Algal blooms have been occurring in many regions worldwide, and allelochemicals are important algaecides used to control harmful algal blooms (HABs). The allelopathic effects of linoleic acid (LA) on the harmful raphidophyte Heterosigma akashiwo were studied, and the possible mechanisms were investigated through analyses of population growth dynamics, cellular ultrastructure and the physiological levels of H. akashiwo. The results showed that the inhibitory effect of LA on H. akashiwo cells increased with an increasing LA concentration. The levels of ROS and MDA were significantly elevated, indicating oxidative stress and lipid peroxidation due to LA exposure. At the same time, LA also activated the antioxidant system, including superoxide dismutase (SOD), catalase (CAT), and POD, and non-enzymatic antioxidants such as reduced AsA and glutathione (GSH). Transmission electron microscopy (TEM) revealed that the morphology of the algal cells was impaired in an LA-dependent manner. Annexin V-FITC/PI double staining and flow cytometric analysis revealed that LA exposure decreased the cellular mitochondrial membrane potential (MMP), increased the rate of apoptosis. LA modulated bcl-2/bax homeostasis and increased the expressions of cytochrome c and caspases-3 and -9, proving that LA induced cell death via the mitochondria-mediated apoptotic pathway. It was suggested that LA had allelopathic effects on H. akashiwo, inducing physiological and morphological changes and finally triggering the apoptosis of H. akashiwo. All of these results showed that LA might have the potential as an algaecide to control harmful algae.

The effect of continuous lighting (CL, 24 h) and light spectrum on growth and nutritional quality of arugula (Eruca sativa), broccoli (Brassica oleracea var. italic), mizuna (Brassica rapa. var. nipposinica), and radish (Raphanus sativus var. radicula) were investigated in growth chambers under light-emitting diode (LED) and fluorescent lighting. Microgreens were grown under four combinations of two photoperiods (16 h and 24 h) providing daily light integral (DLI) of 15.6 and 23.3 mol m−2 day−1, correspondingly) with two light spectra: LED lamps and fluorescent lamps (FLU). The results show that fresh and dry weights as well as leaf mass per area and robust index of harvested arugula, broccoli, mizuna, and radish seedlings were significantly higher under CL compared to 16 h photoperiod regardless of light quality. There were no visible signs of leaf photodamage. In all CL-treated plants higher chlorophyll a/b and carotenoid-to-chlorophyll ratios were observed in all plants except mizuna. CL treatment was beneficial for anthocyanin, flavonoid, and proline accumulation. Higher activities of antioxidant enzymes (catalase, superoxide dismutase, ascorbate peroxidase, and guaiacol peroxidase) were also observed in CL-treated plants. In most cases, the effects were more pronounced under LED lighting. These results indicate that plants under mild oxidative stress induced by CL accumulated more non-enzymatic antioxidants and increased the activities of antioxidant enzymes. This added nutritional value to microgreens that are used as functional foods providing health benefits. We suggest that for arugula, broccoli, mizuna, and radish, an LED CL production strategy is possible and can have economic and nutritional benefits.

In this study, the contribution of nonenzymatic (ascorbate, glutathione) and enzymatic antioxidants (superoxide dismutase, catalase, glutathione reductase, glutathione S-transferase) in the first hours of recovery of the resurrection plant Haberlea rhodopensis from drought- and freezing-induced desiccation was assessed. The initial stage of recovery after desiccation is critical for plants, but less investigated. To better understand the alterations in the activity of antioxidant enzymes, their isoenzyme patterns were determined. Our results showed that ascorbate content remained high during the first 9 h of rehydration of desiccated plants and declined when the leaves′ water content significantly increased. The glutathione content remained high at the first hour of rehydration and then strongly decreased. The changes in ascorbate and glutathione content during recovery from drought- and freezing-induced desiccation showed great similarity. At the beginning of rehydration (1–5 h), the activities of antioxidant enzymes were significantly increased or remained as in dry plants. During 7–24 h of rehydration, certain differences in the enzymatic responses between the two plant groups were registered. The maintenance of a high antioxidant activity and upregulation of individual enzyme isoforms indicated their essential role in protecting plants from oxidative damage during the onset of recovery.

(1) Background: Peach cv. GF305 is commonly used in breeding programs due to its susceptibility to numerous viruses. In this study, we aimed to achieve a methodology for rapid and uniform seed germination of peach cv. GF305 in order to obtain vigorous seedlings; (2) Methods: A combination of cold stratification and H2O2 imbibition was tested on peach seeds with or without endocarp. In addition, the levels of non-enzymatic antioxidants ascorbate and glutathione as well as the hormone profile in seedling roots and shoots were determined; (3) Results: We found that H2O2 imbibition of peach seeds without endocarp after 8 weeks of stratification increased germination rate and resulted in seedlings displaying good vegetative growth. The H2O2 imbibition also affected the levels of ascorbate, glutathione, and the phytohormones abscisic acid and jasmonic acid in peach seedlings; (4) Conclusions: Although stratification periods of 12 weeks have been previously established as being appropriate for this cultivar, we have been able to reduce this stratification time by up to 4 weeks, which may have practical implication in peach nurseries.

