AbstractExisting methods to detect domestic triatomines have low sensitivity. As early house infestation detection is epidemiologically important, the exploration of better methods is required. Hence, we measured the attractiveness of a yeast-baited trap to adults and nymphs of Triatoma infestans, under laboratory conditions.The assays were conducted in an experimental arena, with an experimental and a control traps placed at opposite sides and one refuge in the center area. Insects where released and the number of triatomines in the yeast and control traps were counted, after 3, 6 and 24 hours of the beginning of the experiment. We use generalized linear models within a multimodel inference approach to model the number of insects in the trap, using insect age classes, time after assay initiation and date of the experiment as predictors.Our results show that the attraction to CO2 depends upon the life stage of the insects. During the 24 hours of experiment a constant number of adults were attracted to the yeast trap, while nymphs show attraction only up to the first three hours after the initiation of CO2 liberation. Undoubtedly, the orientation response to chemical cues deserves further studies to be fully understood.

While the control of the major Chagas' disease vector Triatoma infestans has been achieved in many endemic areas of Brazil, data from the inception of the control program in the Triângulo Mineiro (1976-79) suggest that re-infestation by triatomines occurs under certain favorable conditions. The percentage of houses infested in 500 communities of the Triângulo Mineiro region is compared for two years: 1976 and 1979, using linear regression models. Controlling for three major triatomine vectors, household crowding, house demolition and construction, and infested out-buildings are all independent covariates of house infestation in these communities. While several household factors have been suggested as correlates of infestation, the control program focuses on community-level reductions in infestation, but intra-community or regional comparisons have heretofore been unfeasible. Computerized data are becoming available to identify communities at high risk for re-infestation, and for targeting control-program activities.

The beetle Astylus variegatus (Germ.) (Coleoptera: Melyridae) is frequently found in flowers feeding on pollen. Responses of A. variegatus to volatile floral attractants were studied in common beans (Phaseolus vulgaris L.) fields. Traps originally designed to capture Diabrotica speciosa (Germ.) (Coleoptera: Chrysomelidae), consisted of plastic bottles (2 L) with 150 holes (5-mm diameter) yellow gold painted and containing inside a plastic strip (3.5 <FONT FACE=Symbol>´</FONT> 25 cm) with Lagenaria vulgaris (L.) powder (0.28% B cucurbitacin - feeding stimulant and arrestant for diabroticites) sprayed with carbaril insecticide. Treatments consisted of 1,4-dimethoxybenzene (one or two dispensers per trap), 1,4-dimethoxybenze + indole, 1,4-dimethoxybenzene + cinnameldehyde and control. Volatile average release rates (over ten days) was approximately 32 mg day-1 per dispenser under laboratory conditions. 1,4-dimethoxybenzene-lured traps caught significantly more beetles than the control, three and seven days after trap setting. Ten days after the onset of the experiment, there were no differences in number of beetles caught by treatments. Captures were higher in the 1,4-dimethoxybenzene + cinnamaldehyde treatment than in 1,4-dimethoxybenzene only in the first assessment. Adding indole to 1,4-dimethoxybenzene did not improve beetle captures.

Volatile organic compounds (VOCs) are small molecular mass metabolites which compose the volatilome, whose analysis has been widely employed in different areas. This innovative approach has emerged in research as a diagnostic alternative to different diseases in human and veterinary medicine, which still present constraints regarding analytical and diagnostic sensitivity. Such is the case of the infection by mycobacteria responsible for tuberculosis and paratuberculosis in livestock. Although eradication and control programs have been partly managed with success in many countries worldwide, the often low sensitivity of the current diagnostic techniques against Mycobacterium bovis (as well as other mycobacteria from Mycobacterium tuberculosis complex) and Mycobacterium avium subsp. paratuberculosis together with other hurdles such as low mycobacteria loads in samples, a tedious process of microbiological culture, inhibition by many variables, or intermittent shedding of the mycobacteria highlight the importance of evaluating new techniques that open different options and complement the diagnostic paradigm. In this sense, volatilome analysis stands as a potential option because it fulfills part of the mycobacterial diagnosis requirements. The aim of the present review is to compile the information related to the diagnosis of tuberculosis and paratuberculosis in livestock through the analysis of VOCs by using different biological matrices. The analytical techniques used for the evaluation of VOCs are discussed focusing on the advantages and drawbacks offered compared with the routine diagnostic tools. In addition, the differences described in the literature among in vivo and in vitro assays, natural and experimental infections, and the use of specific VOCs (targeted analysis) and complete VOC pattern (non-targeted analysis) are highlighted. This review emphasizes how this methodology could be useful in the problematic diagnosis of tuberculosis and paratuberculosis in livestock and poses challenges to be addressed in future research.

