the standard MERINOSELECT™ indices is predicted to result in a small correlated improvement in the wool quality traits and classer grade, with a slight deterioration in fleece rot score. The improvement in wool handle, character and colour are greater with those indices that have a greater emphasis on reducing fibre diameter than with those increasing fleece weight. Indices with more emphasis on increased fleece weight than on reduced fibre diameter are predicted to result in small increases in neck and body wrinkle scores and a slight deterioration in wool colour. Inclusion of classer grade, handle, character or colour provided slight extra gains in fleece weight for indices that emphasised reduced fibre diameter through increased accuracy of selection.
