Cytology of triploid and pentaploid hybrids of Elymus trachycaulus × Pseudoroegneria spicata and P. spicata ssp. inermis

Emasculated clones of tetraploid and octaploid Elymus trachycaulus crossed with Pseudoroegneria spicata yielded 69 triploids and 13 pentaploid hybrids. The triploid hybrids were morphologically intermediate between the parental species but the pentaploid hybrids resembled E. trachycaulus more closely than P. spicata. A very high degree of plant size variability (very weak to very vigorous) was observed among the 50 triploids that survived to maturity. The variability among the pentaploids, though apparent, was not high. Mean chromosome associations among the selected eight triploids varied from low multivalent formation at meiosis (7.60 I + 6.18 II + 0.24 III + 0.08 IV) to high multivalent-forming meiosis (7.28 I + 5.66 II + 0.32 III + 0.36 IV). The bivalent configurations in these triploids were attributed to homoeologous pairing between the S1 genome of P. spicata and S genomes of E. trachycaulus and the multivalents indicated intergenomal and intragenomal pairings. Mean chromosome associations of 5.22 I + 11.94 II + 1.97 III in 78.7% and 7 I + 14 II in 21.3% of the 150 cells were distributed among the three pentaploid hybrids. The low frequency of trivalents and the absence of multivalents higher than trivalent configurations suggest that homoeologous pairing was substantially reduced and there was no intergenomal and intragenomal pairing in these 2n = 35 x hybrids. Fourteen triploids and three pentaploid hybrids were produced from 4x and 8x forms of E. trachycaulus and P. spicata spp. inermis crosses. The triploids were intermediate between the parents but the pentaploids were more similar to E. trachycaulus. Mean chromosome associations in the triploids (6.020 I + 5.315 II + 0.554 III + 0.235 IV + 0.250 V + 0.048 VI + 0.03 VII) and in the pentaploids (3.50 I + 10.18 II + 3.54 III + 0.13 IV) indicated a very high degree of intergenomal and intragenomal pairing in the triploids; nevertheless, it was greatly reduced in the pentaploids. The Pseudoroegneria spicata ssp. inermis genome may have gene(s) that affect a high degree of intergenomal and intragenomal pairing in triploid hybrids with E. trachycaulus. However, in pentaploid hybrids (SS HH S2) the S and H genomes of E. trachycaulus in double dose seem to have restored its regular meiotic bahaviour, at least partially, and reduced homoeologous pairing as well as intergenomal and intragenomal pairing. The analysis of CTG and CTA expression in malignant tumors described in this review has been showed that different types of tumors are significantly different from each other according to the frequency of CTA mRNA expression. Melanoma, ovarian cancer and lung cancer have a very high frequency of CTA expression. Lymphoma, kidney cancer, pancreatic cancer have a low frequency of CTA expression. Breast cancer, bladder cancer, prostate cancer demonstrate an intermediate level of CTA expression. High degree malignant tumors in late clinical stage with metastases showed a greater incidence of CTA -gene expression. CTA-genes are expressed together in tumor. If the tumor is positive for one CTA-gene then the expression of several genes is possible. Immunogenic CTA-s are a well object for anti-tumor vaccines creating.

The visualization of organic macromolecules such as proteins, nucleic acids, viruses and virus components has reached its high degree of effectiveness owing to refinements and reliability of instruments and to the invention of methods for enhancing the structure of these materials within the electron image. The latter techniques have been most important because what can be seen depends upon the molecular and atomic character of the object as modified which is rarely evident in the pristine material. Structure may thus be displayed by the arts of positive and negative staining, shadow casting, replication and other techniques. Enhancement of contrast, which delineates bounds of isolated macromolecules has been effected progressively over the years as illustrated in Figs. 1, 2, 3 and 4 by these methods. We now look to the future wondering what other visions are waiting to be seen. The instrument designers will need to exact from the arts of fabrication the performance that theory has prescribed as well as methods for phase and interference contrast with explorations of the potentialities of very high and very low voltages. Chemistry must play an increasingly important part in future progress by providing specific stain molecules of high visibility, substrates of vanishing “noise” level and means for preservation of molecular structures that usually exist in a solvated condition.

Abstract We surveyed sequence variation and divergence for the entire 5972-bp transcriptional unit of the white gene in 15 lines of Drosophila melanogaster and one line of D. simulans. We found a very high degree of haplotypic structuring for the polymorphisms in the 3′ half of the gene, as opposed to the polymorphisms in the 5′ half. To determine the evolutionary mechanisms responsible for this pattern, we sequenced a 1612-bp segment of the white gene from an additional 33 lines of D. melanogaster from a European and a North American population. This 1612-bp segment encompasses an 834bp region of the white gene in which the polymorphisms form high frequency haplotypes that cannot be explained by a neutral equilibrium model of molecular evolution. The small number of recombinants in the 834bp region suggests epistatic selection as the cause of the haplotypic structuring, while an investigation of nucleotide diversity supports a directional selection hypothesis. A multi-locus selection model that combines features from both-hypotheses and takes the recent history of D. melanogaster into account may be the best explanation for these data.

