Assessment of α-Cypermethrin Pour-On Application and Diminazene Aceturate for Treating Trypanosome-Related Diseases Caused by Tsetse Flies on Cattle in Mô, Togo

Abstract The effects of tsetse-transmitted trypanosomosis control in high tsetse flies (Glossina spp.) challenge and trypanocidal drug resistance settings remain poorly understood in Togo owing to poor data coverage on the current disease impact. From March 2014 to November 2017, a database of zoo-sanitary surveys integrating the evolution of disease incidence and intervention coverage made it possible to quantify the apparent effects attributable to the control effort, focused on all sedentary cattle breeds in the 1,000 km² area of Mô in Togo. The strategy involved an initial phase with cross-sectional entomological and parasitological. Then, three times a year, 20% of the bovine animals of the study area received α-cypermethrin pour-on, and infected cattle with poor health (798 cattle in 2014 and 358 in 2017) were individually given diminazene aceturate at 7 mg/kg of body weight. The tsetse density in the area decreased significantly, from 1.78 ± 0.37 in March 2014 before the α-cypermethrin application to 0.48 ± 0.07 in February 2017. The α-cypermethrin pour-on application and diminazene aceturate treatment of cattle led to the largest reduction in disease incidence, from 28.1% in 2014 to 7.8% in 2017, an improvement in hematocrit from 24.27 ± 4.9% to 27.5 ± 4.6%, and a reduction in calf mortality from 15.9 ± 11% to 5.9%. Improved access to these interventions for different types of livestock and maintaining their effectiveness, despite high tsetse (Diptera: Glossinidae) challenges, should be the primary focus of control strategies in many areas of Togo.

Abstract BackgroundThe effects of tsetse-transmitted trypanosomosis control in high tsetse challenge and trypanocidal drug resistance settings remain poorly understood in Togo, owing to the poor data underlying the current disease impact. This study was implemented in the framework of the PDRI-Mô project which is an African animal trypanosomosis control project using trypanocides and insecticides on cattle. This project, planned by the agricultural ministry, focused on all the sedentary cattle breeds in the 1000 km² area of the prefecture of Mô. Until 2013, the locality remained unconnected to outside areas, with long-term limited access to quality medicines, veterinary services, animal husbandry and new AAT control technologies. Methods From March 2014 to November 2017, a database of zoo-sanitary surveys integrating the evolution of the incidence of the disease and the coverage of interventions made it possible to quantify the apparent effect attributable to control effort in herds. The strategy involved an initial phase with cross-sectional entomological and parasitological surveys, including a rapid trypanocidal drug sensitivity testing. Treatment efficacy was assessed using parasitological status post-treatment of trypanosome-positive animals in each village, randomly assigned to three groups, one treated with 0.5 mg/kg b.w. Isometamidium chloride, the second with 3.5 mg/kg b.w. Diminazene diaceturate and the third with distillated water as control. Using parasitological status of blood sample collected on day 0, day 14 and day 28 post-treatment as the outcome result, trypanosome phenotype resistance of drugs treatment was determined if relapse occur. Then, three times a year, 20% of the herd received α-cypermethrin pour-on, and parasitaemic cattle with poor health were individually given diminazene aceturate at 7 mg/kg of body weight (b.w.).ResultsThe tsetse density in the area decreased significantly (P-value ˂ 0.001) from 1.78 ± 0.37 in March 2014 prior to the α-cypermethrin application to 0.48 ± 0.07 in February 2017. Prior to the trypanocidal treatment, relapse of trypanosome infections in the cattle was approximately 12.5% for diminazene at 3.5 mg/kg of b.w., 35% for isometamidium at 0.5 mg/kg of b.w. and null for diminazeen at 7 mg/kg of b.w. Target deployment led to the largest reduction in disease incidence from 28.1% in 2014 to 7.8% in 2017, an improvement in haematocrit from 24.27±4.9% to 27.5±4.6% and a reduction in calf mortality from 15.9±11% to 5.9%.ConclusionsImproving the access to these interventions for different types of livestock and maintaining their effectiveness in the face of high tsetse challenges should be the primary focus of control strategies in areas of Togo.

