Oligosaccharides have been playing an important role in biological systems. Synthesis of oligosaccharides requires the protection from hydroxyl groups present in the corresponding monosaccharide units. The existing methods of protection have drawbacks, including formation of anomeric mixtures, change in hydrophilicity or lipophilicity and solubility of the products, participation of the protecting groups in the reactions of the core of monosaccharide units, problems associated with chemoselectivity, regioselectivity and overall stereochemical outcomes of reactions. Additionally, there has been a spectral overlap of these protecting groups with carbohydrate core, which yielded more complex spectra. Therefore, the identification and synthesis of suitable alternative protecting groups have received attention in the oligosaccharide synthesis. The objective of the present study was to synthesize various fluorinated benzyl ethers of methyl-α-D-mannopyronoside and to evaluate these ethers as the alternative protecting groups for enhancing NMR resolution in the oligosaccharide synthesis. Various fluorinated benzyl ethers of methyl-α-D-mannopyronoside were prepared through the reaction of methyl-α-D-mannopyronoside with various fluorinated benzyl bromides by using Williamson ether synthesis method. Spectral analysis of these fluorinated benzyl ethers showed that the peaks of methylene carbons shifted to a value of 10-20 parts per million (ppm) to a high field region in the 13C NMR, compared to the non-fluorinated benzyl ether. As a result, the spectral complexity decreased and enhanced the spectral resolution. In this study, we concluded that fluorinated benzyl ethers could be a suitable alternative to the non-fluorinated benzyl ethers to protect the hydroxyl groups of monosaccharides in the synthesis of oligosaccharides.

Abstract Background The effects of left atrial appendage (LAA) occlusion compared to non-vitamin K antagonist oral anticoagulant (NOAC) therapy in patients with atrial fibrillation (AF) remain unknown. Aims We aimed to evaluate the outcomes in patients with AF who received LAA occlusion vs. NOAC therapy. Methods We utilised data from TriNetX which is a global federated health research network currently containing data for 88.5 million patients. ICD-10 codes were employed to identify AF patients treated with either LAA occlusion or NOAC between 1st December 2010 and 17th January 2019. Clinical outcomes of interest were analysed up to 2 years. Results 108,697 patients were included. Patients who underwent LAA occlusion were younger, more likely to be white Caucasian and male, had a greater incidence of comorbidities, and were less likely to be prescribed other cardiovascular medications. Using propensity score matching, the risk of all-cause mortality was significantly lower among patients who received LAA occlusion compared to NOAC therapy [1.51% vs. 5.60%, RR 0.27 (95% CI 0.14–0.54)], but there were no statistical differences in the composite thrombotic or thromboembolic events [8.17% vs. 7.72%, RR 1.06 (95% CI 0.73–1.53)], ischaemic stroke or TIA [4.69% vs. 5.45%, RR 0.86 (95% CI 0.54–1.38)], venous thromboembolism [1.66% vs. 1.51%, RR 1.10 (95% CI 0.47–2.57)] and intracranial haemorrhage [1.51% vs. 1.51%, RR 1.00 (95% CI 0.42–2.39)]. Conclusion Overall, LAA occlusion might be a suitable alternative to NOAC therapy for stroke prevention in patients with AF. Graphical abstract

