Taxonomic status and origin of some ecological forms of whitefish Coregonus lavaretus (L.) from water bodies of Siberia

The article deals with carrying out DNA barcoding of aquatic invertebrates of Kazakhstan to identify their taxonomic status as organic pollution indicators. 33 species of the Balkhash-Alakol basin and the Zhayik river were analyzed. 21 species correlate (95-100%) with previously published sequences of invertebrates with well-known classifications in the GenBank and BOLD databases. The taxonomic discrepancy in morphometric and genetic parameters in certain species has been revealed. The discrepancy may be caused by the morphological identity in chironomids at a larval stage. The phylogenetic trees of the investigated species within the families Chironomidae and Moinidae have been indicated. Chironomids are represented by ten clades of different types of genetic polymorphism of DNA gene. Genetic links of Moinidae are detected in four groups including a cryptic species from Lake Alakol. It has been stated that in distribution of cryptic taxons in Moina family factors of salinity and depth of the lake are important, as well as differences in depth. Molecular DNA-barcoding of invertebrates of Kazakhstan should be continued with covering a greater number of species and several replications, with qualified primary fixation of subjects of research and a sufficient number of samples. Authenticity of composition defining, species abundance, species characteristics of aquatic invertebrates from the water bodies of poorly explored arid regions is necessary for using them as indicators of the ecological status of water bodies.

The study reveals the results of the analysis of fatty acid composition of muscle, liver and gonad tissues of the whitefish Coregonus lavaretus L. of different ages from lakes Lovozero and Umbozero of the Kola Peninsula. The analysis of fatty acid composition in organs and tissues allows to evaluate the physiological characteristics of the organism. It was found that the content of monoenoic fatty acids in the muscles of the whitefish from lake Lovozero is higher than that of the whitefish from Umbozero. It is probably due to the features of the food base of the objects studied. In the muscles of immature whitefish of lake Umbozero the content of polyenic fatty acids is higher compared to the fish from Lovozero. The content of PUFA significantly increases in the mature whitefish from Umbozero lake perhaps due to the temperature regime in water bodies and different rates of motor activity of fish. Lower levels of PUFA in the liver of the whitefish from Lovozero lake may indicate reduced metabolism in hepatocytes. In the gonads of female whitefish from the studied water bodies, the content of saturated fatty acids is low compared to the polyene fatty acids which is characteristic for the viscosity reduction in the membranes of oocytes.

Background. A population genetic study of Siberian whitefish or pidschian Coregonus lavaretus pidschian Gmelin of the Anabar River (North-West Yakutia) has been done to study genetic structure of the species within a river system and for revealing its phylogeographic relations. Materials and Methods. Five samples of pidschian from different parts of the river were analyzed for 30 isozyme loci. Results and Discussion. The results has demonstrated a significant genetic heterogeneity of the species sampled from lower and middle reaches of the river (0.001 < DN< 0.056). The most likely reason for genetic differentiation of the species within the river system is a colonization of the watershed by independent phylogenetic lineages of whitefish. At present the phylogenetic lineages in Anabar River are presented by sympatric ecological forms of the species – eastern-siberian whitefish and glacial-plains whitefish – which maintain their reproductive isolation. The relationships of ecological forms of whitefish dwelling the Anabar River with other populations of the species in Siberia, and possible ways of penetration of discrete phylogenetic lineages of pidschian in the basin of studied river are discussed. Conclusions. A genetic heterogeneity of whitefish from Anabar River should be taken into account while creating adequate intraspecific taxonomy and considered in conducting fisheries and environmental measures.

Background. The purpose of this work was studying of interaction between morphologically similar forms of the pidcshian-like whitefishes inhabiting restricted territory of “riverine-lacustrine” system situated in the Bolshoi Abakan Rriver basin. Material and methods. In the morphological analysis were used 141 whitefish individuals from Lake Karakul and 26 - from the Bolshoi Abakan River. In the genetic analysis 11 and 26 whitefish individuals were used respectively. For them the sequences of a gene 16S-ND1 of mitochondrial DNA (mtDNA) and ITS1 of nuclear DNA (nDNA) were defined. Results. The whitefish populations clear differ on the gill raker numbers on the first branchial arch, growth. The analysis of the 16S-ND1 mtDNA genes variability revealed among them several divergent lineages and two lineages were revealed on the structure of ITS1 fragment of the nDNA. Conclusions. Based on the obtained data it is proved that two genetically divergent populations of the morphologically same forms of pidcshian-like whitefishes inhabiting the “riverine-lacustrine” system (the Bolshoi Abakan River - Lake Karakul). Our results are supported a hypothesis of multiple hybridization between week differentiated populations/form of pidcshian-like whitefishes inhabiting region during the postglacial period.

In Lake Onega, the whitefish Coregonus lavaretus has been shown to occur as a variety of forms. Medium- and sparsely-ranked whitefish are most abundant. Analysis of available data indicates that whitefish populations from Karelia’s large lakes display the maximum values of various genetic variability indices. This fact seems to be due to the history of the colonization of the lake by the discrete evolutionary whitefish lineages from various Late Quaternary habitats followed by their hybridization. A great variety of Onega whitefish haplotypes is probably related to the genetic heterogeneity of the whitefish who until recently had occurred as five ecological forms ranking as subspecies. The median network obtained suggests that many of the populations studied have become less abundant. The well-defined “star-like” network structure is characteristic of populations that passed through a narrow “bottleneck” in the near past and then expanded rapidly, as indicated by the abundance of rare haplotype varieties. It seems that the retreat of the Scandinavian glacier was not a momentary event but took a long time during which the populations formed were subjected to demographic transformations.