 A national sentinel survey of 2006 shows that Enugu State in south-eastern Nigeria is the worst hit by HIV infections. Despite numerous control strategies having been implemented, the state was still reported by another national HIV sentinel survey in 2010 as the worst hit by HIV/AIDS within the zone, and that youths are the most affected. The researchers conducted a cross-sectional descriptive survey to assess the HIV/AIDS knowledge and risk-taking behaviours of youths at a federal university in Enugu, south-eastern Nigeria. They adopted a multistage sampling method to recruit 460 unmarried, consenting students between 15 and 30 years of age, from three faculties of the university. Almost 90 per cent of the university youths had a sound knowledge of key concepts related to HIV/AIDS; despite this, a subset (7.39%) still engaged in risk-taking behaviours. The major risky behaviours identified include having premarital sex, having multiple sexual partners, the sharing of shaving or razor blades, the use of public clippers and tribal marking. Although an analysis of variance demonstrated no significant relationship (t = −0.036) between university youths’ knowledge of HIV/AIDS and their related risk-taking behaviour, risky behaviours were found to be more (13%) among respondents who had insufficient or incorrect knowledge of the disease. An analysis of covariance, however, showed that gender and socio-economic backgrounds were not determinants of the HIV/AIDS risk-taking behaviours of these youths. Therefore, factors such as attitude or culture are areas that should be focused on, and should then, along with educational awareness campaigns, help to reduce the spread and prevalence of the disease.

Abstract Background Glossina (tsetse flies) biologically transmit trypanosomes that infect both humans and animals. Knowledge of their distribution patterns is a key element to better understand the transmission dynamics of trypanosomosis. Tsetse distribution in Rwanda has not been well enough documented, and little is known on their current distribution. This study determined the current spatial distribution, abundance, diversity, and seasonal variations of tsetse flies in and around the Akagera National Park. Methods A longitudinal stratified sampling following the seasons was used. Biconical traps were deployed in 55 sites for 6 consecutive days of each study month from May 2018 to June 2019 and emptied every 48 h. Flies were identified using FAO keys, and the number of flies per trap day (FTD) was used to determine the apparent density. Pearson chi-square (χ2) and parametrical tests (t-test and ANOVA) were used to determine the variations between the variables. The significance (p < 0.05) at 95% confidence interval was considered. Logistic regression was used to determine the association between tsetse occurrence and the associated predictors. Results A total of 39,516 tsetse flies were collected, of which 73.4 and 26.6% were from inside Akagera NP and the interface area, respectively. Female flies accounted for 61.3 while 38.7% were males. Two species were identified, i.e. G. pallidipes [n = 29,121, 7.4 flies/trap/day (FTD)] and G. morsitans centralis (n = 10,395; 2.6 FTD). The statistical difference in numbers was significant between the two species (p = 0.000). The flies were more abundant during the wet season (15.8 FTD) than the dry season (4.2 FTD). Large numbers of flies were trapped around the swamp areas (69.1 FTD) inside the park and in Nyagatare District (11.2 FTD) at the interface. Glossina morsitans was 0.218 times less likely to occur outside the park. The chance of co-existing between the two species reduced outside the protected area (0.021 times). Conclusions The occurrence of Glossina seems to be limited to the protected Akagera NP and a narrow band of its surrounding areas. This finding will be crucial to design appropriate control strategies. Glossina pallidipes was found in higher numbers and therefore is conceivably the most important vector of trypanosomosis. Regional coordinated control and regular monitoring of Glossina distribution are recommended. Graphic Abstract

Introduction: Endoparasites in captive wildlife might pose a threat to public health; however, very few studies have been conducted on this issue, and much remains to be learned, especially in limited-resource settings. This study aimed to investigate endoparasites of captive wildlife in Bangladesh. Perception and understanding of veterinarians regarding one health and zoonoses were also assessed. Materials and Methods: A cross-sectional study was conducted from October 2019 to August 2020. A total of 45 fecal samples from 18 different species of wild animals (i.e., 11 species of mammals: herbivores, carnivores, and omnivores, six birds, and a single reptile species) were collected randomly. Parasitological assessments were done by modified formalin ether sedimentation technique and rechecked by Sheather’s sugar floatation technique. Molecular identification of Spirometra spp. was conducted by amplifying the cytochrome c oxidase 1 (cox1) gene. Questionnaire surveys among 15 veterinarians and an in-depth interview (IDI) with a zoo officer were conducted. Results: Helminths (Spirometra sp., Capillaria sp., Ascaridia/Heterakis, opisthorchiid, strongyles, acuariid, hookworms, roundworms, and unidentified nematode larvae) and protozoa (coccidian oocyst) were identified, and the overall prevalence was 48.9% (22/45). The cox1 sequences (341 bp) of the Bangladesh-origin Spirometra species from lion showed 99.3–99.7% similarity to the reference sequences of Spirometra decipiens (GenBank No: KJ599679.1; MT122766). The majority of study participants (86.6%) agreed about the importance of endoparasite control in zoo animals, and 73.3% expressed that the one health concept should be promoted in Bangladesh. Only 6.7% of veterinarians perceived confidence in diagnosing parasitic diseases and preventing antiparasiticidal resistance. Conclusions: In the present survey, we found a considerable prevalence of endoparasites in captive wildlife. For the first time, zoonotically important S. decipiens from lion was molecularly characterized in Bangladesh. Veterinarian training is required to improve parasite control knowledge and practice. This study highlights the need for routine parasitological assessment, promotion of one health, and improvement of the implementation of current parasite control strategies in zoo animals.

