Maize endosperm consists of about 70 % starch, which makes it an excellent substrate for fermentation. However, due to its low diastatic power, it is used in brewing as an adjunct, mainly. In order to include both red and blue maize as an enzyme source in the brewing process, the effect of temperature and time germination on the diastatic power of malts was studied. The research consisted in a completely randomized three-factor experimental design where the involved factors were colour of maize (blue and red), germination temperature (15, 20, and 25 °C), and germination time (3, 4, 5, 6, 7, 8, and 9 days). The response variables were germination percentage, acrospire length, malting yield, and diastatic power. Data was analysed through Analysis of variance and Comparison Multiple Tukey’s Test. Results showed that both temperature and germination time encouraged the acrospire length, which had a negative effect on malting yield. Regarding to diastatic power, it maintained an increase from third to seventh germination day, at the three tested temperatures. Additionally, as the germination temperature increased, the diastatic power also increased. The highest diastatic power for blue and red maize malts were 39 and 42 °L, respectively, and it was reached when these malts were germinated for 7 days at 25 °C. It was concluded that, by germinating both blue and red maize under the resulting optimum conditions, the obtained malts would be capable of converting their own starch.

The precision of the malting test made in equipment already described (Can. J. Research, C, 15: 204–216. 1937) was studied by making four batches of malt each of which contained duplicate malts made by eight treatments representing the combination of steeping for 48 or 60 hr., maintaining the germination chamber at 53° or 54.5° F., and kilning for 36 hr. at 100° to 175° F. or for 52 hr. at 90° to 165° F. The standard errors of duplicate tests made in the same and in different batches were found to be: extract, 0.08 and 0.09%; moisture, 0.04 and 0.05%; color, 0.04 and 0.05 units; diastatic power 1.0 and 2.8° L.; permanently soluble nitrogen as percentage of wort solids, 0.01 and 0.02%; malting loss 0.06 and 0.29%; and sprouts, 0.06 and 0.18%.On the average, increasing the time of steeping decreased extract by 0.08%; but increased diastatic power by 3.2° L., permanently soluble nitrogen by 0.05%, malting loss by 0.98% and sprouts by 0.44%. Increasing the temperature of the germination chamber increased diastatic power by 4.2° L., permanently soluble nitrogen by 0.04%, malting loss by 0.93%, and sprouts by 0.52%. Increasing the time and decreasing the temperature of kilning increased extract by 0.08% and diastatic power by 14.8° L. Statistical analyses show that the test is sufficiently precise to prove that these effects, though small, are significant.

ABSTRACT   Runtunuwu, S.D. et al. 2008. Assembling Hybrid Coconut of GSK x DMT Based on RAPD (RANDOMLY AMPLIFIED POLYMORPHIC DNA) Marker. Eugenia 14 (1) : 134-152.   The aimed of this research was : 1. assembling hybrid coconut GSK x DMT (Genjah Salak x Dalam Mapanget) that seeds growth was relatifly homogeneous based on RAPD (Randomly Amplified Polymorphic DNA) marker and 2. to found the assembling method of hybrid coconut that will produce massive seeds relatifely short time will homogeneous plant. It was 65 individu trees observe for the average of famale flower per bunch. The result was 25 individu of coconut GSK has the average flower production > 40 per bunch was analyze the homogeneous genetic with the RAPD marker. Based on the analyze RAPD that were 25 individu of GSK coconut trees have the same genetic average 88 % and 14 individu among that was 100 % have same genetic. Further more that 14 individu of GSK was crossing with the 3 individu of DMT that have high yield per year its was DMT 1188, 1172 and 781. Based on the evaluation for the color of buds, high of buds, the steam circle, the petiole color and the germination time of hybrid coconut seeds from the crossing of GSK x DMT 1188 produce more than    70 % seeds that have same genetic, also for crossing of GSK x DMT 1172 have 9 combination and have more than 70 % that same genetic, 10 combination from crossing GSK x DMT 781 have more than 80 % same seeds growth. Therefore, using the RAPD marker were successfully produced 28 crossing of the hybrid coconut GSK x DMT that have relatifly homogeneous seeds growth.   Keywords : assembling, hybrid coconut GSK x DMT, RAPD.
