Aims: This study was designed to evaluate the phytochemical constituents and lipid peroxidation activities of extracts of two species of groundnut peels (Var-Hypogaea-Big and Var-vulgaris-Small). Study Design: Twenty-four (24) male wistar albino rats where randomized into four (4) groups of six (6) rats each. Group A was the baseline, Groups B and C were given feed formulated with 1% groundnut peel extract of Var-hypogaea and Var-vulgaris respectively, group D is the control group fed with a standard feed. Results: The phytochemical analysis was carried out using standard methods. Lipid peroxidation was determined by the thiobarbituric acid-reacting substances (TBARS) assay method. The phytochemical analysis revealed that the extract from the groundnut peels of Var-hypogaea-big contain alkaloids, tannins, saponnins, terpenoid, flavonoids phlobotannin, and glycosides. Also, the groundnut peel extracts of Var-vulgaris-small reveal the presence of alkaloids, tannins, saponnins, terpenoid, flavonoids phlobotannin and glycosides. There was a significant (p<0.05) increase in the malondialdehyde level of the groups that were fed with the feed compounded with both species of the groundnut compared with the baseline and normal control. However, the malondialdehyde level of the group fed with var-vulgaris was higher than that of Var-hypogaea. Conclusion: These results suggest that the groundnut peel extract of Var-hypogaea and Var-vulgaris is capable of inducing lipid peroxidation in living tissues. The groundnut peel does not only serve as a covering to the groundnut seed but may also protect the seeds from the effect of pests since it exhibits lipid peroxidative activity.

An implantable artificial endocrine pancreas consisting of a glucose sensor and a closed-loop insulin delivery system could potentially replace the need for glucose self-monitoring and regulation among insulin dependent diabetics. Achieving such a break through largely depends on the development of an appropriate, biocompatible membrane for the sensor. Biocompatibility is crucial since changes in the glucose sensors membrane resulting from attack by orinter action with living tissues can interfere with sensor reliability and accuracy. If such interactions can be understood, however, compensations can be made for their effects. Current polymer technology offers several possible membranes that meet the unique chemical dynamics required of a glucose sensor. Two of the most promising polymer membranes are polytetrafluoroethylene (PTFE) and silicone (Si). Low-voltage scanning electron microscopy, which is an excellent technique for characterizing a variety of polymeric and non-conducting materials, 27 was applied to the examination of experimental sensor membranes.

Abstract. Physical exercise increases free radicals production; antioxidant supplementation may improve the muscle fiber’s ability to scavenge ROS and protect muscles against exercise-induced oxidative damage. This study was designed to examine the effects of all-trans resveratrol supplementation as an antioxidant to mediate anti-oxidation and lipid per-oxidation responses to exercise in male Wistar rats. Sixty-four male Wistar rats were randomly divided into four equal number (n = 16) including training + supplement (TS), training (T), supplement (S) and control (C) group. The rats in TS and S groups received a dose of 10 mg/kg resveratrol per day via gavage. The training groups ran on a rodent treadmill 5 times per week at the speed of 10 m/min for 10 min; the speed gradually increased to 30 m/min for 60 minutes at the end of 12th week. The acute phase of exercise protocol included a speed of 25 m/min set to an inclination of 10° to the exhaustion point. Superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) activity, non-enzymatic antioxidants bilirubin, uric acid, lipid peroxidation levels (MDA) and the total antioxidant capacity (TAC) were measured after the exercise termination. The data were analyzed by using one-way ANOVA. The result showed that endurance training caused a significant increase in MDA level [4.5 ± 0.75 (C group) vs. 5.9 ± 0.41 nmol/l (T group)] whereas it decreased the total antioxidant capacity [8.5 ± 1.35 (C group) vs. 7.1 ± 0.55 mmol/l (T group)] (p = 0.001). In addition, GPx and CAT decreased but not significantly (p > 0.05). The training and t-resveratrol supplementation had no significant effect on the acute response of all variables except MDA [4.3 ± 1.4 (C group) vs. 4.0 ± 0.90 nmol/l (TS group)] (p = 0.001) and TAC [8.5 ± 0.90 (C group) vs. 6.6 ± 0.80 mmol/l (TS group)] (p = 0.004). It was concluded that resveratrol supplementation may prevent exercise-induced oxidative stress by preventing lipid peroxidation.

Melatonin reduces high levels of lipid peroxidation induced by potassium iodate in porcine thyroid

Abstract. Iodine is essential for thyroid hormone synthesis. Under normal iodine supply, calculated physiological iodine concentration in the thyroid is approx. 9 mM. Either potassium iodide (KI) or potassium iodate (KIO3) are used in iodine prophylaxis. KI is confirmed as absolutely safe. KIO3 possesses chemical properties suggesting its potential toxicity. Melatonin (N-acetyl-5-methoxytryptamine) is an effective antioxidant and free radical scavenger. Study aims: to evaluate potential protective effects of melatonin against oxidative damage to membrane lipids (lipid peroxidation, LPO) induced by KI or KIO3 in porcine thyroid. Homogenates of twenty four (24) thyroids were incubated in presence of either KI or KIO3 without/with melatonin (5 mM). As melatonin was not effective against KI-induced LPO, in the next step only KIO3 was used. Homogenates were incubated in presence of KIO3 (200; 100; 50; 25; 20; 15; 10; 7.5; 5.0; 2.5; 1.25 mM) without/with melatonin or 17ß-estradiol. Five experiments were performed with different concentrations of melatonin (5.0; 2.5; 1.25; 1.0; 0.625 mM) and one with 17ß-estradiol (1.0 mM). Malondialdehyde + 4-hydroxyalkenals (MDA + 4-HDA) concentration (LPO index) was measured spectrophotometrically. KIO3 increased LPO with the strongest damaging effect (MDA + 4-HDA level: ≈1.28 nmol/mg protein, p < 0.05) revealed at concentrations of around 15 mM, thus corresponding to physiological iodine concentrations in the thyroid. Melatonin reduced LPO (MDA + 4-HDA levels: from ≈0.97 to ≈0,76 and from ≈0,64 to ≈0,49 nmol/mg protein, p < 0.05) induced by KIO3 at concentrations of 10 mM or 7.5 mM. Conclusion: Melatonin can reduce very strong oxidative damage to membrane lipids caused by KIO3 used in doses resulting in physiological iodine concentrations in the thyroid.

Dyslipidemia and oxidative modifications of lipid are frequently associated in patients with chronic kidney diseases (CKD) and considered the most important risk factors for cardiovascular events. Melatonin is a well-known potent antioxidant and has beneficial effect on lipid metabolism. the study was designed to evaluate if Melatonin could improve lipid profile and ameliorates lipid peroxidation. This single blind placebo controlled clinical study carried out on 41 patients with CKD who were randomized into two groups, control groups (n=20) those who received placebo cap and melatonin group those who received 5mg melatonin (n=21). Lipid profile [total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL-C), low-density lipoprotein (LDL-C)] and parameters of lipid peroxidation [oxidized LDL (oxLDL) and malondialdehyde (MDA) were measured before and after 12 weeks of the treatment. After 12 weeks of treatment, melatonin significantly increased HDL-C and decreased LDL-C compared to the initial value. The elevation in HDL-C and reduction in LDL-C were significantly different from that in placebo group. Also, both oxLDL and MDA levels significantly lowered by melatonin compared to the baseline and to the placebo group. Collectively, the results of our study showed that melatonin has advantageous effect on lipid profile and inhibit lipid peroxidation in patients with CKD.
