ABSTRACTColor vision requires the activity of cone photoreceptors to be compared in post-receptoral circuitry. Decades of psychophysical measurements have quantified the nature of these comparative interactions on a coarse scale. How such findings generalize to a cellular scale remains unclear. To answer that question, we quantified the influence of surrounding light on the appearance of spots targeted to individual cones. The eye’s aberrations were corrected with adaptive optics and retinal position was precisely tracked in real-time to compensate for natural movement. Subjects reported the color appearance of each spot. A majority of L-and M-cones consistently gave rise to the sensation of white, while a smaller group repeatedly elicited hue sensations. When blue sensations were reported they were more likely mediated by M- than L-cones. Blue sensations were elicited from M-cones against a short-wavelength light that preferentially elevated the quantal catch in surrounding S-cones, while stimulation of the same cones against a white background elicited green sensations. In one of two subjects, proximity to S-cones increased the probability of blue reports when M-cones were probed. We propose that M-cone increments excited both green and blue opponent pathways, but the relative activity of neighboring cones favored one pathway over the other.

An enzyme from leaves of sumach (Rhus typhina) was partially purified that catalyzes the β-glucogallin (l-O-galloylglucose)-dependent galloylation of 1,6-digalloylglucose, thus forming 1,2,6-trigalloylglucose and free glucose. This acyltransferase had a molecular weight of ca. 750,000 and a pH optimum at 5.0-5 .5. Besides β-glucogallin (Km = 3.9 mM ) , also related 1-O-phenylcarboxylglucoses acted as acyl donors. On the other hand, the acceptor substrate, 1,6-digalloylglucose (Km = 0.9 mM ) , could only be replaced by 1,6-diprotocatechuoylglucose (relative activity 46%); however, also tri-, tetra-, and pentagalloylglucoses were galloylated. A pronounced stimulation of the enzymatic reaction was observed upon addition of penta- or hexagalloylglucose into the assay mixtures. The systematic name “ β-glucogallin: 1,6-di-O-galloylglucose 2-O-galloyltransferase” (EC 2.3.1. - ) is proposed for the enzyme


In a previous contribution (Hogben and Slome, 1931) evidence was brought forward to show that the white background response does not depend on the same mechanism of coordination as the black background response, which is produced by reflex liberation of a hormone (“B” substance) of the pars intermedia in the pituitary gland; and experiments pointing to the existence of another internal secretion (“W” substance), connected directly or indirectly with the activity of the pars tuberalis, were described. The existence of separate receptor components of the retina controlling the two systems was left for subsequent enquiry. Of two possible hypotheses concerning the nature of the receptive mechanism, the most likely one is illustrated diagrammatically in fig. 1. In normal situations, when an animal is illuminated on a black background, light can only fall on the floor of the retina. If it is aquatic, the maximum divergence of any two rays which strike the eye is twice the critical angle for air and water, so that in the absence of reflexion of rays from sur­rounding objects below the surface of separation all rays will presumably be brought to a sharp focus in shallow water. There are thus three distinct possibilities which arise from the way in which the animal is illuminated if, as in Xenopus , the eyes are situated on the top of the head: ( a ) in darkness no part of the retina is stimulated, the same being true of the eyeless animal; ( b ) when the animal is exposed to a black background only a sharply localized region of the retina is stimulated; ( c ) when the animal is exposed to a white background the whole of the retina is illuminated owing to the scattering of rays in all directions from the surroundings. For convenience of description the usual black back­ground situation will be described hereafter as one in which only the “floor” of the retina is stimulated, and the white background situation as one in which the floor and the “ periphery ” of the retina are both stimulated together. If then, the receptor elements of the floor and periphery initiate different systems of reflex arcs the phenomena of the background response in Amphibia and Reptiles may be interpreted as follows. In Reptiles we may suppose that stimulation of floor reflexly excites the melanophores to expand, while stimulation of peripheral photoreceptors excites them to contract, being presumably prepotent in the final common path. In Amphibia two alternatives may be con­sidered: ( a ) that floor elements reflexly excite liberation of “B” and that peripheral photoreceptors, being prepotent, reflexly inhibit libera­tion of “B"; ( b ) that floor elements reflexly excite liberation of “B”, and peripheral photoreceptors reflexly excite production of the antago­nistic substance “W” in quantity sufficient to over-ride the effect of “B”. The crucial test of the truth of the general hypothesis that the floor and peripheral elements of the retina initiate different processes of coordina­tion was suggested by Keeble and Gamble (1904-6) in their experiments on Crustacea. If it is true, a normal animal illuminated from below in a black tank with a white top should react in exactly the same way as a normal animal when illuminated in a black tank from above. On the other hand, a normal animal illuminated from below in a black tank with a black top should react like an eyeless animal in the same situation, because the floor elements would not be subject to stimulation. The experiment may be varied as indicated below. In order to obtain signi­ficant results two classes of precautions must be carefully observed. One is that the physical dimensions of the tank must not exceed certain limits, since the maximal divergence of two rays is rigidly fixed when an animal is illuminated from above. The other is that there must be no air-water interface to permit reflexion of the incident rays downwards. Aside from the fact that no bubbles must be allowed to collect, this condition presents a practical difficulty if the animal has to come to the surface to breathe.

