Survival of Escherichia coli O157:H7 and Campylobacter jejuni that were separately inoculated into bottled purified drinking water was investigated during storage at 22, 4, and −18°C for 5, 7, and 2 days, respectively. Two inoculation levels were used, 1 and 10 CFU/ml (102 and 103 CFU/100 ml). In samples inoculated with 102 CFU/100 ml, C. jejuni was not detectable (>2-log reduction) after storage under the conditions specified above. E. coli O157:H7 was detected on nonselective and selective media at log reductions of 1.08 to 1.25 after storage at 22°C, 1.19 to 1.56 after storage at 4°C, and 1.54 to 1.98 after storage at −18°C. When the higher inoculation level of 103 CFU/100 ml was used, C. jejuni was able to survive at 22 and 4°C, with 2.25- and 2.17-log reductions, respectively, observed on nonselective media. At these higher inoculation levels, E. coli O157:H7 was detectable at 22, 4, and −18°C, with log reductions of 0.76, 0.97, and 1.21, respectively, achieved on nonselective media. Additionally, E. coli O157:H7 showed significant differences in culturability (P < 0.05) on the nonselective and selective culture media under the different storage conditions, with storage at −18°C for 2 days being the treatment most inhibiting. The percentage of sublethal injury of E. coli O157:H7 ranged from ~33 to 75%, indicating that microbial examination of bottled water must be done carefully, otherwise false-negative results or underestimation of bacterial numbers could pose a health risk when low levels of pathogens are present.

Information on lairages (regarding design, construction materials, and use of bedding and cleaning regimes) was collected for 21 commercial cattle and/or sheep abattoirs in southwest England. Overall, roughened or grooved concrete was the most common lairage flooring material. Straw bedding was used in the majority of lairages and was changed between animal batches, daily, weekly, and monthly in roughly 5, 60, 15, and 10%, respectively, of the surveyed lairages. Lairages were commonly washed with cold water with no detergents and/or disinfectants, and only about half the lairages were washed daily. Also, a three-pathogen cocktail inoculum comprising Escherichia coli O157 (NCTC 12900), Salmonella Kedougou (VLA S488/01), and Campylobacter jejuni (VLA C4) (at 8, 8, and 7 log CFU/ml or 8, 8, and 7 log CFU/g, respectively) was suspended in either broth (for nonfecal contamination) or bovine feces (for fecal contamination). Samples of the four most common substrates present in lairages (concrete, straw, metal, and hide) were contaminated in vitro with either fecal or nonfecal inocula and subsequently held in the laboratory at 10 or 25°C for 1 week. Bacterial counts for these samples were monitored daily and used to assess the number of days required for a 90% reduction of each pathogen population. In most cases, pathogens survived for >1 week, with survival rates being higher for straw or hide than for concrete or metal and higher for fecal contamination than for nonfecal contamination. Overall, if survival rates for the three pathogens under practical lairage conditions were similar to the in vitro survival rates found in this study, contamination of lairages with pathogens could be carried over from one batch of animals to another and/or from one day to the next.

Ngộ độc thực phẩm, với một trong những nguyên nhân chính do nhiễm khuẩn vẫn luôn là mối lo ngại, mang tính toàn cầu, được Tổ chức Sức khỏe Thế giới rất quan tâm. Việc xác định chính xác đối tượng vi khuẩn nhiễm vẫn luôn là một nhu cầu cấp thiết của các labo lâm sàng. Nghiên cứu trước đây của chúng tôi đã được công bố với việc thành công bước đầu trong việc xây dựng một quy trình dựa trên kĩ thuật PCR-Reverse Dot Blot (PCR-RDB) nhằm xác định đồng thời 12 vi khuẩn gây bệnh đường ruột ch yếu, bao gồm Bacillus cereus, Clostridium botulinum, Clostridium perfringen, Staphylococcus aureus, Listeria monocytogenes, Escherichia coli O157:H7, Salmonella spp., Shigella spp.., Vibrio cholerae, Vibrio parahaemolyticus, Yersinia enterocolitica và Brucella spp. Chúng tôi tiếp tục phát triển nghiên cứu này nhằm hoàn thiện quy trình PCR-RDB bằng việc thiết kế bổ sung trên màng các loại mẫu dò nhằm làm chứng dương, chứng âm, chứng màu và chứng kiểm tra tín hiệu nền. Bên cạnh đó, xét nhu cầu lâm sàng, việc bổ sung hai loại mẫu dò đề dò hai vi khuẩn Campylobacter jejuni và Yersinia enterocolitica O:8 gây bệnh đường ruột chủ yếu ở trẻ em, là cần thiết. Quy trình PCR-RDB được hoàn thiện trong nghiên cứu này vì vậy có khả năng phát hiện đồng thời 14 vi khuẩn gây bệnh đường ruột, một cách đặc hiệu, với độ nhạy đạt 10^2 bản sao/ml, đã được thử nghiệm trên 30 mẫu phân, ghi nhận kết quả hoàn toàn khớp với kít thương mại PowerCheckTM 20 Pathogen Multiplex Real-time PCR Kit (Korea).

We report on two children with Escherichia coli O157 infection, one of whom developed haemolytic uraemic syndrome (HUS). Both had drunk raw cows’ or goats’ milk in the week before their illness. Molecular subtyping identified a sorbitol fermenting Escherichia coli O157:H isolate from a dairy cow. This isolate differed from Shiga toxin producing O157:H strains isolated from the 6 year old boy with HUS. This result underlines the need to search for other causes of infection, despite documented consumption of unpasteurised milk. In the second patient, human sorbitol non-fermenting O157:H isolates and animal isolates from goats were indistinguishable. The isolation of indistinguishable sorbitol non-fermenting Escherichia coli O157:H from contact animals supports the association between HUS and consumption of raw goats’ milk, and re-emphasises the importance of pasteurising milk.
