Essential Oil Composition, Free-Radical-Scavenging and Antibacterial Effect from Stems of Ephedra alata alenda in Tunisia

Abstract Background Asparagus flagellaris leaves are ethnomedicinally used to treat syphilis, gonorrhea and other sexually transmitted diseases (STDs), with no reports on the volatile constituents. This study was aimed to quantitatively and qualitatively characterise the composition of essential oil, evaluates the free radical scavenging and antimicrobial capacity of the essential oil. Methods The essential oil was isolated by hydrodistillation method using all-glass Clevenger-type apparatus, while the identification and quantification of constituents were performed by gas chromatography-mass spectrometry (GC-MS) technique. The antioxidant activity on 2, 2-diphenyl-1-picrylhydrazyl (DPPH) was evaluated and the oil was also tested against 10 strains of microorganisms consisting of 6 bacteria: Escherichia coli, Salmonella typhi, Klebsiella pneumonia, Pseudomonas aeruginosa, Staphylococcus aureus and Bacillus subtilis and 4 fungi: Candida albicans, Penicillium notatum, Aspergillus niger and Rhizopus spp. using broth dilution and surface plate methods, respectively. Results A pale yellow essential oil with a characteristic scent was obtained, with a yield of 0.80% (w/w). A total of 28 compounds accounting for 97.41% of the total oil contents were identified. The oil was predominated by Thymol and its derivatives, accounting for 57.48%. The most abundance (% area) constituents of the essential oil were 5-Thymyl tiglate (18.49%), Thymyl-2-methyl butyrate (17.34%), Thymol hydroquinone dimethyl ether (10.52%), Thymol methyl ether (9.42%) and 5-Propyl-1, 3-benzodioxole (4.59%). The essential oil showed a significant free radical scavenging activity compared to the standard antioxidant drugs used in this study, with % inhibition varying from 88.06 ± 0.0001 to 93.05 ± 0.0006. The leaf essential oil exhibited antimicrobial activity on all the tested organisms at 500–125 μg/mL, with an 18–10 mm inhibitory zone. Conclusion The leaf essential oil of A. flagellaris contains notable chemical compounds responsible for its antioxidant and antimicrobial activities.

The genus Sideritis, belonging to the Labiateae family, is represented by more than 150 species distributed mainly in the Mediterranean regions. Member of the genus have been used as beverages, flavorings, and medication. We aimed to shed light on the antioxidant potential of the essential oil of Sideritis rubriflora Hub.-Mor. In this study, antioxidant capacity (DPPH and ABTS radical scavenging, CUPRAC and FRAP, phosphomolibdenum assay, and metal chelating activity) of essential oil of S. rubriflora was investigated with colorimetric methods. Also, essential oil composition of the plant was determined by gas chromatography/mass spectrophotometry (GC-MS). 48 components, representing 95.4% of essential oil of S. rubriflora were identified. β-pinene (10.7%) and Germacrene D (10.7%) were the main constituents of the essential oil. Generally, essential oil of S. rubriflora has shown moderate free radical, reducing power, metal chelating. The reported results supported that the possible use of essential oil of S. rubriflora is a source of natural agents for phytopharmaceutical applications.

In previous work [1], the essential oil of the aerial parts of Marrubium vulgare L. obtained by hydrodistillation was analysed by gas chromatography coupled to mass spectrometry (GC-MS) in order to determine their chemical composition. Fifty (50) components in the oil of M. vulgare were identified. The results demonstrated that the major components of the essential oil were: 4,8,12,16-Tetramethyl heptadecan-4-olid (16.97 %), Germacrene D-4-ol (9.61 %), α- pinéne (9.37 %), Phytol (4.87 %), Dehydro-sabina ketone (4.12 %), Piperitone (3.27 %), δ-Cadinene (3.13 %), 1-Octen-3-ol (2.35 %) and Benzaldehyde (2.31 %). In this study, the antioxidant properties of essential oil were examined. The results showed that this oil can be considered an effective source of antioxidants of natural origin. This is the first report on chemical composition of M. vulgare essential oil cultivated in Algeria and the original study on the antioxidant activity of M. vulgare essential oil. The antioxidant activity was investigated with one method: 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging method.

Aerial parts of Mentha pulegium L. collected from south east of Algeria (Reguiba, El-Oued) possessed an essential oil in 2.34% (v/w) yield. GC and GC-MS analysis of the oil revealed recognition of Thirty-seven compounds, representing 95.02% (area percent) of the total oil composition. Oil was rich in Oxygenated hydrocarbons, exhibited higher percentage of Pulegone (46.31%) followed by Piperitenone (23.3%), Menthone (6.2%) and Limonene (4.7%). The antioxidant activity by DPPH free radical scavenging bioassay (IC50 = 157 µl/ml).