The spectacular changes of CH Cyg spectrum during the last period of activity and the flickering in short wavelength light suggest the possibility of rapid line variations (Walker et al.,1969). In November 1982 a series of spectral observations has been accomplished at Rozhen Observatory to investigate the subject.In that period the spectrum of CH Cyg is dominated by type A shell lines. They are supposed to originate in an accretion disk surrounding the hot star of the binary system. The flickering in the optical wavelengths is characteristic for CH Cyg during periods of activity. Evidently it is related with the mass transfer. The investigation of its origin and relations with the other observational characteristics of the star could elucidate the accretion mechanism in this system.

ABSTRACT Whether the short-term regulation of thyroidal protein synthesis by TSH occurs at the transcriptional or the translational level was tested by measuring the effect of actinomycin D (act D) on the TSH-induced stimulation of L-14C-leucine incorporation into the thyroidal proteins of rats. TSH was injected 6 h before the rats were killed. The thyroid glands were then removed and incubated in vitro in the presence of L-14C-leucine for 2 h. The pronounced stimulation of leucine incorporation in the TSH-treated animals was depressed as compared with controls but still significant even when the animals had been pre-treated with 100 μg act D 24 and 7 h before sacrifice. On the other hand, act D strongly decreased incorporation of 3H-uridine into RNA. Short-term regulation of thyroidal protein synthesis by TSH appears to be partly but not wholly dependent on neosynthesis of RNA. Hence regulation may partly occur at the translation level of protein synthesis.

This chapter presents the equations and calculations for energy approximation. It establishes the estimates (261) and (262) of the Main Lemma (10.1) for continuous solutions; these estimates state that we are able to accurately prescribe the energy that the correction adds to the solution, as well as bound the difference between the time derivatives of these two quantities. The chapter also introduces the proposition for prescribing energy, followed by the relevant computations. Each integral contributing to the other term can be estimated. Another proposition for estimating control over the rate of energy variation is given. Finally, the coarse scale material derivative is considered.

Endo-D-galacturonanase of Aspergillus sp. was irreversibly adsorbed on polyethyleneterephthalate in an acetate 0.1 mol l-1 buffer solution of pH 4.2. Immobilization of the enzyme resulted in lowering of its activity, the measure of which depended on the amount of the enzyme fixed on the carrier. The highest relative activity (42.4%) had the preparation containing 5.25 mg of the enzyme per 1 g of the carrier. The velocity and intensity of the sorption of the enzyme depended on the ionic strength of the medium, whilst pH, on the other hand, was of no influence. Endo-D-galacturonanase immobilized in a 0.1 mol l-1 buffer was characteristic a) of its fixation strength in salt solutions of various ionic strength and pH, in a 3 mol l-1 guanidine solution, and also in sodium pectate and pectin solutions, b) of its high stability during a long-lasting storage at 4 °C, c) of its operational stability. The immobilization led to a partial change of the action pattern onto the high-molecular substrate, manifested in lowering the decrease of viscosity to degradation degree ratio.

