Working memory (WM) is one of the most investigated cognitive functions albeit the extent to which individual characteristics impact on performance is still unclear, especially when older adults are involved. The present study considers repeated practice of a visual N -Back task with three difficulty levels (1-, 2-, and 3-Back) in healthy young and older individuals. Our results reveal that, for both age groups, the expected mental fatigue was countered by a learning effect, in terms of accuracies and reaction times, which turned out to benefit females more than males, for all three N -Back levels. We conclude that future WM studies, in particular when relying on repeated N -Back sessions, should account for learning effects in relation to mental fatigue and gender, in both young and older adults.

Background: Early prognosis of high-risk older adults for amnestic mild cognitive impairment (aMCI), using noninvasive and sensitive neuromarkers, is key for early prevention of Alzheimer’s disease. We have developed individualized measures in electrophysiological brain signals during working memory that distinguish patients with aMCI from age-matched cognitively intact older individuals. Objective: Here we test longitudinally the prognosis of the baseline neuromarkers for aMCI risk. We hypothesized that the older individuals diagnosed with incident aMCI already have aMCI-like brain signatures years before diagnosis. Methods: Electroencephalogram (EEG) and memory performance were recorded during a working memory task at baseline. The individualized baseline neuromarkers, annual cognitive status, and longitudinal changes in memory recall scores up to 10 years were analyzed. Results: Seven of the 19 cognitively normal older adults were diagnosed with incident aMCI for a median 5.2 years later. The seven converters’ frontal brainwaves were statistically identical to those patients with diagnosed aMCI (n = 14) at baseline. Importantly, the converters’ baseline memory-related brainwaves (reduced mean frontal responses to memory targets) were significantly different from those who remained normal. Furthermore, differentiation pattern of left frontal memory-related responses (targets versus nontargets) was associated with an increased risk hazard of aMCI (HR = 1.47, 95% CI 1.03, 2.08). Conclusion: The memory-related neuromarkers detect MCI-like brain signatures about five years before diagnosis. The individualized frontal neuromarkers index increased MCI risk at baseline. These noninvasive neuromarkers during our Bluegrass memory task have great potential to be used repeatedly for individualized prognosis of MCI risk and progression before clinical diagnosis.

Studies examining the effect of embodied cognition have shown that linking one’s body movements to a cognitive task can enhance performance. The current study investigated whether concurrent walking while encoding or recalling spatial information improves working memory performance, and whether 10-year-old children, young adults, or older adults (Mage = 72 years) are affected differently by embodiment. The goal of the Spatial Memory Task was to encode and recall sequences of increasing length by reproducing positions of target fields in the correct order. The nine targets were positioned in a random configuration on a large square carpet (2.5 m × 2.5 m). During encoding and recall, participants either did not move, or they walked into the target fields. In a within-subjects design, all possible combinations of encoding and recall conditions were tested in counterbalanced order. Contrary to our predictions, moving particularly impaired encoding, but also recall. These negative effects were present in all age groups, but older adults’ memory was hampered even more strongly by walking during encoding and recall. Our results indicate that embodiment may not help people to memorize spatial information, but can create a dual-task situation instead.

The promise of transcranial direct-current stimulation (tDCS) as a modulator of cognition has appealed to researchers, media, and the general public. Researchers have suggested that tDCS may increase effects of cognitive training. In this study of 123 older adults, we examined the interactive effects of 20 sessions of anodal tDCS over the left prefrontal cortex (vs. sham tDCS) and simultaneous working memory training (vs. control training) on change in cognitive abilities. Stimulation did not modulate gains from pre- to posttest on latent factors of either trained or untrained tasks in a statistically significant manner. A supporting meta-analysis ( n = 266), including younger as well as older individuals, showed that, when combined with training, tDCS was not much more effective than sham tDCS at changing working memory performance ( g = 0.07, 95% confidence interval, or CI = [−0.21, 0.34]) and global cognition performance ( g = −0.01, 95% CI = [−0.29, 0.26]) assessed in the absence of stimulation. These results question the general usefulness of current tDCS protocols for enhancing the effects of cognitive training on cognitive ability.

Both selective attention and visual working memory (WM) performance are vulnerable to age related decline. Older adults perform worse on, and are less able to modulate oscillatory power in the alpha frequency range (8-12 Hz) than younger adults in WM tasks involving predictive cues about ‘where’ or ‘when’ a stimulus will be present. However, no study has investigated whether alpha power is modulated by cues predicting ‘how long’ an encoding duration will be. To test this, we recorded electroencephalography (EEG) while 24 younger (aged 18-33 years) and 23 older (aged 60-77 years) adults completed a modified delay match-to-sample task where participants were cued to the duration (either 0.1 s or 0.5 s) of an encoding stimulus consisting of 4 coloured squares. We found: (1) predictive cues increased WM capacity, but long encoding duration trials led to reduced WM capacity in both age groups, compared to short encoding duration trials; (2) no evidence for differences in preparatory alpha power between predictive and neutral cues for either short or long encoding durations, but preparatory alpha suppression was weaker in older adults; (3) retention period oscillatory power differed between short and long encoding duration trials, but these differences were no longer present when comparing the trial types from the onset of the encoding stimulus; and (4) oscillatory power in the preparatory and retention periods were not related to task performance. Our results suggest that preparatory alpha power is not modulated by predictive cues towards encoding duration during visual WM, however, reductions in alpha/beta oscillatory power during visual WM retention may be linked to the encoding stimulus, rather than a process specific to WM retention.

