Ruminants derived products have a prominent role in diets and economy worldwide; therefore, the capability to control the rumen microbial ecosystem, for ameliorating their quality, is of fundamental importance in the livestock sector. The aim of this study was to evaluate the effect of dietary supplementation with chestnut and quebracho tannins on microbial community and fatty acid profile, in the rumen fluid of dairy ewes. Multivariate analysis of PCR-DGGE profiles of rumen microbial communities showed a correlation among the presence of chestnut or quebracho in the diet, the specificButyrivibriogroup DGGE profiles, the increase in 18:3cis9,cis12, andcis15; 18:2cis9 andcis12; 18:2cis9 andtrans11; 18:2trans11 andcis15; and 18:1trans11 content, and the decrease in 18:0 concentration. Phylogenetic analysis of DGGE band sequences revealed the presence of bacteria representatives related to the generaHungatella,Ruminococcus, andEubacteriumand unclassified Lachnospiraceae family members, suggesting that these taxa could be affected by tannins presence in the diets. The results of this study showed that tannins from chestnut and quebracho can reduce the biohydrogenation of unsaturated fatty acids through changes in rumen microbial communities.

ABSTRACT The microbial community composition of colonized cotton and leachate samples from a landfill was quantified using small subunit (SSU) rRNA probes (quantitative rRNA hybridization). Relative quantification of bacteria, eukaryotes, and archaea revealed variations in the landfill microbial community between samples from different areas of the landfill site and indicated the presence of potentially novel archaea. Anaerobic fungi were quantified in rumen fluid samples but were not sufficiently abundant for direct detection in the landfill samples.

The effect of Saccharomyces cerevisiae fermentation products (SCFP) on improving growth and health of calves could be attributed to the ability of SCFP to modulate the microbiota in the gastrointestinal tract (GIT). However, the changes in microbial community along the gut in calves supplemented with SCFP have not been investigated extensively. The aims of this study were to investigate the effect of SCFP on microbial communities in each sites of GIT using high-throughput sequencing technique. Fifteen Holstein male calves were used and randomly assigned to 1 of the 3 treatments including a calf starter containing 0 (Control, CON), 0.5 (SCFP1) or 1% SCFP (SCFP2, Original XPC, Diamond V, Cedar Rapids, IA, USA) of dry matter from day 4 to 56. The supplemented calves were fed with an additional 1 g/d SCFP (SmartCare, Diamond V, Cedar Rapids, IA, USA) in milk from day 2 to 30. Rumen fluid was sampled at day 28 of age via esophageal tube. All calves were slaughtered and gastrointestinal samples collected on day 56. Inclusion of SCFP increased the microbial species richness in the large intestine. The SCFP also affected the bacterial community at an early age in the rumen and later in rectum microbiota. Supplementation of SCFP stimulated colonization by fibrolytic bacteria (Lachnospiraceae and Ruminococcaceae) in rumen and large intestine, respectively. No differences were found between SCFP1 and SCFP2. This is the first study to analyze the effect of SCFP on bacterial community of the GIT microbiota in calves. The results provide the basic bacterial community information, which helps us understand the mechanism of action of SCFP for improving the health and performance of pre-weaning calf.

Recently, transgenic crops have been considered as possible donors of transgenes that could be taken up by micro-organisms under appropriate conditions. In anin vitrorumen simulation system, effects of ampicillin on microbial communities growing either on rumen contents with transgenic maize carrying a gene that confers resistance to ampicillin or its isogenic counterpart as substrates were examined continuously over 13 d. Rate of production of SCFA was measured to determine functional changes in the rumen model and single-strand conformational polymorphism was used to detect alterations in structure of the microbial community. Rumen contents treated with ampicillin displayed a marked decrease in the rate of production of SCFA and diversity of the microbial community was reduced severely. In the presence of transgenic maize, however, the patterns of change of rumen micro-organisms and their metabolic profiles were different from that of rumen fluid incorporating maize bred conventionally. Recovery of propionate production was observed both in the rumen fluid fed transgenic and conventional maize after a delay of several days but recovery occurred earlier in fermenters fed transgenic maize. Alterations in the microbial population structures resulting from the ampicillin challenge were not reversed during the experimental run although there was evidence of adaptation of the microbial communities over time in the presence of the antibiotic, showing that populations with different microbial structures could resume a pre-challenge metabolic profile following the introduction of ampicillin, irrespective of the source of the plant material in the growth medium.

Microbial communities drive diverse processes that impact nearly everything on this planet, from global biogeochemical cycles to human health. Harnessing the power of these microorganisms could provide solutions to many of the challenges that face society. However, naturally occurring microbial communities are not optimized for anthropogenic use. An emerging area of research is focusing on engineering synthetic microbial communities to carry out predefined functions. Microbial community engineers are applying design principles like top-down and bottom-up approaches to create synthetic microbial communities having a myriad of real-life applications in health care, disease prevention, and environmental remediation. Multiple genetic engineering tools and delivery approaches can be used to ‘knock-in' new gene functions into microbial communities. A systematic study of the microbial interactions, community assembling principles, and engineering tools are necessary for us to understand the microbial community and to better utilize them. Continued analysis and effort are required to further the current and potential applications of synthetic microbial communities.
