Chemical composition and in vitro digestibility of lines of Lolium perenne selected for high concentrations of water-soluble carbohydrate

Differences in the water-soluble carbohydrate concentrations of herbage of northern European perennial ryegrass cultivars (Aurora, Melle, Cariad) grown under southern Australian conditions, and a New Zealand perennial ryegrass cultivar (Ellett) which yields well in southern Australia, were investigated in relation to their nutritive value. The water-soluble carbohydrates (WSC), total nitrogen, in vitro dry matter digestibility (IVDMD), neutral detergent fibre (NDF), and digestibility of NDF (NDFD) were measured in all cultivars. Aurora and Cariad exhibited higher WSC concentrations than the other cultivars, particularly during summer. This buffered the decline in IVDMD that was due to declining NDFD at that time of the year and resulted in an improvement in IVDMD of between 2 and 6%. Although WSC and nitrogen concentrations of the herbage were negatively correlated, this was due mainly to divergent seasonal variation in these components of the herbage.

Abstract This study examined 169 of the newest varieties of three ryegrass species, perennial (Lolium perenne L.), Italian (Lolium multiflorum Lam.) and hybrid (Lolium boucheanum Kunth), from Recommended List trials in Ireland. The traits examined were yield, dry matter concentration, three nutritive value traits (in vitro dry matter digestibility, water-soluble carbohydrate on a dry matter basis and crude protein concentration) and two ensilability traits (buffering capacity and water soluble carbohydrate concentration on an aqueous phase basis). Varietal monocultures of each species underwent a six cut combined simulated grazing and silage management in each of two years following sowing. Perennial ryegrass yielded less than both other species in one-year-old swards, but less than only Italian ryegrass in two-year-old swards, but generally had the higher in vitro dry matter digestibility and crude protein values. Italian ryegrass displayed the most favourable ensilability characteristics of the three species with perennial ryegrass less favourable and hybrid ryegrass intermediate. Overall, despite the high yields and favourable nutritive value and ensilability traits recorded, the general differences between the three ryegrass species studied were in line with industry expectations. These findings justify assessing the nutritive value and ensilability of ryegrass species, in addition to yield, to allow farmers select species that match farming enterprise requirements.

1. Three sheep fitted with duodenal re-entrant cannulas and three with large rumen fistulas were given red clover, perennial ryegrass and a 2:1 (w/w) mixture of grass and clover in two Latin square arrangements. Measurements were made of voluntary intake, digestibility, flow of nutrients into the duodenum and the flow of Cr-EDTA marker through the reticulo-rumen.2. Organic matter (OM) digestibility was similar for the three feeds but the voluntary intake decreased in the order mixture > red clover > perennial ryegrass. There was an increase in the rate of marker flow from the rumen and a decrease in retention time of the same order. Rumen volume did not change significantly.3. There was a reduction in the mean particle size of rumen contents in the order perennial ryegrass > clover > mixture. The in vitro digestibility of particles decreased with size; the reduction being more rapid for clover than perennial ryegrass.4. The proportion of ingested digestible OM appearing at the duodenum increased from 18.4% to 26.7% to 30.0% for perennial ryegrass, clover and the mixture respectively.5. A higher proportion of digestible cellulose and hemicellulose disappeared over the stomach for the perennial ryegrass feed compared to the clover and the mixture but over 96% of water soluble carbohydrates and starch disappeared over the stomach for all three feeds.6. The apparent digestibility of nitrogen was similar for all three feeds but the proportion of undigested feed N appearing at the duodenum was calculated to be greater by a factor of 1.71 and 2.52 for clover and mixture feeds compared to grass.7. It was concluded that the higher nutritive value of red clover compared to perennial ryegrass was due to an increased rate of flow of nutrients througth the reticulo-rumen and an increase in the proportion of digestible OM digested post ruminally.

