A total of 510 service water samples from cooling towers throughout South Africa were analysed for the presence of Legionella bacteria. Legionella was detected using an immuno-labelling technique based on the most probable number principle. Only cultural (viable) bacteria were counted. Legionellae were found in most of the samples tested. However, in only 4% of the samples a high level of legionellae was recorded. No correlation was found between the numbers of legionellae and those of standard plate counts. Biocide treatment was shown to be effective in the removal of the bacteria from cooling towers after a 3-month treatment period.

During the last decade, application of molecular methods using cultivation-independent approaches has provided new insights into the composition and structure of denitrifying communities in various environments. However, little is known about their abundance, and quantification is still performed using cultivation-based approaches, which are not only biased by the inability to cultivate of many micro-organisms but also fastidious and time-consuming. Two types of cultivation-independent approaches have recently been developed to quantify denitrifiers. The first type, which is based on the hybridization technique, comprises the use of Southern hybridization and DNA arrays. The second type, based on PCR, comprises the use of MPN (most probable number)-PCR, competitive PCR or real-time PCR. In this review, these different approaches will be presented with examples of their application in environmental studies.

Concepts of the standard surface plate method and the most probable number method (MPN) were combined to provide a new enumeration technique (plate-MPN). Three discrete 0.01-ml samples of an appropriate decimal dilution were inoculated onto each quadrant of a pre-dried petri plate. The discrete spots from the inoculum were then observed for growth after incubation. Results were interpreted analogous to a 3-tube MPN test using presently available tables. Application of the test to pure cultures and mixed flora provided no evidence to indicate the plate-MPN technique to be any less accurate than the standard technique for microbial counts. The plate-MPN technique was less precise than the standard technique. However, the plate-MPN technique has many advantages over traditional methods.

SimPlate for Total Plate Count–Color Indicator (TPC-CI, IDEXX Laboratories, Inc., Westbrook, Me.) is a new medium that incorporates the redox dye resazurin to detect and quantify bacteria in food. Enumeration is achieved by the most probable number method using a SimPlate device. Viable bacteria are detected in each well of the SimPlate device by the biochemical reduction of resazurin, which is blue, to the pink resorufin or the clear dihydroresorufin indicators. Results after 24 h of incubation for TPC-CI are highly correlated with standard plate count agar after 48 h of incubation. Correlation coefficients from studies conducted at five laboratories ranged from 0.94 to 0.98 in side-by-side comparisons against standard plate count agar. Four additional test sites, using alternative methods for determining the aerobic plate count in food, reported similar results in comparison studies (r = 0.91 to 0.97). The slopes from linear regression analysis at all sites ranged from 0.91 to 0.98, with y intercepts ranging from 0.11 to 0.84. Samples used for the validation of TPC-CI included raw food products (i.e., liver and grains), which may contain natural enzymes that interfere with enzyme-based detection methods. No interference was seen from the foods tested. These results suggest that TPC-CI is a suitable alternative to existing plate count methods and has reduced incubation time.

Abstract Background: Legionnaires’ disease is a potentially lethal pneumonia contracted through inhalation of aerosolized water contaminated with Legionella bacteria. Detection and control of L. pneumophila, the primary species responsible for the disease, is critical to public health. In Québec, cooling towers and evaporative condensers are required to follow a maintenance and testing program to ensure L. pneumophila concentrations remain at acceptable levels. Objective: This study compared a new culture method based on the most probable number approach, Legiolert®, with the formal culture method used at EnvironeX for regulatory compliance testing to quantify L. pneumophila from cooling tower waters in Québec. Methods: A split-sample analysis was performed in which 401 samples from cooling towers in Québec were tested with both methods. Results: Results with 74 positive samples showed that Legiolert provided a significant increase in sensitivity for L. pneumophila compared with the agar plate method. Cooling tower samples often contain non-Legionella flora that necessitate multiple treatment and plating conditions to prevent interference with the test. Legiolert showed little to no impact from non-Legionella organisms in this study. Conclusions: Overall, Legiolert showed several advantages over the agar plate method, including increased sensitivity, reduced interference, a simplified test procedure, and an easy-to-read positive signal.

Precision parameters from four microbiological analytical methods (coliform most probable number [MPN], fecal coliform MPN, Staphylococcus aureus plate count and standard plate count) were computed for the Shellfish Quality Assurance Program of the U.S. Food and Drug Administration (FDA). The pooled reproducibility variance (SR2) for the four methods from 1973 to 1989 were 0.0778, 0.1181, 0.0137, and 0.0087, respectively.

The microbial populations of groundwaters were analyzed in a region under the influence of a landfill (piezometer L12) in the town of São Carlos, São Paulo, Brazil, and in an area not influenced by the landfill (piezometer L5). Heterotrophic bacteria were counted by spread plate method and the number of protozoa was estimated by the most probable number method. There was a larger number of organisms in well L12, with a mean value of 15.76 x 104 CFU/ml for bacteria and 9.7 MPN/ml for protozoa, whereas the mean values for piezometer L5 were 2.88 x 104 CFU/ml for bacteria and 3.4 MPN/ml for protozoa. The greater abundance detected in piezometer L12 may be related to the influence of the leachate through the landfill on the microbial populations, also demonstrated by deoxygenation and by the high conductivity values (3530 µS/cm) compared to piezometer L5 (2.47 mg/L dissolved oxygen and 42 µS/cm conductivity). The most commonly detected protozoa were amoebae and flagellates. The density of flagellate protozoa determined under microaerophilic conditions was 10 times higher than that determined under aerobic conditions.

The present study was aimed to isolate and identify micro-organisms load of drinking water of Mardan city, Khyber Pakhtunkhwa Pakistan. A total of 27 samples of drinking water were collected from different locations of the study area. Total Plate Count was determined by pour plate method, while total coliforms, total fecal coliforms and E. coli were determined by multiple tube fermentation method. Of the total collected samples, 17 (62.96%) samples were contaminated with either one or more than one type of microorganisms. The results of most probable number test showed that 13 (48.15%) samples were unsatisfactory. It was concluded that the water should be treated before consumption for drinking purpose. Regular assessment of the water quality is recommended as regular monitoring of the water quality for improvement not only prevents disease and hazards but also checks the water resources from becoming further polluted. The effects of stress and resuscitation on selective enumeration of coliforms, Escherichia coli and enterococci in mechanically deboned raw poultry meat and in dried foods were studied using a hydrophobic grid membrane filter (HGMF) technique. The effects of four different stresses, sublethal heating, freezing, acid pH and drying, were examined on 25 to 30 samples per indicator organism for each type of stress. Counts obtained with resuscitation were compared statistically to direct selective counts for each series of samples. Also, both the direct and resuscitative HGMF results were compared to a 5-tube most probable number method for coliforms and E. coli and to a spread plate method for the enterococci. The use of appropriate resuscitation procedures not only produced a significant increase in counts over the direct HGMF procedure, but also yielded HGMF results that were statistically equivalent to those obtained by conventional methods.
