Abstract Objectives The purpose of this paper was to construct a reliable methodology to discriminate the geographical origins of Chuanminshen violaceum Sheh et Shan planted in different regions in Sichuan, China. Materials and methods Fatty acid profiles of roots and leaves of C. violaceum planted in various regions of Sichuan Province in China, namely Guangyuan (GY), Langzhong (LZ), Jintang (JT), Bazhong (BZ), and Shuangling (SL), were determined using GC-MS followed by multivariate statistical analyses, including orthogonal partial least-squares discriminant analysis and hierarchical clustering analysis. Results Leaves of C. violaceum showed the highest contents of hexadecatrienoic acid (3.21 g/kg), linoleic acid (6.62 g/kg), and α-linolenic acid (7.24 g/kg), which were all higher than those contained in roots. Chuanminshen violaceum samples collected from LZ, JT, and GY could be clearly distinguished based on fatty acid profiles of leaves and those collected from LZ, GY, and BZ could be clearly distinguished based on fatty acid profiles of roots. Conclusions Chemometric method is used as a potential approach for analyses of fatty acid profiles of roots and leaves to control the quality of C. violaceum and their powered products.

Ultrahigh-performance liquid chromatography Quadrupole-Orbitrap tandem mass spectrometry (UHPLC-Q-Orbitrap-MS/MS) was used to compare the composition of ginsenosides in white ginseng (WG) and extruded white ginseng (EWG). A total of 45 saponins, including original neutral ginsenosides, malonyl-ginsenosides, and chemical transformation of ginsenosides, were successfully identified in both WG and EWG. Multivariate statistical analyses including supervised orthogonal partial least squared discrimination analysis (OPLS-DA) and hierarchical clustering analysis (HCA) were used to analyze components of white ginseng before and after extrusion. As a result, three ginsenosides (malonyl (M)-Rb1, M-Rb2, and M-Rc) were found to be increased in WG, while three ginsenosides (Rb2, Rc, and Rg1) were elevated in EWG. In the OPLS-DA S-plot, the different compositions of ginsenoside that were distinguished between WG and EWG were screened out. Experimental results indicate that the UHPLC-Q-Orbitrap-MS/MS is a useful tool to characterize variations of ginsenosides in WG and EWG.

A precise HPLC-DAD-based quantification together with the metabolomics statistical method was developed to distinguish and control the quality of Fallopia multiflora, a popular medicinal material in Vietnam. Multivariate statistical methods such as hierarchical clustering analysis and principal component analysis were utilized to compare and discriminate six natural and twelve commercial samples. 2,3,4′,5-Tetrahydroxystilbene 2-O-β-D-glucopyranoside (THSG) (1), emodin (4), and the new compound 6-hydroxymusizin 8-O-α-D-apiofuranosyl-(1⟶6)-β-D-glucopyranoside (5) could be considered as important markers for classification of F. multiflora. Furthermore, seven phenolics were quantified that the variation in the contents of selected metabolites revealed the differences in the quality of natural and commercial samples. Recovery of the compounds from the analytes was more than 98%, while the limits of detection (LOD) and the limits of quantitation (LOQ) ranged from 0.5 to 6.6 μg/ml and 1.5 to 19.8 μg/ml, respectively. The linearity, LOD, LOQ, precision, and accuracy satisfied the criteria FDA guidance on bioanalytical methods. Overall, this method is a promising tool for discrimination and quality assurance of F. multiflora products.

The objective of this study was to evaluate the effect of vaccination against gonadotropin-releasing factor (GnRF) on carcass characteristics and meat quality and back fat quality. In total, 400 healthy male pigs were randomly assigned into two treatment groups: physically castrated males (CM) and vaccinated males (IM). CM pigs were physically castrated within 3 days of age. IM pigs received the GnRF vaccine twice, at 14 and 21 weeks of age. At the end of the experiment, 20 pigs of each treatment were weighed individually and slaughtered at 25 weeks of age. The longissimus muscle was collected to evaluate the meat quality including pH, drip loss, shear force, cooking loss, intramuscular fat, fatty acid composition and inosine monophosphate. Subcutaneous fat was sampled to evaluate fatty acid composition. IM pigs had a lower dressing percentage and were leaner (P < 0.01). The inosine monophosphate content was higher in pork from IM pigs than from CM pigs (P < 0.05). IM pigs had a higher content of polyunsaturated fatty acids in longissimus muscles than CM pigs (P < 0.01), mostly because of the higher linoleic acid content (P < 0.01). IM pigs had higher levels of unsaturated fatty acids in subcutaneous fat than CM pigs (P < 0.01), mostly because of the higher linoleic acid and linolenic acid (P < 0.01) and the lower myristic acid content (P < 0.01). There were no adverse effects of the GnRF vaccine on slaughter performance and meat quality of pigs. These results suggest that GnRF vaccine administered to pigs increases the inosine monophosphate content in longissimus muscles and the content of polyunsaturated fatty acids in longissimus muscles and back fat. Consequently, vaccination with Improvac for boar taint control will provide a good alternative to physical castration of male pigs.