Summary. Two lines of perennial ryegrass (Lolium perenne L.), cv. Aurora and breeding line Ba 11351, from the United Kingdom with elevated concentrations of water-soluble carbohydrates in the shoot were compared with the standard cultivars, Ellett, Vedette and Kangaroo Valley, in pure grass swards under irrigation at Kyabram, Victoria, and Gatton, Queensland, and under natural rainfall at Condah, Victoria, during 1995–97. Near infrared reflectance spectroscopy was used to predict the water-soluble carbohydrate, crude protein, in vitro dry matter digestibility, neutral and acid detergent fibre, and Klason lignin concentrations of the perennial ryegrass herbage. Herbage yield and water-soluble carbohydrate differed between cultivars at each site at most harvests, with the high water-soluble carbohydrate lines usually yielding less and having higher water-soluble carbohydrate concentrations than the 3 standard cultivars. However, the high water-soluble carbohydrate lines also had higher water-soluble carbohydrate concentrations at harvests where their yield was equal to the standard cultivars. The other nutritive value traits differed significantly at more than half of the 32 harvests: the high water-soluble carbohydrate lines had higher crude protein and dry matter digestibility, and lower neutral detergent fibre, the neutral detergent fibre containing less acid detergent fibre and lignin than did the standard cultivars. The high water-soluble carbohydrate lines were more susceptible to crown rust during spring and summer than the standard cultivars at Kyabram and Gatton: heavy infections reduced yield, water-soluble carbohydrate, dry matter digestibility and crude protein. Higher water-soluble carbohydrate may depend on only a few genes, as does rust resistance and it seems likely that high yielding, high water-soluble carbohydrate cultivars can be developed by recombination and selection.

Tetraploidy or the use of diploid genotypes with genes conferring high water-soluble carbohydrate concentrations are two mechanisms to increase the nutritive value of perennial ryegrass. This experiment compared the morphology, nutritive value and diurnal variation in water-soluble carbohydrate (WSC) concentrations of 56-day-old plants from six perennial ryegrass cultivars grown under controlled environment conditions. Three of these cultivars were diploid (Melle, Aurora and Cariad) and three were tetraploids (Meltra, Prospero and AberOnyx) which had been derived from the respective diploid cultivars. Two of the diploid cultivars (Cariad and Aurora) had previously been selected for high concentrations of water-soluble carbohydrates. The tetraploid cultivars had fewer (mean 59), larger tillers than the diploids (mean 83). However, with the exception of Melle and Meltra the dry matter yield of the diploid cultivars was not significantly different from their tetraploid derivatives. The effect of tetraploidy on WSC concentrations was dependent on the genetic background of the cultivars. Melle, which had not been previously selected for increased WSC, had a significantly lower WSC concentration than its tetraploid derivative, Meltra. However, tetraploidy did not further increase the WSC concentration in those cultivars previously selected for high WSC concentrations. WSC concentrations in the leaf of both Aurora and Melle rose by 65–70 g/kg throughout the photoperiod, suggesting that differences in the total WSC concentration of these cultivars were not due to any increase in the amount of carbon fixed by Aurora but rather due to differences in the allocation of carbon during growth and development. This experiment demonstrated that tetraploidy was not beneficial in improving the WSC concentration of perennial ryegrass when imposed on two diploid cultivars which had the genetic potential for increased WSC accumulation. However, tetraploidy significantly increased the WSC concentration and by implication the nutritive value of a cultivar derived from a perennial ryegrass cultivar with standard WSC concentrations.

The effects of site and season on the nutritive value of 16 perennial ryegrass cultivars and 60 half-sib families were assessed at 2 locations in South West Victoria. Crude protein, water-soluble carbohydrates, neutral detergent fibre and in vitro digestibility were measured on vegetative herbage, harvested in either autumn or spring. While no heritable genetic variation for nutritive value parameters was detected in this set of families, consistent differences in the nutritive value of cultivars were measured across sites and seasons. The cultivars Yatsyn1 and Ellett were consistently high in both water-soluble carbohydrates and in vitro digestibility. The differences in mean nutritive value between high and low ranking cultivars were ~40 g/kg water-soluble carbohydrates and 3–5% in vitro digestibility. These consistent differences in forage quality demonstrate the value of measuring forage quality during cultivar evaluation. The identification of cultivars with improved nutritive value will also facilitate the crossing of the alleles that confer this improvement into other genetic backgrounds.