The antioxidant parameters of black pine (Pinus nigra J.F. Arnold) needles samples collected from different areas in the province of Diyarbakır, Turkey were studied. Black pine needles samples were collected from three different locations (Egil, Kosuyolu, and Diyarbakır industrial area) in the province of Diyarbakır in summer and winter seasons. The activity of enzymatic antioxidants such as superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), glutathione S-transferase (GST) and malondialdehyde (MDA) levels of the samples were determined by ELISA method. Results showed that SOD enzyme activities of the samples collected in summer from Kosuyolu and the industrial area decreased compared to the control group, but increased in the samples collected in winter from the same areas. When compared to the control group, it was observed that the MDA levels of the samples collected from Kosuyolu and the industrial area increased. Bangladesh J. Seed ageing is associated with a high concentration of reactive oxygen species (ROS). Apple (Malus domestica Borkh.) seeds belong to the orthodox type. Due to a deep dormancy, they may be stored in dry condition at 5 °C for a long time, without viability loss. In the laboratory, artificial ageing of apple seeds is performed by imbibition in wet sand at warm temperature (33 °C). The aim of the work was to study nitric oxide (NO) as a seed vigour preservation agent. Embryos isolated from apple seeds subjected to accelerated ageing for 7, 14, 21 or 40 days were fumigated with NO. Embryo quality was estimated by TTC and MDA tests. ROS level was confirmed by NBT staining. We analysed the alteration in transcript levels of CAT, SOD and POX. NO fumigation of embryos of seeds aged for 21 days stimulated germination and increased ROS level which correlated to the elevated expression of RBOH. The increased total antioxidant capacity after NO fumigation was accompanied by the increased transcript levels of genes encoding enzymatic antioxidants, that could protect against ROS overaccumulation. Moreover, post-aged NO application diminished the nitro-oxidative modification of RNA, proving NO action as a remedy in oxidative remodelling after seeds ageing.

Abstract There is recently a fast growing interest in green nanotechnology as a better approach for managing diabetes mellitus (DM) and its associated complications. This study therefore aimed at exploring the protective potentials of biogenic phytomediated magnesium-based nanoparticles using Monodora myristica seed (Mg(OH)2NP-Mm) against streptozotocin-induced diabetic hepatopathy, nephropathy and neuropathy in Wistar rats. Mg(OH)2NP-Mm was biosynthesized and characterized using standard techniques. Forty-eight (48) adult male Wistar rats weighing 150-200 g, were indiscriminately grouped into eight (8) groups of six (6) rats (n=6) each. Diabetes was induced with a low dose of streptozotocin (STZ) (55 mg/kg bw) and diabetic animals administered 50, 100, 150 and 200 mg/kg bw Mg(OH)2NP-Mm for 21 days, while control groups received glibenclamide (5 mg/kg bw) and Mg(OH)2-STD (150 mg/kg bw), respectively. In this study, treatment with Mg(OH)2NP-Mm caused a significant (p < 0.05) improvement in fasting blood sugar (FBG), serum hepatic biomarkers (AST, ALT and ALP activities), renal clearance markers (creatinine and urea), total protein (TP) and bilirubin. Enzymatic and non-enzymatic antioxidants, as well as histomorphological examinations indicated a significant (p < 0.05) restoration of the hepatic, renal and brain tissues. This finding therefore, hypothesizes attenuation of redox imbalance which probably could be the basis for the protective effect demonstrated by Mg(OH)2NP-Mm in the tissues examined from STZ-induced diabetic rats.