AbstractHead-and-neck squamous cell carcinomas (HNSCCs) are relatively common in patients with Fanconi anemia (FA), a hereditary chromosomal instability disorder. Standard chemo-radiation therapy is not tolerated in FA due to an overall somatic hypersensitivity to such treatment. The question is how to find a suitable alternative treatment. We used whole-exome and whole genome mRNA sequencing to identify major genomic and transcriptomic events associated with FA-HNSCC. CRISPR-engineered FA-knockout models were used to validate a number of top hits that were likely to be druggable. We identified deletion of 18q21.2 and amplification of 11q22.2 as prevailing copy-number alterations in FA HNSCCs, the latter of which was associated with strong overexpression of the cancer-related genes YAP1, BIRC2, BIRC3 (at 11q22.1-2). We then found the drug AZD5582, a known small molecule inhibitor of BIRC2-3, to selectively kill FA tumor cells that overexpressed BIRC2-3. This occurred at drug concentrations that did not affect the viability of untransformed FA cells. Our data indicate that 11q22.2 amplifications are relatively common oncogenic events in FA-HNSCCs, as holds for non FA-HNSCC. Therefore, chemotherapeutic inhibition of overexpressed BIRC2-3 may provide the basis for an approach to develop a clinically realistic treatment of FA-HNSCCs that carry 11q22.2 amplifications.

As the global search for new methods to combat global warming and climate change continues, renewable fuels and hydrogen have emerged as saviours for environmentally polluting industries such as aviation. Sustainable aviation is the goal of the aviation industry today. There is increasing interest in achieving carbon-neutral flight to combat global warming. Hydrogen has proven to be a suitable alternative fuel. It is abundant, clean, and produces no carbon emissions, but only water after use, which has the potential to cool the environment. This paper traces the historical growth and future of the aviation and aerospace industry. It examines how hydrogen can be used in the air and on the ground to lower the aviation industry’s impact on the environment. In addition, while aircraft are an essential part of the aviation industry, other support services add to the overall impact on the environment. Hydrogen can be used to fuel the energy needs of these services. However, for hydrogen technology to be accepted and implemented, other issues such as government policy, education, and employability must be addressed. Improvement in the performance and emissions of hydrogen as an alternative energy and fuel has grown in the last decade. However, other issues such as the storage and cost and the entire value chain require significant work for hydrogen to be implemented. The international community’s alternative renewable energy and hydrogen roadmaps can provide a long-term blueprint for developing the alternative energy industry. This will inform the private and public sectors so that the industry can adjust its plan accordingly.

This study investigated the health-promoting activities of methanolic extracts of Ajwa date seed and fruit pulp extracts through in vitro studies. These studies confirmed potential antioxidant, anti-hemolytic, anti-proteolytic, and anti-bacterial activities associated with Ajwa dates. The EC50 values of fruit pulp and seed extracts in methanol were reported to be 1580.35 ± 0.37 and 1272.68 ± 0.27 µg/mL, respectively, in the DPPH test. The maximum percentage of hydrogen peroxide-reducing activity was 71.3 and 65.38% for both extracts at 600 µg/mL. Fruit pulp and seed extracts inhibited heat-induced BSA denaturation by 68.11 and 60.308%, heat-induced hemolysis by 63.84% and 58.10%, and hypersalinity-induced hemolysis by 61.71% and 57.27%, and showed the maximum anti-proteinase potential of 56.8 and 51.31% at 600 μg/mL, respectively. Seed and fruit pulp inhibited heat-induced egg albumin denaturation at the same concentration by 44.31 and 50.84%, respectively. Ajwa seed showed minimum browning intensity by 63.2%, percent aggregation index by 64.2%, and amyloid structure by 63.8% at 600 μg/mL. At 100 mg/mL, Ajwa seed extract exhibited good antibacterial activity. Molecular docking analysis showed that ten active constituents of Ajwa seeds bind with the critical antioxidant enzymes, catalase (1DGH) and superoxide dismutase (5YTU). The functional residues involved in such interactions include Arg72, Ala357, and Leu144 in 1DGH, and Gly37, Pro13, and Asp11 in 5YTU. Hence, Ajwa dates can be used to develop a suitable alternative therapy in various diseases, including diabetes and possibly COVID-19-associated complications.

