AbstractWe report the transmission dynamics of SARS-CoV-2 across different settings from the initial COVID-19 cluster in Brunei, arisen from 19 attendees at the Malaysian Tablighi Jama’at gathering and resulted in 52 locally transmitted cases. Highest non-primary attack rates(ARs) were observed at a subsequent local religious gathering (14.8% [95%CI: 7.1,27.7]) and in the household (10.6% [95%CI: 7.3,15.1]. Household ARs of symptomatic cases were higher (14.4% [95%CI: 8.8,19.9]) than asymptomatic (4.4% [95%CI: 0.0,10.5]) or presymptomatic cases (6.1% [95%CI: 0.3,11.8]). Low ARs (<1%) were observed for workplace and social settings. Our analysis highlights that SARS-CoV-2 transmission varies depending on environmental, behavioural and host factors. We identify ‘red flags’ of potential super-spreading events, namely densely populated gatherings for prolonged periods in enclosed settings, presence of individuals with recent travel history, and group behaviours such as communal eating, sleeping and sharing of personal hygiene facilities. We propose differentiated testing strategies that account for transmission risk.Article summary lineWe highlight the variability of SARS-CoV-2 transmission across different settings, identify settings at highest risk, and characterize the role of environmental, behavioural, and host factors in driving SARS-CoV-2 transmission.

Abstract We sought to assess the proportion of elicited close contacts diagnosed with COVID-19 at the start, and before exiting quarantine, in San Francisco, USA. From June 8th to August 31st, 6946 contacts were identified; 3008 (46.3%) tested, 940 (13.5%) tested positive; 90% tested positive in first 9 days of quarantine.

The Coronavirus Disease-2019 (COVID-19) pandemic, caused by the novel severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), has claimed 1.2 million people globally since December 2019. Although the host factors underpinning COVID-19 pathology are not fully understood, type I interferon (IFN-I) response is considered crucial for SARS-CoV-2 pathogenesis. Perturbations in IFN-I signaling and associated interferon-inducible genes (ISG) are among the primary disease severity indicators in COVID-19. Consequently, IFN-I therapy, either alone or in- combination with existing antiviral or anti-inflammatory drugs, is tested in many ongoing clinical trials to reduce COVID-19 mortality. Since signaling by the IFN-I family of molecules regulates host immune response to other infectious and non-infectious diseases, any imbalance in this family of cytokines would impact the clinical outcome of COVID-19, as well as other co-existing diseases. Therefore, it is imperative to evaluate the beneficial-versus-detrimental effects of IFN-I immunotherapy for COVID-19 patients with divergent disease severity and other co-existing conditions. This review article summarizes the role of IFN-I signaling in infectious and non-infectious diseases of humans. It highlights the precautionary measures to be considered before administering IFN-I to COVID-19 patients having other co-existing disorders. Finally, suggestions are proposed to improve IFN-I immunotherapy to COVID-19.

Several host factors influence HIV-1 infection and replication. The p53-mediated antiviral role in monocytes-derived macrophages (MDMs) was previously highlighted. Indeed, an increase in p53 level results in a stronger restriction against HIV-1 early replication steps through SAMHD1 activity. In this study, we investigated the potential role of some p53 isoforms in HIV-1 infection. Transfection of isoform-specific siRNA induces distinctive effects on the virus life cycle. For example, in contrast to a siRNA targeting all isoforms, a knockdown of Δ133p53 transcripts reduces virus replication in MDMs that is correlated with a decrease in phosphorylated inactive SAMHD1. Combination of Δ133p53 knockdown and Nutlin-3, a pharmacological inhibitor of MDM2 that stabilizes p53, further reduces susceptibility of MDMs to HIV-1 infection, thus suggesting an inhibitory role of Δ133p53 towards p53 antiviral activity. In contrast, p53β knockdown in MDMs increases the viral production independently of SAMHD1. Moreover, experiments with a Nef-deficient virus show that this viral protein plays a protective role against the antiviral environment mediated by p53. Finally, HIV-1 infection affects the expression pattern of p53 isoforms by increasing p53β and p53γ mRNA levels while stabilizing the protein level of p53α and some isoforms from the p53β subclass. The balance between the various p53 isoforms is therefore an important factor in the overall susceptibility of macrophages to HIV-1 infection, fine-tuning the p53 response against HIV-1. This study brings a new understanding of the complex role of p53 in virus replication processes in myeloid cells. Importance As of today, HIV-1 is still considered as a global pandemic without a functional cure, partly because of the presence of stable viral reservoirs. Macrophages constitute one of these cell reservoirs, contributing to the viral persistence. Studies investigating the host factors involved in cell susceptibility to HIV-1 infection might lead to a better understanding of the reservoir formation and will eventually allow the development of an efficient cure. Our team previously showed the antiviral role of p53 in macrophages, which acts by compromising the early steps of HIV-1 replication. In this study, we demonstrate the involvement of p53 isoforms, which regulates p53 activity and define the cellular environment influencing viral replication. In addition, the results concerning the potential role of p53 in antiviral innate immunity could be transposed to other fields of virology and suggest that knowledge in oncology can be applied to HIV-1 research.

