Abstract Previous studies suggested the presence of species complex in the so-called Simulium asakoae Takaoka & Davies (Diptera: Simuliidae) in Thailand due to its high morphological variability and genetic divergence. To investigate whether the true S. asakoae is present in Thailand, we performed a detailed morphological identification of S. asakoae and compared its DNA barcodes with the morphospecies S. asakoae from Myanmar and the typical S. asakoae from Malaysia. Phylogenetic analysis revealed the Thai materials analyzed in this study were indeed genetically similar with those from Myanmar and Malaysia, though genetic distances 0–2.27% were observed. We tentatively regard this divergence as intraspecific variation, and the automatic barcode gap discovery analysis further supports them as a single species.

Exploring the effectiveness of DNA barcoding in species identification is prerequisite for biodiversity conservation and environmental monitoring. Aquatic oligochaete could serve as an excellent indicator in aquatic monitoring programs. However, few studies have examined the effectiveness of DNA barcoding in these specific organisms. The mitochondrial COI gene and nuclear ITS2 gene of 83 specimens belonging to 36 species of 18 genera were sequenced in this study. The results showed that there was a barcode gap between species of Naididae, and the intraspecific genetic distances of each species were smaller than interspecific genetic distances. The classification results of ABGD (Automatic Barcode Gap Discovery) were consistent with those of morphological identification except for Tubifex tubifex and Lumbriculus variegatus. All species were successfully distinguished in the phylogenetic tree based on ITS2 gene, which was coincident with morphological result. Our results provided evidence that DNA barcoding can be used as an effective and convenient tool for species identification of the family Naididae and even aquatic oligochaete.

Molecular and cytogenetic data have provided evidence of cryptic speciation in the widespread South American trahira, Hoplias malabaricus. In the present study, karyotypes and DNA barcode sequences of specimens from seven populations inhabiting the lower Amazon River were analyzed in order to characterize the levels of genetic divergence within a single karyomorph. All the specimens presented karyotypes with 2n = 40 chromosomes (20m+20sm) that were consistent with the species' C karyomorph. The DNA barcodes revealed six haplogroups, with clear divergence between populations from Brazil and Argentina. The results support the species complex hypothesis and indicate that a single karyomorph of H. malabaricus may harbor more than one species

Abstract In this study, two new records of goatfishes Upeneus spottocaudalis and U. sundaicus from the South China Sea, combing evidence from morphology and DNA barcodes for species identification. ML tree and NJ tree result showed that the sequences of U. spottocaudalis and U. sundaicus were clustered with the homologous sequences form GenBank, respectively, and the intraspecific genetic distances of U. spottocaudalis (0.2%) and U. sundaicus (0.3%) were less than 2%. Automatic Barcode Gap Discovery (ABGD) analysis also supported this result of classification.

Leuctra adjariae sp. n. and Leuctra georgiae sp. n. (Plecoptera: Leuctridae) are described as two new apterous stonefly species from the Meskheti Range (Lesser Caucasus) in southwestern Georgia. Descriptions and illustrations are provided for both sexes and diagnostic characters are discussed. Males and females of the two species are associated by DNA barcodes. Comparisons with corresponding regions of COI between L. adjariae sp. n. and L. georgiae sp. n. produced K2P genetic distances of 8.38%, values well associated with interspecific variation. The well-supported monophyly as well as results of an ABGD analysis confirms the validity of both new species. Capnioneura gouanerae Vinçon & Sivec, 2011, previously described and known only from Turkey, is reported for the first time for the Caucasus.

Abstract Previous phylogenetic analyses of small ruminant lentivirus (SRLV) sequences found in Poland revealed the circulation of subtype A1 in both sheep and goats, subtypes B1 in goats, and subtypes B2, A12, and A13 in sheep only. This study aimed to analyze the genetic nature of SRLV circulating in sheep and goats from single-species flocks. In order to analyze the degree of genetic variability, the fragments of gag and env genes of 24 SRLV strains were amplified by PCR, cloned into plasmid vectors, sequenced, and consensus sequences were aligned to each other and to reference sequences available from GenBank. Phylogenetic analysis was performed using the Geneious tree-builder tool, and phylogenetic trees were constructed using Mr Bayes (using the general time reversible substitution model) within Geneious Pro 5.3. Pairwise genetic distances were calculated in MEGA 6. Phylogenetic analysis revealed that the strains were highly heterogeneous and represented ovine strains belonging to subtypes A12 and B2 and caprine strains grouped in subtypes B1, B2, A1, and A12. In addition, two novel subtypes, A16 and A17, were found in goats. The mean pairwise genetic distances of gag and env sequences of both clusters were above 15 per cent nucleotide divergence when compared to all other subtypes within group A, which is a criterion required to distinguish a new subtype. Additionally, the existence of two separated clusters was confirmed by high bootstrap values. Co-infections with strains belonging to different subtypes within A and B groups were detected in one sheep and four goats originating from four flocks. Since the co-infection with more than one lentivirus genotype offers an opportunity for viral recombination, the possible recombination events were tested based on RDP analysis. For all co-infected animals, no evidence of recombination was found within the gag gene; however, env sequences showed some recombination patterns in three samples. In conclusion, we have demonstrated extended genetic variability of SRLV in sheep and goats from Poland with the existence of co-infection and recombination events.