A supplement of dietary fat can improve oocyte quality in ruminants. However, to our knowledge, the effect of the type dietary fat (differing in fatty acid profile) on oocyte and embryo production has never been reported in cattle. Therefore, in this study the effect of the type of fat supplement on the number and quality of oocytes collected by ovum pickup (OPU) and on the production of embryos was investigated in Holstein heifers. The experiment was conducted over 2 years: year 1: 8 heifers, and year 2: 10 heifers (16 to 20 months old, body weight 368 � 8.2 kg and BCS 2.3 � 0.1). Heifers were given a diet of hay (67% DM) and a concentrate (33% DM). The concentrate (130 g fat/kg DM) was formulated with either extruded linseeds (L, rich in α-linolenic acid (ω-3 fatty acid),n = 9) or extruded soybeans (S, rich in linoleic acid (ω-6 fatty acid), n = 9). Oocytes were collected by OPU for 6 weeks (2 sessions/week) and their morphological quality was assessed (Q1, excellent; Q2, good; Q3, fair; and Q4, low). The oocytes from one session/week were frozen and those from the other session were used to produce embryos (in vitro maturation and fertilization). Embryo quality was judged following IETS criteria (EQ1, excellent; EQ2, good; EQ3, fair; and EQ4, low). Blood samples were collected weekly throughout the experiment for the analysis of insulin, insulin-like growth factor-1, glucose, non-esterified fatty acids, β-hydroxybutyrate, and urea. Statistical analysis was performed on the growth rate, BCS, hormone, metabolite, and fatty acid data and on the oocyte and embryo data using Split plot ANOVA. Growth rate was not affected by diet (S, 0.93 � 0.21 kg d-1 vs. L, 0.90 � 0.21 kg d-1), and there was no effect of dietary treatment on plasma hormone and metabolite concentrations. However, treatment L increased the proportion of α-linolenic acid (P &lt; 0.0001), and treatment S increased the proportion of linoleic acid (P &lt; 0.0001) in the plasma. Neither the oocyte characteristics (number of oocytes collected, 5.3 � 1.0 vs. 5.7 � 1.0; their quality: Q1, 1.1 � 0.3 vs. 1.0 � 0.3; Q2, 1.8 � 0.3 vs. 1.6 � 0.3; Q3, 1.4 � 0.4 vs. 2.1 � 0.4; and Q4, 1.0 � 0.2 vs. 1.1 � 0.2; and oocytes inseminated, 4.4 � 0.8 vs. 5.1 � 0.8, and cleaved, 3.4 � 0.7 vs. 3.5 � 0.7 per heifer per session; L vs. S, respectively) nor the embryo characteristics (number of embryos, 1.1 � 0.3 vs. 0.9 � 0.3; their quality: EQ1, 0.4 � 0.1 vs. 0.3 � 0.1; EQ2, 0.4 � 0.1 vs. 0.3 � 0.1; EQ3, 0.2 � 0.1 vs. 0.3 � 0.1; and EQ4, 0.04 � 0.03 vs. 0.05 � 0.03 per heifer per session; L vs. S, respectively) were affected by dietary treatment. In conclusion, under our experimental conditions, the type of fatty acid (ω-3 vs. ω-6) does not modify significantly the numbers of oocytes and embryos produced by OPU and their quality in dairy eifers.

Fatty acid profiling, meat and organoleptic quality assessments were conducted on 160 carcasses of crossbred steers born into one of two calving systems and later assigned to one of two postweaning feeding systems. The steers were weaned from either an early calving or late calving system and later assigned to either a rapid-gain feeding (RF) or a slow-gain feeding (SF) system. The RF steers received a silage–hay diet during the backgrounding period prior to finishing, while the SF steers received a hay diet at backgrounding and then grazed alfalfa–meadow bromegrass pasture and annual cereal swaths prior to finishing. All treatment groups received a conventional diet during finishing until the steers attained a target backfat thickness or body weight. Fatty acid analyses were conducted on longissimus dorsi muscle (LDM) and subcutaneous fat samples. Other analyses included LDM composition, tenderness and taste panel evaluations. Total saturated fatty acid was greater (P<0.02) in samples from RF steers, while total monounsaturated fatty acid was greater (P<0.01) in SF steers. The SF steers had greater (P<0.01) conjugated linoleic acid concentration. There was no main or interaction effect (P>0.05) on beef aroma, flavour and tenderness but the SF steaks had lower (P=0.02) cooking losses than RF steaks. The SF strategy has the potential to create a value chain that would lead to finished steers with higher backfat omega-3, conjugated linoleic acid and trans vaccenic acid and less cooking moisture losses.

<p>Western Canadian bison meat is renowned for its natural healthfulness; however, studies on the dietary effects on the fatty acid (FA) profile are limited. Herein, we evaluated the FA profiles of retail bison (<em>longissimus dorsi</em>) representing grain-fed (Grain), grass-fed (Grass) and grass-fed plus oat and pea screening supplement from early (Early-con) or late (Late-con) season harvested finishing regimes. Bison meat contained less than 30 mg fat/g meat, and was lowest for Early-con bison. Proportions of polyunsaturated FA (PUFA) were greatest in Early-con and lowest for Late-con bison. Early-con bison had the greatest proportion of omega-6 (n-6) FA and Late-con bison the lowest, yet as mg/g meat, total n-6 content did not differ. In contrast, Grass and Early-con bison had greater proportions of 18:3n-3, 20:5n-3, 22:5n-3, 22:6n-3 and total omega-3 (n-3) FA. The n-3 content for Grain, Grass, Early-con and Late-con bison were 38, 90, 69 and 69 mg/100 g meat, respectively. The 3:1 n-6/n-3 ratios of Grass, Early-con and Late-con bison were superior to the 7:1 ratio of Grain bison. Proportions of potentially beneficial biohydrogenation intermediates (BI), including <em>t</em>11-18:1 and <em>c</em>9, <em>t</em>11-conjugated linoleic acid, were greater for Early-con and Late-con bison. Proportions of <em>cis</em>-monounsaturated FA were similar for both Grain and Grass bison; however, Late-con was greater than Early-con bison. Cumulatively, Grass, Early-con and Late-con bison were more desirable compared to Grain on account of greater proportions of n-3 FA and a lower n-6/n-3 ratio. Furthermore, seasonal supplementation enhanced the BI proportions with potential beneficial bioactivity in Early-con and Late-con bison.</p>