Background: Older adults perform better in tasks which include positive stimuli, referred to as the positivity effect. However, recent research suggests that the positivity effect could be attenuated when additional challenges such as stress or cognitive demands are introduced. Moreover, it is well established that older adults are relatively resilient to many of the adverse effects of sleep deprivation. Our aim was to investigate if the positivity effect in older adults is affected by one night of total sleep deprivation using an emotional working memory task. Methods: A healthy sample of 48 older adults (60-72 years) was either sleep deprived for one night (n = 24) or had a normal night’s sleep (n = 24). They performed an emotional working memory n-back (n = 1 & 3) task containing positive, negative and neutral pictures. Results: Performance in terms of accuracy and reaction times was best for positive stimuli and worst for negative stimuli. This positivity effect was not altered by sleep deprivation. Results also showed that, despite significantly increased sleepiness, there was no effect of sleep deprivation on working memory performance. A working memory load × valence interaction on the reaction times revealed that the beneficial effect of positive stimuli was only present in the 1-back condition. Conclusion: While the positivity effect and general working memory abilities in older adults are intact after one night of sleep deprivation, increased cognitive demand attenuates the positivity effect on working memory speed.

AbstractWorking memory (WM) is vulnerable to age-related decline, particularly under high loads. Visual alpha oscillations contribute to WM performance in younger adults, and although alpha decreases in power and frequency with age, it is unclear if alpha activity supports WM in older adults. We recorded electroencephalography (EEG) while 24 younger (aged 18-35 years) and 30 older (aged 50-86) adults performed a modified Sternberg task with varying load conditions. Older adults demonstrated slower reaction times at all loads, but there were no significant age differences in accuracy. Regardless of age, alpha power decreased, and alpha frequency increased with load during encoding, and the magnitude of alpha suppression during retention was larger at higher loads. While alpha power during retention was lower than fixation in older, but not younger adults, the relative change from fixation was not significantly different between age groups. Individual differences in alpha power did not predict performance for either age groups or at any WM loads. Future research should elaborate the functional significance of alpha power and frequency changes that accompany WM performance in cognitive ageing.

Normal adult aging is known to be associated with lower performance on tasks assessing the short-term storage of information. However, whether or not there are additional age-related deficits associated with concurrent storage and processing demands within working memory remains unclear. Methodological differences across studies are considered critical factors responsible for the variability in the magnitude of the reported age effects. Here we synthesized comparisons of younger and older adults' performance on tasks measuring storage alone against those combining storage with concurrent processing of information. We also considered the influence of task-related moderator variables. Meta-analysis of effect sizes revealed a small but disproportionate effect of processing on older adults' memory performance. Moderator analysis indicated that equating single task storage performance across age groups (titration) and the nature of the stimulus material were important determinants of memory accuracy. Titration of storage task difficulty was found to lead to smaller, and non-significant, age-differences in dual task costs. These results were corroborated by supplementary Brinley and state-trace analyses. We discuss these findings in relation to the extant literature and current working memory theory as well as possibilities for future research to address the residual heterogeneity in effect sizes.

Abstract Objectives In young adults, the ability to verbally recall instructions in working memory is enhanced if the sequences are physically enacted by the participant (self-enactment) or the experimenter (demonstration) during encoding. Here we examine the effects of self-enactment and demonstration at encoding on working memory performance in older and younger adults. Method Fifty young (18–23 years) and 40 older (60–89 years) adults listened to sequences of novel action-object pairs before verbally recalling them in the correct order. There were three different encoding conditions: spoken only, spoken + demonstration, and spoken + self-enactment. We included two different levels of difficulty to investigate whether task complexity moderated the effect of encoding condition and whether this differed between age groups. Results Relative to the spoken only condition, demonstration significantly improved young and older adults’ serial recall performance, but self-enactment only enhanced performance in the young adults, and this boost was smaller than the one gained through demonstration. Discussion Our findings suggest that additional spatial-motoric information is beneficial for older adults when the actions are demonstrated to them, but not when the individual must enact the instructions themselves.

AbstractAlterations of resting-state EEG microstates have been associated with various neurological disorders and behavioral states. Interestingly, age-related differences in EEG microstate organization have also been reported, and it has been suggested that resting-state EEG activity may predict cognitive capacities in healthy individuals across the lifespan. In this exploratory study, we performed a microstate analysis of resting-state brain activity and tested allocentric spatial working memory performance in healthy adult individuals: twenty 25–30-year-olds and twenty-five 64–75-year-olds. We found a lower spatial working memory performance in older adults, as well as age-related differences in the five EEG microstate maps A, B, C, C′ and D, but especially in microstate maps C and C′. These two maps have been linked to neuronal activity in the frontal and parietal brain regions which are associated with working memory and attention, cognitive functions that have been shown to be sensitive to aging. Older adults exhibited lower global explained variance and occurrence of maps C and C′. Moreover, although there was a higher probability to transition from any map towards maps C, C′ and D in young and older adults, this probability was lower in older adults. Finally, although age-related differences in resting-state EEG microstates paralleled differences in allocentric spatial working memory performance, we found no evidence that any individual or combination of resting-state EEG microstate parameter(s) could reliably predict individual spatial working memory performance. Whether the temporal dynamics of EEG microstates may be used to assess healthy cognitive aging from resting-state brain activity requires further investigation.