AbstractGoats are critical in mixed smallholder agricultural systems in lower and middle-income countries, while fleas are important human and animal health concerns around the world. Convenience sampling was used to describe and consider risk factors for flea infestations of peri-urban goats, with the aim of informing the iterative development of animal husbandry and management based control strategies. Seven hundred and ninety-two goats were examined in 228 households across 10 peri-urban communities surrounding Blantyre in southern Malawi. The prevalence of Ctenocephalides felis fleas was 18.3, 37.1 and 100% at the levels of individual goats, households and communities, respectively, highlighting a neglected human and animal health concern. Constant introduction of new livestock coupled to a lack of biosecurity within communities, the ubiquitous presence of dog and cat hosts for C. felis, the frequency and thoroughness of cleaning overnight goat accommodation, and goat age less than 12 months old were identified as risk factors for flea infestation. This focal cross-sectional study highlights the significance of fleas in peri-urban communities and uncovers trends and commonalities that are needed to inform sustainable disease management. The majority of the peri-urban goat keepers were female, had resided in the same community throughout their whole life and had primary level education. Advice on the planned management of fleas in livestock needs to be tailored towards this demographic group. This approach affords an opportunity to promote public health measures to address household flea infestations and zoonotic disease spread.

Specific and sensitive quantitative diagnostics, based on real-time (TaqMan) polymerase chain reaction (PCR) and PCR enzyme-linked immunosorbent assay, were developed to detect dry-rot-causing Fusarium spp. (F. avenaceum, F. coeruleum, F. culmorum, and F. sulphureum). Each assay detected Fusarium spp. on potato seed stocks with equal efficiency. Four potato stocks, sampled over two seed generations from Scottish stores, were contaminated with F. avenaceum, F. sulphureum, F. culmorum, F. coeruleum or a combination of species, and there was a general trend towards increased Fusarium spp. contamination in the second generation of seed sampled. F. sulphureum and F. coeruleum caused significantly (P < 0.05) more disease in storage than the other species when disease-free tubers of potato cvs. Spunta and Morene were inoculated at a range of inoculum concentrations (0, 104, 105, and 106 conidia/ml). Increased DNA levels were correlated with increased disease severity between 8 and 12 weeks of storage. The threshold inoculum levels resulting in significant disease development on both cultivars were estimated to be 104 conidia/ml for F. sulphureum and 105 conidia/ml for F. coeruleum. To study the effect of soil infestation and harvest date on disease incidence, seed tubers of cvs. Morene and Spunta were planted in a field plot artificially infested with the four Fusarium spp. F. culmorum and F. sulphureum were detected in soil taken from these plots at harvest, and F. sulphureum DNA levels increased significantly (P < 0.05) at the final harvest. All four Fusarium spp. were detected in progeny tubers. There was a trend toward higher levels of F. culmorum detected in progeny tubers at the earliest harvest date, and higher levels of F. sulphureum at the final harvest. The use of diagnostic assays to detect fungal storage rot pathogens and implications for disease control strategies are discussed.