The uptake of tritiated thymine into cells of a heterogeneous population of Prorocentrum micans was investigated using light-microscope and electron-microscope autoradiography. Specificity of thymine uptake into DNA was demonstrated by the specific removal of label from wax-embedded material using DNase and by the high degree of localization of nuclear label to chromosomes in the electron-microscope autoradiographs. All nuclei, including both dividing and non-dividing cells, showed a substantial uptake of label, indicating that nuclear DNA synthesis in Prorocentrum micans is a continuous process. The level of DNA synthesis does show considerable variation, however, with very high levels in some interphase nuclei. The continuous replication of nuclear DNA provides further evidence of dinoflagellate affinity to the prokaryotes, and indicates that Prorocentrum micans is a very primitive eukaryote cell.

This study presents a decision-support method to estimate the next year performance of corporate Operating Income Margin (OIM). It is based on a unique combination of cross-section model and the rules-based evaluation mechanism. The estimate is done in terms of broad categories, and not precise numerical values. The model is constructed as follows: its dependent variable (OIM) is one year ahead vs. the corresponding explanatory variables. This structure of the model allows us to view explanatory variables as reflecting financial potential of corporations. The evaluation component consists of a set of rules designed to identify the companies whose “potential” clearly points to an opportunity to invest. For the method presented here to succeed, it is necessary to utilize a highly reliable modeling method, even if it is “Fuzzy”. We apply Soft Regression (SR), which is a Soft Computing modeling tool based on Fuzzy Logic, and utilize all available proxy variables by creating intervals of values. Advantages of utilizing SR, and the intervals’-based modeling are extensively discussed. Modeling results for five consecutive years are consistent and stable, thus indicating high degree of reliability. Testing indicates very high success rate for the stock market related domain, the lowest being 87.9%.

Context.—Myeloid sarcoma of the head and neck region can pose diagnostic challenges because of the low frequency of myeloid sarcoma and the potential for tumors of almost any lineage to occur in the head and neck. Objective.—To study the clinicopathologic and immunohistochemical characteristics of myeloid sarcoma in the head and neck region and to review the differential diagnosis. Design.—We searched for cases of myeloid sarcoma involving the head and neck region for a 24-year period at our institution. The medical records and pathology slides were reviewed. Additional immunohistochemical stains were performed. Results.—We identified 17 patients, age 17 to 85 years. Most tumors involved the oral cavity. Myeloid sarcoma was the initial diagnosis in 9 patients (53%); the remaining 8 patients (47%) had a history of bone marrow disease. Immunohistochemical analysis using antibodies specific for lysozyme, CD43, and CD68 were highly sensitive for diagnosis but were not specific. By contrast, assessment for myeloperoxidase in this study was less sensitive but more specific. We also used antibodies specific for CD11c and CD33 in a subset of cases, and these reagents seem helpful as well. Conclusions.—The clinical presentation of myeloid sarcoma involving the head and neck, particularly the mouth, is often nonspecific, and a high degree of suspicion for the possibility of myeloid sarcoma is needed. Immunohistochemistry is very helpful for establishing the diagnosis.

AbstractHESS J1731−347 a.k.a. SNR G353.6−0.7 is one of the five known very high energy (VHE, Energy > 0.1 TeV) shell-type supernova remnants. We carried out Giant Metrewave Radio Telescope (GMRT) observations of this TeV SNR in 1390, 610 and 325 MHz bands. We detected the 325 and 610 MHz radio counterparts of the SNR G353.6−0.7 (Nayana et al. 2017). We also determined the spectral indices of individual filaments and our values are consistent with the non-thermal radio emission. We compared the radio morphology with that of VHE emission. The peak in radio emission corresponds to the faintest feature in the VHE emission. We explain this anti-correlated emission in a possible leptonic origin of the VHE γ-rays.

Low frequency internal friction of Ti49Ni51 binary and Ti50Ni40Cu10 ternary shape memory alloys has been measured. The effect of solution and aging heat treatments on the damping property was examined. The temperature spectrum of internal friction for TiNi binary alloy consists, in general, of two peaks; one is a transition peak which is associated with the parent-martensite transformation and is rather unstable in a sense that it strongly depends on the frequency and decreases considerably when held at a constant temperature. The other one is a very high peak of the order of 10-2, which appears at around 200K. It appears both on cooling and on heating with no temperature hysteresis, and is very stable. The behavior of the peak is strongly influenced by the heat treatments. The trial of two-stage aging with a purpose of improving the damping capacity has been proved unsatisfactory. TiNiCu has a very high damping, the highest internal friction reaching 0.2, but by quenching from very high temperature, say 1373K, the damping is remarkably lowered. For the realization of high damping the quenching from a certain temperature range around 1173K seems the most preferable condition.

The incorporation of tritiated thymidine in Amoeba proteus was reinvestigated in order to see if it could be associated with microscopically detectable structures. Staining experiments with basic dyes, including the fluorochrome acridine orange, revealed the presence of large numbers of 0.3 to 0.5 µ particles in the cytoplasm of all cells studied. The effect of nuclease digestion on the dye affinity of the particles suggests that they contain DNA as well as RNA. Centrifugation of living cells at 10,000 g leads to the sedimentation of the particles in the centrifugal third of the ameba near the nucleus. Analysis of centrifuged cells which had been incubated with H3-thymidine showed a very high degree of correlation between the location of the nucleic acid-containing granules and that of acid-insoluble, deoxyribonuclease-sensitive labeled molecules and leads to the conclusion that cytoplasmic DNA synthesis in Amoeba proteus occurs in association with these particles.