Taking point from a post-9/11 spiritual warfare narrative in which models of asymmetric war are used to reconceptualize the demonic, the Introduction argues that figures of the demonic are both consolidating and deconstructive of systems of power, particularly those tied to sovereignty, identity, and empire. Weaving together two definitions of demonology, by Bruce Lincoln and Marcella Althaus-Reid, respectively, it demonstrates that demonology operates as a rubric of knowledge aimed at the classification, comprehension, and control of nonhuman and dehumanized others—the demonized—who simultaneously unsettle those rubrics of knowledge by exposing their categories as constructed and not natural. Mobilizing queer and critical race theory, it then situates the demon’s deconstructive quality in its figuration of passing and counterfeiture, which unsettle territorial boundaries, stable identities, and linear models of temporality.

By immunizing a mouse with human metastatic breast tumor cells from patient effusions and infiltrated lymph nodes, a monoclonal antibody (MLuC2), which identifies a new carcinoma-associated marker, was raised. The reactivity of this reagent was studied by immunohistochemistry on live and fixed cells from tumor cell lines and on frozen sections from surgical specimens. Besides reacting with 73% of breast carcinomas, MLuC2 also reacted with 93% of non-small cell lung carcinoma (NSCLC) and with a few normal tissues. The MLuC2-recognized molecule (CaMLuC2), whose MW was 90 KDa according to immunoblotting experiments, was found to be detectable in the serum and could therefore be of particular interest for serological diagnostic applications. Since the CaMLuC2 epitope was not polyexpressed on the bearing molecule, we produced a new generation of MAbs in order to define epitopes coexpressed with CaMLuC2 on the same 90 KDa molecule, and which are therefore suitable to develop a double-determinant immunoradiometric assay (DDIRMA) for the detection of this marker in the sera of lung carcinoma patients. Different analyses by immunohistochemistry, binding inhibition tests and DDIRMA, proved that the two new reagents developed, MLuC8 and MLuC9, recognize the same or closely related epitopes, which are however different from CaMLuC2, but which are all present on the same molecule. Preliminary immunoradiometric tests performed on sera from lung cancer and control patients showed a good specificity but a low sensitivity. In fact, only 42% of the 28 tested sera samples from NSCLC patients scored positive despite the fact that more than 90% of the NSCLC expressed the relevant antigen

Background: Many graft configurations are clinically used for valve-sparing aortic root replacement, some specifically focused on recapitulating neosinus geometry. However, the specific impact of such neosinuses on valvular and root biomechanics and the potential influence on long-term durability are unknown. Methods: Using a custom 3-dimenstional–printed heart simulator with porcine aortic roots (n=5), the anticommissural plication, Stanford modification, straight graft (SG), Uni-Graft, and Valsalva graft configurations were tested in series using an incomplete counterbalanced measures design, with the native root as a control, to mitigate ordering effects. Hemodynamic and videometric data were analyzed using linear models with conduit as the fixed effect of interest and valve as a fixed nuisance effect with post hoc pairwise testing using Tukey’s correction. Results: Hemodynamics were clinically similar between grafts and control aortic roots. Regurgitant fraction varied between grafts, with SG and Uni-Graft groups having the lowest regurgitant fractions and anticommissural plication having the highest. Root distensibility was significantly lower in SG versus both control roots and all other grafts aside from the Stanford modification ( P ≤0.01 for each). All grafts except SG had significantly higher cusp opening velocities versus native roots ( P <0.01 for each). Relative cusp opening forces were similar between SG, Uni-Graft, and control groups, whereas anticommissural plication, Stanford modification, and Valsalva grafts had significantly higher opening forces versus controls ( P <0.01). Cusp closing velocities were similar between native roots and the SG group, and were significantly lower than observed in the other conduits ( P ≤0.01 for each). Only SG and Uni-Graft groups experienced relative cusp closing forces approaching that of the native root, whereas relative forces were >5-fold higher in the anticommissural plication, Stanford modification, and Valsalva graft groups. Conclusions: In this ex vivo modeling system, clinically used valve-sparing aortic root replacement conduit configurations have comparable hemodynamics but differ in biomechanical performance, with the straight graft most closely recapitulating native aortic root biomechanics.