Energy demand and the use of commodity consumer products, such as chemicals, plastics, and transportation fuels, are growing nowadays. These products, which are mainly derived from fossil resources and contribute to environmental pollution and CO2 emissions, will be used up eventually. Therefore, a renewable inexhaustible energy source is required. Plant biomass resources can be used as a suitable alternative source due to their green, clean attributes and low carbon emissions. Lignin is a class of complex aromatic polymers. It is highly abundant and a major constituent in the structural cell walls of all higher vascular land plants. Lignin can be used as an alternative source for fine chemicals and raw material for biofuel production. There are many chemical processes that can be potentially utilized to increase the degradation rate of lignin into biofuels or value-added chemicals. In this study, two lignin degradation methods, CuO–NaOH oxidation and tetramethyl ammonium hydroxide (TMAH) thermochemolysis, will be addressed. Both methods showed a high capacity to produce a large molecular dataset, resulting in tedious and time-consuming data analysis. To overcome this issue, an unsupervised machine learning technique called principal component analysis (PCA) is implemented.

Abstract Modeling and forecasting are among the most powerful and widely-used tools in decision support systems. The Fuzzy Linear Regression (FLR) is the most fundamental method in the fuzzy modeling area in which the uncertain relationship between the target and explanatory variables is estimated and has been frequently used in a broad range of real-world applications efficaciously. The operation logic in this method is to minimize the vagueness of the model, defined as the sum of individual spreads of the fuzzy coefficients. Although this process is coherent and can obtain the narrowest α-cut interval and exceptionally the most accurate results in the training data sets, it can not guarantee to achieve the desired level of generalization. While the quality of made managerial decisions in the modeling-based field is dependent on the generalization ability of the used method. On the other hand, the generalizability of a method is generally dependent on the precision as well as reliability of results, simultaneously. In this paper, a novel methodology is presented for the fuzzy linear regression modeling; in which in contrast to conventional methods, the constructed models' reliability is maximized instead of minimizing the vagueness. In the proposed model, fuzzy parameters are estimated in such a way that the variety of the ambiguity of the model is minimized in different data conditions. In other words, the weighted variance of different ambiguities in each validation data situation is minimized in order to estimate the unknown fuzzy parameters. To comprehensively assess the proposed method's performance, 74 benchmark datasets are regarded from the UCI. Empirical outcomes show that, in 64.86% of case studies, the proposed method has better generalizability, i.e., narrower α-cut interval as well as more accurate results in the interval and point estimation, than classic versions. It is obviously demonstrated the importance of the outcomes' reliability in addition to the precision that is not considered in the traditional FLR modeling processes. Hence, the presented EFLR method can be considered as a suitable alternative in fuzzy modeling fields, especially when more generalization is favorable.

Cholecystectomy is a procedure used to rectify severe gallbladder disease that is not amenable to medical management. It is most commonly performed in dogs with gallbladder mucocoele, a condition affecting mainly small breeds such as the Border Terrier, Cocker Spaniel and Shetland Sheepdog. Traditionally, this has been done by open laparotomy but, in the human field, where cholecystectomy is a common and routine procedure, laparoscopic cholecystectomy has almost completely superseded open laparotomy, as it has been shown to be safer and have lower rates of morbidity. This article describes a technique for laparoscopic cholecystectomy in the dog as a suitable alternative to open laparotomy.

The diversification of pedagogic tools remains essential for a fruitful learning experience among the Gen Z students by embedding technology such as gamification in learning. Recent literature has discussed the acquisition of 21st century skills and the educational challenges generally faced by Asian students due to their cultural traits. Against this background, the findings of this study open reflections relating to the benefit of gamification in acquiring 21st century skills. The objective of this chapter is to identify whether gamification is a suitable pedagogic tool among Malaysian law students in order to support the acquisition of 21st century skills. The respondents are from Year 1 and Year 2 of a three-year Bachelor of Laws degree with the majority being Malaysian students. The data collected showed that gamification helps in the acquisition of 21st century skills. Evidently, it showed that gamification can be a suitable alternative pedagogic tool to support the students to learn skills such as critical thinking, creativity, innovation, leadership, or communication.
