Discrimination Between Fructus Gardeniae (ZZ) and Fructus Gardeniae Grandiflorae (SZZ) Based on Fingerprint Coupled with Chemometrics and Quantitative Analysis

Abstract Fructus Gardeniae, known as Zhi-zi in China, has been used as Chinese herbal medicine and functional health food for thousands of years. Fructus Gardeniae Grandiflorae, named as Shui-zhizi, is a counterfeit herb of Fructus Gardeniae. In order to discriminate these two varieties, based on ultra-high-performance liquid chromatography, an analysis method of fingerprints of Fructus Gardeniae and Fructus Gardeniae Grandiflorae was established. With hierarchical clustering analysis and principal component analysis, they were separated into two groups. Analyzed with partial least squares discriminant analysis, there were differences in chemical compositions between Fructus Gardeniae and Fructus Gardeniae Grandiflorae. Six compounds, crocin I, genipin-1-β-D-gentiobioside and four other unknown compositions were identified as differential marker compositions between them. Furthermore, seven active substances in them were determined simultaneously. Thus, an integral method of ultra-high-performance liquid chromatography fingerprint combined with chemometrics analysis and quantitative assessment was established. It could be utilized in characterization, quality evaluation of Fructus Gardeniae and could be applied for discriminating Fructus Gardeniae from Fructus Gardeniae Grandiflorae.

Shilajit has a longstanding use as an anti-aging and memory enhancing drug. It is known to have excellent anti-bacterial effects and is believed to be effective for cognitive enhancement, but is difficult to standardize because of the lack of quality control standards. This study, for the first time, proposes a quality control standard using a simultaneous analytical method for the drug’s multi-compound content using high-performance liquid chromatography-ultraviolet detection (HPLC-UV) as an aid for the internationalization of Mongolian Shilajit. Phenolic compounds 1-6 were isolated from Mongolian Shilajit extract using bioassay-guided isolation, and the isolated compounds were evaluated for cognitive-related anti-Alzheimer’s disease (AD) activities using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging, acetylcholinesterase (AChE), butyrylcholinesterase (BChE), β-site amyloid precursor protein-cleaving enzyme 1 (BACE1), and advanced glycation end-product (AGE) formation assays. The isolated compounds showed good effects for each activity. In addition, the isolated compounds were successfully quantified using a validated quantitative HPLC analysis method. As a result, the isolated compounds were suggested as standard marker compounds for Mongolian Shilajit. Also, we proved that the original material of Mongolian Shilajit is a lichen named Xanthoparmelia somloensis (Gyel.) Hale using HPLC-UV, ultra-high-performance liquid chromatography-electrospray ionization/hybrid linear trap-quadruple-orbitrap-high-resolution mass spectrometry (UHPLC-ESI/LTQ-HRMS).

Objective: generalization and analysis of scientific data on the component composition, pharmacological properties and standardization of senna leaves. Materials and methods. The article provides an overview of scientific publications in the journals of the search platforms Scopus and PubMed, as well as the databases of the Russian Science Citation Index. The analysis of the range of herbal medicinal products was carried out according to the data of the State Registers of Medicines of the Republic of Belarus, the Russian Federation and the Republic of Kazakhstan using content analysis, comparison and generalization of data. Results. In addition to the laxative effect, determined by the presence of anthracene derivatives in senna leaves, the complex of their biologically active substances has antimicrobial, hypoglycemic, antioxidant, antifungal and antitumor effects. Also, the chemical composition of the plant is represented by fatty acids, triglycerides, flavonoids, macro- and microelements. The analysis of medicinal herbal preparations based on senna leaves registered in the Russian Federation, the Republic of Belarus and the Republic of Kazakhstan has been carried out. The differences in the methods of their standardization, given in the normative documentation on quality, have been revealed. The scientific experience of using the method of high performance liquid chromatography for the determination of anthracene derivatives in senna leaves is generalized. Conclusion. The complex of biologically active substances of senna leaves exhibits various pharmacological effects, which, possibly, will expand the range of combined preparations based on them. The use of a modern and selective method of high-performance liquid chromatography in the analysis of senna leaves is a very promising direction in the field of standardization of this type of medicinal plant material.

Interesterification becomes a very powerful tool in food industry. A blend of coconut oil and palm stearin is enzymatically interesterified by lipase (EC 3.1.1.3) in an aquarium reactor. The interesterified blend obtained is then incorporated in madeleines, mini croissants, and mini rolls. Physicochemical parameters’ assessment for molasses used is in good agreement with the international standards. Fatty acid composition of the interesterified blend and sugar content of molasses were assessed by gas chromatography (GC) and high performance liquid chromatography (HPLC). A sensory evaluation of the madeleines, mini croissants, and buns has been carried out by untrained tasters, with a statistical analysis by a principal component analysis (PCA). Chromatographic characterization by Gas Chromatography revealed fatty acids, ranging from C6: 0 to C22: 0. Liquid sugar’s content by high performance liquid chromatography revealed three main sugars: sucrose, glucose, and fructose. Results of the sensory analysis showed the good quality of the prepared products.