The primary growth of eight varieties of three species–white clover (3), red clover (4) and lucerne (1)–was sampled at fortnightly intervals and the percentage dry matter, in vitro digestibility, crude protein, water soluble carbohydrates, P, Ca, K, Na and Mg were determined.Differences between species were nearly always significant and the general order of merit was white clover, red clover and lucerne. The exceptions were for dry-matter percentage where this order was reversed, and red clover had the lowest Na and highest Mg content.

Near-infrared spectroscopy (NIRS) was used to predict the nutritive value of forage species available to the critically endangered northern hairy-nosed wombat (Lasiorhinus krefftii). Nutritive attributes of the forage successfully estimated included total nitrogen concentration, fibre (including neutral detergent fibre, acid detergent fibre and acid lignin), organic matter, water soluble carbohydrates and in vitro dry matter digestibility. The reported results demonstrate the seasonal variability of the forage resource available to L. krefftii in its tropical savanna habitat. Multivariate modelling of the spectra enabled the nutritive value of forage samples to be estimated with coefficients of determination (r2) of 0.770–0.995 and standard errors of the cross-validation of 0.070–2.850 using a modified partial least-squares analysis technique. The standard error of the laboratory was 0.02–1.42. This study demonstrates that broad-based NIRS predictive equations can be used to predict the nutritive value of a number of plant types available to a herbivore over time. By using NIRS the analyst can rapidly analyse large numbers of samples with limited reduction of precision, thereby enabling large-scale ecological applications that may have previously been impeded by time and costs.

Maize cultivation for silage could be a sustainable option in Boreal conditions, especially when combined with nutrient recycling. Effects of digestate (sludge from biogas of domestic origin) application in comparison with synthetic fertilizer and two maturity stages on chemical composition and in vitro digestibility of whole-crop maize were investigated. Starch, neutral detergent fiber, water soluble carbohydrate (WSC) and digestible organic matter (DOM) contents of maize did not differ in response to the two fertilizer treatments. However, starch, DOM and metabolizable energy of maize increased, while ash, crude protein and WSC contents decreased with increasing maize maturity. Heavy metals in maize fertilized with digestate remained low. The results indicate that whole-crop maize fertilized with digestate and harvested at 150 days after sowing is a promising feed and has good nutritive value, even in Boreal conditions.

The effects of paraquat and glyphosate on the nutritive value of dry residues of vulpia [Vulpia bromoides (L.) S. F. Gray], and its subsequent regeneration the following year were examined at Wagga Wagga during the spring, summer and winter of 1986/87 and 1987/88. Paraquat (100 and 200 g a.i./ha) and glyphosate (135 and 270 g a.i./ha) were applied as spraytopping treatments at heading, anthesis and early grain filling stages of vulpia. For both herbicides, time of application was critical to the level of regeneration obtained. Glyphosate gave 84 and 83% control when applied at heading and anthesis respectively, delaying application until early grain filling reduced the level of vulpia control to 28%. Paraquat gave 81% control when applied at anthesis, while delaying application until early grain filling, or earlier application at heading, gave 59% control. Crude protein (CP) and water-soluble carbohydrate (WSC) concentrations, and in vitro organic matter digestibilities (OMD) were measured in vulpia residues collected for 16 weeks after herbicide application. Paraquat increased CP levels most when applied at heading (from 4-8 to 7.2% in 1986, and from 4.9 to 6.5% in 1987). Glyphosate increased CP levels most when applied at heading in 1986 (from 4.8 to 5.4%), but at anthesis in 1987 (from 4.9 to 6.5%). Glyphosate increased WSC most when applied at heading (from 5.7 to 10.6% in 1986, and from 3.5 to 6.3% in 19871, while paraquat reduced WSC in both years. Application of glyphosate at heading was the only treatment to increase OMD (from 50.5 to 54.7%).
