In RBL-2H3 rat leukemic mast cells, crosslinking IgE-receptor complexes with anti-IgE antibody leads to degranulation. Receptor crosslinking also stimulates the redistribution of receptors on the cell surface, a process that can be observed by labeling the anti-IgE with 15 nm protein A-gold particles as described in Stump et al. (1989), followed by back-scattered electron imaging (BEI) in the scanning electron microscope. We report that anti-IgE binding stimulates the redistribution of IgE-receptor complexes at 37“C from a dispersed topography (singlets and doublets; S/D) to distributions dominated sequentially by short chains, small clusters and large aggregates of crosslinked receptors. These patterns can be observed (Figure 1), quantified (Figure 2) and analyzed statistically. Cells incubated with 1 μg/ml anti-IgE, a concentration that stimulates maximum net secretion, redistribute receptors as far as chains and small clusters during a 15 min incubation period. At 3 and 10 μg/ml anti-IgE, net secretion is reduced and the majority of receptors redistribute rapidly into clusters and large aggregates.

In this symposium I will present our studies about the molecular architecture and function of the cytomatrix of the nerve cells. The nerve cell is a highly polarized cell composed of highly branched dendrites, cell body, and a single long axon along the direction of the impulse propagation. Each part of the neuron takes characteristic shapes for which the cytoskeleton provides the framework. The neuronal cytoskeletons play important roles on neuronal morphogenesis, organelle transport and the synaptic transmission. In the axon neurofilaments (NF) form dense arrays, while microtubules (MT) are arranged as small clusters among the NFs. On the other hand, MTs are distributed uniformly, whereas NFs tend to run solitarily or form small fascicles in the dendrites Quick freeze deep etch electron microscopy revealed various kinds of strands among MTs, NFs and membranous organelles (MO). These structures form major elements of the cytomatrix in the neuron. To investigate molecular nature and function of these filaments first we studied molecular structures of microtubule associated proteins (MAP1A, MAP1B, MAP2, MAP2C and tau), and microtubules reconstituted from MAPs and tubulin in vitro. These MAPs were all fibrous molecules with different length and formed arm like projections from the microtubule surface.

<div> <div> <div> <p>Many-body potential energy functions (PEFs) based on the TTM-nrg and MB-nrg theoretical/computational frameworks are developed from coupled cluster reference data for neat methane and mixed methane/water systems. It is shown that that the MB-nrg PEFs achieve subchemical accuracy in the representation of individual many-body effects in small clusters and enables predictive simulations from the gas to the liquid phase. Analysis of structural properties calculated from molecular dynamics simulations of liquid methane and methane/water mixtures using both TTM-nrg and MB-nrg PEFs indicates that, while accounting for polarization effects is important for a correct description of many-body interactions in the liquid phase, an accurate representation of short-range interactions, as provided by the MB-nrg PEFs, is necessary for a quantitative description of the local solvation structure in liquid mixtures. </p> </div> </div> </div>

Abstract Fragmentation of granular clusters may be studied by experiments and by granular mechanics simulation. When comparing results, it is often assumed that results can be compared when scaled to the same value of $$E/E_{\mathrm{sep}}$$ E / E sep , where E denotes the collision energy and $$E_{\mathrm{sep}}$$ E sep is the energy needed to break every contact in the granular clusters. The ratio $$E/E_{\mathrm{sep}}\propto v^2$$ E / E sep ∝ v 2 depends on the collision velocity v but not on the number of grains per cluster, N. We test this hypothesis using granular-mechanics simulations on silica clusters containing a few thousand grains in the velocity range where fragmentation starts. We find that a good parameter to compare different systems is given by $$E/(N^{\alpha }E_{\mathrm{sep}})$$ E / ( N α E sep ) , where $$\alpha \sim 2/3$$ α ∼ 2 / 3 . The occurrence of the extra factor $$N^{\alpha }$$ N α is caused by energy dissipation during the collision such that large clusters request a higher impact energy for reaching the same level of fragmentation than small clusters. Energy is dissipated during the collision mainly by normal and tangential (sliding) forces between grains. For large values of the viscoelastic friction parameter, we find smaller cluster fragmentation, since fragment velocities are smaller and allow for fragment recombination. Graphic abstract

The relationship between the cluster morphology formed during natural or artificial aging and the paint-bake hardening response in an Al-0.62Mg-0.93Si (mass%) alloy have been investigated using atom probe tomography (APT). Increasing the subsequent aging time at 170 °C causes a gradual increase in hardness in the artificially aged materials, while the retardation period of the hardness increase appears in the naturally aged materials at the early stage of aging. The statistically-proved records in the APT analysis have shown that the artificially aged materials have some large clusters. It is revealed that the hardening at the early stage of the subsequent aging at 170 °C is not promoted in the long-time naturally aged material although the number density of small clusters increases approximately 1.3 times by prolonged natural aging.Hence, we believe that the small clusters are hard to transform continuously into the β'' phase during aging at 170 °C. As for the naturally aged materials, the long-time aging leads to a significant drop in hardness at the early stage of aging at 170 °C. It is speculated that the Mg-Si mixed clusters formed after long-time natural aging can be reversed during the subsequent heat treatment.

In Europe, foodborne transmission has been clearly associated to sporadic cases and small clusters of hepatitis E in humans linked to the consumption of contaminated pig liver sausages, raw venison, or undercooked wild boar meat. In Europe, zoonotic HEV-genotype 3 strains are widespread in pig farms but little information is available on the prevalence of HEV positive pigs at slaughterhouse. In the present study, the prevalence of HEV-RNA positive pigs was assessed on 585 animals from 4 abattoirs located across Italy. Twenty-one pigs (3.6%) tested positive for HEV in either feces or liver by real-time RT-PCR. In these 21 pigs, eight diaphragm muscles resulted positive for HEV-RNA. Among animals collected in one abattoir, 4 out of 91 plasma tested positive for HEV-RNA. ELISA tests for the detection of total antibodies against HEV showed a high seroprevalence (76.8%), confirming the frequent exposure of pigs to the virus. The phylogenetic analyses conducted on sequences of both ORF1 and ORF2 fragments, shows the circulation of HEV-3c and of a novel unclassified subtype. This study provides information on HEV occurrence in pigs at the slaughterhouse, confirming that muscles are rarely contaminated by HEV-RNA compared to liver, which is the most frequently positive for HEV.