Lotus is an aquatic plant that is sensitive to water loss, but its seeds are longevous after seed embryo dehydration and maturation. The great difference between the responses of vegetative organs and seeds to dehydration is related to the special protective mechanism in embryos. In this study, tandem mass tags (TMT)-labeled proteomics and parallel reaction monitoring (PRM) technologies were used to obtain novel insights into the physiological regulatory networks during lotus seed dehydration process. Totally, 60,266 secondary spectra and 32,093 unique peptides were detected. A total of 5,477 proteins and 815 differentially expressed proteins (DEPs) were identified based on TMT data. Of these, 582 DEPs were continuously downregulated and 228 proteins were significantly up-regulated during the whole dehydration process. Bioinformatics and protein-protein interaction network analyses indicated that carbohydrate metabolism (including glycolysis/gluconeogenesis, galactose, starch and sucrose metabolism, pentose phosphate pathway, and cell wall organization), protein processing in ER, DNA repair, and antioxidative events had positive responses to lotus embryo dehydration. On the contrary, energy metabolism (metabolic pathway, photosynthesis, pyruvate metabolism, fatty acid biosynthesis) and secondary metabolism (terpenoid backbone, steroid, flavonoid biosynthesis) gradually become static status during lotus embryo water loss and maturation. Furthermore, non-enzymatic antioxidants and pentose phosphate pathway play major roles in antioxidant protection during dehydration process in lotus embryo. Abscisic acid (ABA) signaling and the accumulation of oligosaccharides, late embryogenesis abundant proteins, and heat shock proteins may be the key factors to ensure the continuous dehydration and storage tolerance of lotus seed embryo. Stress physiology detection showed that H2O2 was the main reactive oxygen species (ROS) component inducing oxidative stress damage, and glutathione and vitamin E acted as the major antioxidant to maintain the REDOX balance of lotus embryo during the dehydration process. These results provide new insights to reveal the physiological regulatory networks of the protective mechanism of embryo dehydration in lotus.

To address the demand for natural fibers, developing new varieties that are resistant to abiotic stress is necessary. The present study was designed to investigate the physiological and biochemical traits of three varieties of C. capularis (Y49, Y38, and Y1) and four varieties C. olitorius (T8, W57, M33, M18) under low temperature to identify the cold-tolerant varieties and elucidate the mechanisms involved in enhancing cold tolerance. Research findings revealed that the varieties Y49 and M33 exhibited the highest chlorophyll and carotenoid content. Biochemical profiles revealed that varieties Y49 and M33 were found to be able to withstand low-temperature stress by accumulating different enzymatic and non-enzymatic antioxidants, such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APx), glutathione (GSH), and phenolics, which participated in reducing the content of malondialdehyde (MDA) and hydrogen peroxide (H2O2) caused by low temperature. Osmolytes compounds, such as total soluble sugar, significantly increased in Y49 and M33; and proline content decreased in all varieties except Y49 and M33 after low-temperature exposure. The rise in these osmolytes molecules can be a defense mechanism for the jute’s osmotic readjustment to reduce the oxidative damage induced by low temperature. Furthermore, PCA and hierarchical cluster analysis distinguished the seven varieties into three separate groups. Results confirmed that group I (Y49 and M33 varieties) were low-temperature tolerant, group II (M18, W57) were intermediate, whereas III groups (Y38, T8, and Y1) were low temperature susceptible. PCA also explained 85.72% of the variance of raw data and clearly distinguished three groups that are similar to the cluster heat map. The study thus confirmed the tolerance of selected varieties that might be an efficient adaptation strategy and utilized them for establishing breeding programs for cold tolerance.

Abstract Cardiovascular disease (CVD) is a global illness causing 31% of global mortality. Though many factors contribute to CVD, oxidative stress advances atherosclerosis through several complementary components, such as the initiation of lipid peroxidation by iron. Blood donation may decrease the risk of CVD due to reducing the iron level. Literature reported that blood donors have a lower risk of CVD, possibly due to the lower iron levels. Various effects of blood donation are involved in preventing type II diabetes. However, little is known of the exact mechanism of the benefits of blood donation. In this study, samples were collected from 33 healthy male participants pre- (1 day) and post-donation (1 day, 1, 2 and 3 weeks) and the effect of the blood donation on the iron, lipids and enzymatic antioxidants profiles were assessed. A repeated-measures ANOVA was used for comparing the quantitative variables between the visits. We found that the iron decreased significantly by week 1 (–25.3%). Ferritin decreased significantly at weeks 1, 2, and 3 (–26.3%, –40.3%, –36.7%, respectively). The superoxide dismutase increased significantly at post-donation day 1, weeks 1, 2, and 3 (17.9%,35.7%, 31.1%, 36.6%, respectively) and in correlation with time [r (165) = 0.50, P <0 .01]. Glutathione peroxide decreased significantly at week 1 (–25.0%). Glutathione reductase decreased significantly 1-day post donation (–5.7%) then increased over the next three weeks [r (165) = 0.3, P <0.01]. Finally, the lipids were significantly reduced 24 hours after the donation but not at week 1, 2 and 3. We conclude that blood donation, resulting in a lowered body iron concentration, is an effective way to increase superoxide dismutase and glutathione reductase, which prevent the initiation of lipid oxidation. Our results could be used to advocate for the benefits of blood donation. However, further studies are required to assess the role of blood donation in plaque formation and arteriosclerosis.