The mechanism whereby gastrin-type receptor and muscarinic M3-type receptor regulate free intracellular Ca2+ concentration ([Ca2+]i) was studied in rabbit gastric parietal cells stimulated by either gastrin or carbachol. Both agonists induced a biphasic [Ca2+]i response: a transient [Ca2+]i rise, followed by a sustained steady state depending on extracellular Ca2+. Gastrin and carbachol also caused a rapid and transient increase in Mn2+ influx (a tracer for bivalent-cation entry). Pre-stimulation of cells with one agonist drastically decreased both [Ca2+]i increase and Mn2+ influx induced by the other. Neither diltiazem nor pertussistoxin treatment had any effect on agonist-stimulated Mn2+ entry. Thapsigargin, a Ca(2+)-pump inhibitor, induced a biphasic [Ca2+]i increase, and enhanced the rate of Mn2+ entry. Preincubation of cells with thapsigargin inhibits the [Ca2+]i increase as well as Mn2+ entry stimulated by gastrin or by carbachol. Thapsigargin induced a weak but significant increase in Ins(1,4,5)P3 content, but this agent had no effect on the agonist-evoked Ins(1,4,5)P3 response. In permeabilized parietal cells, Ins(1,4,5)P3 and caffeine caused an immediate Ca2+ release from intracellular pools, followed by a reloading of Ca2+ pools which can be prevented in the presence of thapsigargin. We conclude that (i) gastrin and carbachol mobilize common Ca2+ intracellular stores, (ii) Ca2+ permeability secondary to receptor activation involves neither a voltage-sensitive Ca2+ channel nor a GTP-binding protein from the G1 family, and (iii) agonists regulate common Ca2+ channels in depleting intracellular Ca2+ stores.

AbstractWe investigated morphological changes of retinal arteries to determine their association with the blood flow and systemic variables in type 2 diabetes patients. The patients included 47 non-diabetic retinopathy eyes, 36 mild or moderate nonproliferative diabetic retinopathy (M-NPDR) eyes, 22 severe NPDR (S-NPDR) eyes, 32 PDR eyes, and 24 normal eyes as controls. The mean wall to lumen ratio (WLR) measured by adaptive optics camera was significantly higher in the PDR groups than in all of the other groups (all P < 0.001). However, the external diameter of the retinal vessels was not significantly different among the groups. The mean blur rate (MBR)-vessel determined by laser speckle flowgraphy was significantly lower in the PDR group than in the other groups (P < 0.001). The WLR was correlated with MBR-vessel (r = − 0.337, P < 0.001), duration of disease (r = 0.191, P = 0.042), stage of DM (r = 0.643, P < 0.001), systolic blood pressure (r = 0.166, P < 0.037), and presence of systemic hypertension (r = 0.443, P < 0.001). Multiple regression analysis demonstrated that MBR-vessel (β = − 0.389, P < 0.001), presence of systemic hypertension (β = 0.334, P = 0.001), and LDL (β = 0.199, P = 0.045) were independent factors significantly associated with the WLR. The increased retinal vessel wall thickness led to a narrowing of lumen diameter and a decrease in the blood flow in the PDR group.

If you would rather say to yourself as the Secretary of State said to the Air Conference, “ There won't be another war for ten years, so why worry? ” then no doubt you will think with him that it is better to let other nations have alk the bother and expense of trying to advance; after all, we are jolly fine fellows and can soon pick up. If, on the other hand, you have imagination which gives you a nasty queasy sensation when you think of what might be, then perhaps the following notes, albeit disjointed and mostly stale, may at least conjure up in you thoughts of your own on the subject. This is all that is needed to help, our advancement in the air–the stimulation of spoken and written thoughts by the British nation, for if every taxpayer in the British Empire says “ Air Force,” then the Press and Parliament will say it too.