AbstractSeveral species of the genus Cryptolestes Ganglbauer, 1899 (Coleoptera: Laemophloeidae) are commonly found in stored products. In this study, five species of Cryptolestes, with almost worldwide distribution, were obtained from laboratories in China, Czech Republic and the USA: Cryptolestes ferrugineus (Stephens, 1831), Cryptolestes pusillus (Schönherr, 1817), Cryptolestes turcicus (Grouvelle, 1876), Cryptolestes pusilloides (Steel & Howe, 1952) and Cryptolestes capensis (Waltl, 1834). Molecular identification based on a 658 bp fragment from the mitochondrial DNA cytochrome c oxidase subunit I (COI) was adopted to overcome some problems of morphological identification of Cryptolestes species. The utility of COI sequences as DNA barcodes in discriminating the five Cryptolestes species was evaluated on adults and larvae by analysing Kimura 2-parameter distances, phylogenetic tree and haplotype networks. The results showed that molecular approaches based on DNA barcodes were able to accurately identify these species. This is the first study using DNA barcoding to identify Cryptolestes species and the gathered DNA sequences will complement the biological barcode database.

Globorotalia truncatulinoides is an extant species of planktic foraminiferans commonly used for stratigraphic and paleoenvironmental analyses. It originated ∼2.8 m.y. ago in subtropical areas of the South Pacific, spread to all subtropical and temperate regions of the world ocean, and expanded its range to southern subantarctic waters between 500 and 200 Ka. The wide geographic distribution of G. truncatulinoides is associated with a latitudinal morphological variability considered as an ecophenotypic variation within a single species. Here, we present the first molecular, morphological, and ecological evidence that G. truncatulinoides corresponds to a complex of four genetic species adapted to particular hydrographic conditions. The different species are separated by significant genetic distances in several ribosomal genes (SSU, ITS-1, 5.8S, ITS-2). Species 1 and species 2 characterize subtropical waters, species 3 is abundant exclusively in the Subantarctic Convergence, while species 4 inhabits subantarctic waters. By using an absolute molecular clock, we deduce the time of divergence between the subtropical and frontal/subantarctic species at ∼300 Ka, which is in agreement with stratigraphic data and suggests an adaptive radiation of the species allowing it to colonize the nutrient-rich and cold subantarctic waters. This genetic dichotomy is associated with a morphological differentiation identified using outline analysis. Species of the same regions are more similar in test shape but can be distinguished by coiling direction. The evolutionary patterns recognized here by combining DNA and morphological analyses from plankton-tow specimens mirror and allow a new interpretation of the data available from Recent sediments. They highlight the importance of adaptation and heterochronic processes, leading to cryptic speciation, in planktic foraminifera.

We report the presence of seven species of Coleoptera newly found on the Maltese Islands in the central Mediterranean. The first species records from the Maltese Islands include: Ancylopus melanocephalus (Olivier, 1808) (Endomychidae), Aplidia transversa (Fabricius, 1801) (Scarabaeidae), Cercyon quisquilius (Linnaeus, 1761) (Hydrophilidae), Hyperaspis duvergeri Fürsch, 1985 (Coccinellidae), Lebia cruxminor (Linnaeus, 1758) (Carabidae), Smicronyx pauperculus Wollaston, 1864 (Curculionidae), and Oxytelus sculptus Gravenhorst, 1806 (Staphylinidae). The morphological identification of each newly reported species was also confirmed through DNA barcoding.

A new species of Moenkhausia is described from tributaries of the rio Paraguay, Brazil. The new species is diagnosed from congeners by characters related to body coloration, the number of lateral line scales, the degree of poring of the lateral line, and number of scales rows above and below the lateral line. Molecular analyses using partial sequences of the mitochondrial gene Cytochrome Oxidase I from specimens of the new species and specimens belonging to morphologically similar species demonstrated that the new species is easily differentiated by their high genetic distance and by their position in the phylogenetic hypothesis obtained through the Maximum Parsimony methodology. The analyses of three samples of M. oligolepis also revealed that they have high genetic distances and belong to different monophyletic groups suggesting that this species corresponds to a species complex rather than a single species.