Abstract A description is provided for Aspergillus fumigatus. Information is included on the disease caused by the organism, its transmission, geographical distribution, and hosts. HOSTS: Common as a saprophyte in soil and fermenting vegetable matter. Pathogenic to mammals (including man), birds and insects. DISEASES: The most serious diseases caused by this fungus are the acute and chronic infections of the respiratory system (aspergillosis, aspergilloma) of man (Austwick, 1965; and many other references, e.g., RMVM 3, 677; 4, 316; 4, 549, 551) and animals (Ainsworth & Austwick, 1959; Austwick, 1965). It may also infect the cardiovascular, digestive, genital, musculoskeletal, nervous, and urinary systems, such infections usually following haematogenous spread of the pathogen from a primary focus in the lungs or air sacs. It is the most common cause of mycotic abortion in cattle (Ainsworth & Austwick, 1959; Austwick, RMVM 5, 660; Dijkstra; RMVM 5, 92) and less often in horses (Hensel et al. ; RMVM 4, 1227). Aspergillus fumigatus may also infect the ear (otomycosis) and mastoid cavity, the eye and sinuses (mycoses). Aspergillosis is a major disease of birds and is a cause of considerable losses in the poultry industry, particularly of very young birds (brooder pneumonia) (Vertinskii et al. ; RMVM 5, 758; Vallejo; 5, 940). It is also common in captive wild birds, especially water birds (Ainsworth & Rewell; RMVM 1, 2090; Dathe; 4, 1948; Saez; 4, 811), but reports of infections in free-living wild birds are more rare (Beer; RMVM 4, 2177; McDiarmid; 2, 1761; Rosen, 5, 558). A. fumigatus also invades eggs. Aspergillus fumigatus spores may induce allergic reactions in man, resulting in hay fever, asthma or allergic bronchopulmonary aspergillosis. It also produces toxins (Ainsworth & Austwick, 1959; Austwick, 1965). GEOGRAPHICAL DISTRIBUTION: World-wide. TRANSMISSION: Soil and air-borne. Disease incidence appears to be related to heavy infection from house dust, mouldy hay or straw, litter, or feeding stuffs (Austwick, 1965; Austwick, RMVM 5, 3b).

In response to increased pest and disease problems, potato farmers use pesticides, which could raise environmental and health concerns. This study sought to promote proper and safe pesticide-handling practices by providing data needed to guide pesticide regulation policy and training for extension staff and farmers. A household survey was conducted in three major potato-growing agroecological zones of Uganda. Two hundred and four potato farmers were interviewed about the type and source of pesticides they use in potato cultivation, the frequency of applications, the use of protective clothing, and cases of pesticide poisoning. The types of pesticides used in potato were fungicides (72%), insecticides (62%), and herbicides (3%). Overall, use of personal protective equipment was low, that is, gumboots (73%), gloves (7%), face masks (16%), and long sleeve shirts (42%). Forty-three percent of farmers who applied pesticides reported having experienced skin itching, 25% skin burning sensation, 43% coughing, 60% a runny nose, 27% teary eyes, and 42% dizziness. An IPM approach involving only moderately to slightly hazardous pesticides when pest and disease incidence has reached economic injury levels and by considering all safety measures during application and storage would be environmentally recommendable and result in reduced health risks.

Purpose Play occupation has been identified as an essential part of children’s lives, and it subsequently features in paediatric occupational therapy. However, few studies address the current place of play and play occupation in occupational therapy practice. This study aims to address this gap in knowledge by exploring paediatric occupational therapists’ perspectives on the place of play and play occupation in occupational therapy practice in Ireland. Design/methodology/approach A cross-sectional online survey was conducted to gather data about the current use of play in the occupational therapy for children under 12 years. Convenience sampling and snowball recruitment techniques were used to recruit paediatric occupational therapists. Data were analysed using descriptive statistics and qualitative content analysis. Findings In total, 65 therapists responded to the survey (estimated response rate, 32%). Results are organised into four sections: demographics and practice context, play assessment practices, use of play in practice and perceived barriers to play-centred practice. Respondents reported that they valued play as a childhood occupation. However, the survey findings identified that the primary focus was on play as a means to an end. Lack of education on play (research, theory and interventions) and pressures in the workplace have been identified as barriers to play-centred practice. Research limitations/implications Findings indicate that there is a mismatch between therapists valuing play as an occupation and how play is used in occupational therapy practice. Unless clarifications are made about play occupation as being different to skills acquisition in childhood, play occupation will continue to get overlooked as an authentic concern of occupation-centred practice. Thus, play as occupation deserves further attention from educators, researchers and practitioners as a means of strengthening occupation-centred practice, in particular play-centred practice in the paediatric context. Originality/value Play has been described as an important occupation in childhood, and consequently, it features in paediatric occupational therapy. However, little is known about the current place of play in occupational therapy practice. This study addresses this gap by considering the current place of play in occupational therapy practice in Ireland.