One of the effects is not maintaining hygiene during menstruation is able to hitkankes Rahim neck (cervical). Based on data from the World Health Organization (WHO),cervical cancer is the second most cancer in women aged 15-45 years after breast cancer. Noless than 500,000 new cases with 280,000 patient deaths occur each year worldwide.Indonesia was ranked first by the victims died at least 555 women per day and 200,000women annually. This study aims to determine the correlation between knowledge andattitude towards personal hygiene during menstruation action at SMAN 2 Bengkulu City. Thisresearch is descriptive analytic. The number of respondents 84 people with a samplingtechnique that stratified random sampling. Presentation of data is done by using a frequencydistribution table. The collection of data taken using a questionnaire. The data were analyzedusing univariate and bivariate analysis with Chi-Square.The results showed that therespondents are classified as good knowledge of (54.8%), attitude unfavorabel or does notsupport (53.6%), and the biggest acts (52.4%) is good. From the bivariate analysis were foundno correlation between knowledge against acts of personal hygiene during menstruation (p =0.794), and no relation attitude towards personal hygiene actions during menstruation (p =0.975).Required role of schools, educators, parents to be more proactive in enhancingknowledge and useful information about the process of menstruation and how to maintainhygiene during menstruation.

<p><span lang="EN-US">Islam has comprehensive roles in some aspects of human activity. It enlarged from theological aspect to political aspects. Some former colonized countries where Islam was coexisted, this religion became an embryo of nationalist movements during colonization era. This essay scrutinizes the role of Islam in escalating nationalism during colonization era and it relation with the states in post colonization era in four former colonized countries namely Brunei Darussalam, India, Indonesia, and Malaysia. By using comparative method, the essay researched some main literature (library research) related to Islam and nationalism. It was founded that Islam has significant roles in nationalist movement in the four analyzed countries through several channels. Meanwhile, in the post-independence era, the relation between Islam and state system are variably. In India, Islam is separated from state system (secular). In contrast, Islamic ideology became the main sources of state system in Brunei Darussalam (adopted entirely) and Malaysia (adopted partially). Then, Islam in Indonesia seems like “a gray zone” because the country does not using Islamic law but still adopting Islamic thoughts in several cases. </span></p>

Purpose – The purpose of this paper is to analyse the Swedish National Agency for Education’s launch of the nationwide Lgr11 curriculum reform and how local education authorities (LEAs) in one municipality translated and responded to the National Agency’s directives. Design/methodology/approach – This paper presents empirical data from a qualitative study of documents and interviews using the analytical perspective from Scott (2001, 2008) to explore regulative, normative and cultural-cognitive aspects of the National Agency’s communications. To analyse the local translations made by LEAs at the central municipal level, analytical categories of assimilation, loose coupling and transformation were used. Findings – The overall results show that the National Agency primarily communicated its policy instructions to LEAs using normative and cultural-cognitive arguments and directives. The lack of sharper regulative directives, such as for division of labour, decision making, mandates and developmental roles, reduced the potential for LEAs to become influential actors in organising local implementation. An analysis using the assistance of assimilation categories, loose coupling and transformation of the paper also shows that LEAs need system support to accomplish more innovative middle-tier translations through elements of loose coupling and transformation – to become catalysts for school system improvement. Research limitations/implications – The research is designed to understand actions and interpretations within specific institutional, organisational and social settings. Originality/value – This paper contributes to former findings by offering a novel perspective for understanding policy translation and the role of middle-tier intermediaries in decentralised education systems.
