Morphological and molecular characterisation of Xiphinema mazandaranense n. sp. (Dorylaimida: Longidoridae), a new member of the Xiphinema pyrenaicum species complex

Xiphinema mazandaranense n. sp. is described and illustrated. The new species belongs to morphospecies group 6, which is characterised by having two equally developed female genital branches that have spines in the uteri and a short, rounded tail. The new species is characterised by having spines in the tubular portion of the uterus, body 3.7-5.2 mm long, odontostyle 163-173 μm long, odontophore 96-100 μm long, body 62-80 μm diam., rounded tail, four juvenile stages, males rare with three ventromedian supplements next to the adcloacal pair and spicules 85 μm long. The polytomous identification codes of the new species are: A4-B3-C7b-D6-E456-F45-G4-H2-I23-J7b-K2-L1. The new species appears closely related to the members of X. pyrenaicum group which are characterised by a rounded tail with or without an inconspicuous projecting bulge and a uterus devoid of Z-differentiation but showing spiniform structures. The new species differs from members of the X. pyrenaicum group, which includes the recently described X. iranicum, by a more rounded tail without any projection. Beside morphological and morphometric data, molecular analyses of the near-full-length small subunit rDNA gene (SSU) placed the new species in close relationship with some species belonging to Xiphinema morphospecies group 6 and further separated this species from the X. pyrenaicum complex.

Xiphinema zagrosense sp. n. is described based on morphological and molecular characters. The new species belongs to morphospecies group 6 characterised by having two equally developed female genital branches with spines in uterus and a short, rounded tail. The new species is characterised by having spines in the tubular part of the uterus, body length 3.8-5.0 mm, odontostyle length 151-169 μm, odontophore 94-105 μm, conical, dorsally convex tail, four juvenile stages and absence of males. The new species appears closely related to the members of X. pyrenaicum group characterised by a rounded tail with or without an inconspicuous digitate end and a uterus devoid of Z-differentiation but showing spiniform structures. It is compared with X. aceri, X. adenohystherum, X. iranicum, X. mazandaranense, X. nuragicum, X. pyrenaicum, X. robbinsi, X. sphaerocephalum and X. vuittenezi. In addition to morphological and morphometric data, molecular analyses of the near-full-length small subunit rDNA gene (SSU) and large subunit rDNA gene (LSU) D2/D3 placed the new species in close relation with some species belonging to Xiphinema group 6 but differs from other species in the X. pyrenaicum complex.

Xiphinema granatum n. sp. from pomegranate in Saveh, Markazi province, central Iran, is described and illustrated. The new species belongs to species group 8 of the genus Xiphinema sensu Loof & Luc, 1990. The new species is characterised by a body 3.5-4.1 mm long, 37-49 μm diam., odontostyle and odontophore 118-132 and 65-74 μm long, respectively, presence of sperm in the female genital tracts, uterus devoid of any Z-differentiation or spines, and abundant males with spicules 45-58 μm long. The polytomous identification codes of the new species are: A4, B4, C5a, D5, E56, F4, G23, H2, I3, J4, K2, L2. Beside morphological and morphometric data, molecular analyses of D2-D3 fragments of 28S rRNA gene placed the new species in close relationship with X. vuittenezi and X. italiae and some species belonging to Xiphinema morphospecies group 6, but clearly separated from X. index. In addition, an Iranian population of Longidorus pisi from the same locality is morphologically and molecularly characterised.

A new species, Eudorylaimus piceae n. sp., extracted from rhizosphere soil of Picea crassifolia from Inner Mongolia, China was identified. The new species is characterized by these combined characters: body length of 1.03–1.27 mm; lip region distinctly offset; odontostyle 20–22 μm and 1.1–1.4 times the lip region diameter in length; odontophore 1.1–1.2 times the odontostyle length; basal expansion of pharynx occupying 42%–50.5% of the total neck length; genital system didelphic-amphidelphic; vulva transverse; vagina extending inwards 32%–43%; V value averaging more than 60; pars refringens vaginae with two trapezoidal sclerotizations and pars distalis vaginae weakly sclerotized; prerectum 72–107 μm long, 2.3–3.3 times the anal body diameter, and rectum 1–1.6 times the anal body diameter in length; tail conoid and bent ventrally, c’ value 1.5–1.8 and males unknown. Phylogenetic analyses based on sequences of 18S small subunit rDNA and the D2-D3 expansion region of 28S rDNA are presented.

Abstract Hemiplectus muscorum, the type and single representative of its genus, is redescribed on the basis of abundant new material collected in the UK, Canada and the USA using both light and scanning electron microscopy. The phylogenetic relationships of the species are inferred from morphological as well as molecular data. Maximum parsimony, neighbour joining and maximum likelihood analyses of small subunit (SSU) rRNA sequences support a position nested among the Plectidae. This conflicts with our morphological assumptions of character polarity, as it implies that the absence of a valvate bulb in Hemiplectus is a reversal rather than a plesiomorphy. The excretory system of Hemiplectus is described more precisely. Its structure is highly reminiscent of the system in Plectus but differs in the presence of an anterior and posterior pair of pseudocoelomocytes flanking the renette cell. A pair of lateral somatic setae is identified as possible homologues of the 'deirids' in Plectus and Rhabditida. Measurements and descriptions are given of all four juvenile stages.

Summary Six species of the genus Tylencholaimellus, one new and five known, collected in virgin and cultivated soils of East Azarbaijan province, Iran, are studied. Tylencholaimellus zeinabadensis sp. n. is morphologically characterised by its 1.2-1.7 mm long body, lip region 10-13 μm wide and offset by deep and broad constriction, odontostyle 16-20 μm long, stylet 22-30 μm long, neck 186-234 μm long, pharyngeal bulb 30-43 μm long, anterior uterine sac 55-103 μm long or 0.9-1.7 corresponding body diam. long, V = 34-41, tail short and rounded to conoid (20-30 μm, c = 45-83, c′ = 0.5-1.0), spicules 39-50 μm long, and one or two ventromedian supplements with hiatus. Molecular analyses using the D2-D3 expansion regions of the 28S rRNA gene sequences of the new species confirmed the monophyly of this genus based upon currently available data. A population of T. affinis is described and illustrated, and new data (morphometrics, pictures and distribution) are provided for T. loofi, T. paracinctus, T. polonicus and T. striatus.

Summary A new species of Aporcelinus, collected in a natural habitat of central Mexico, is described and illustrated, including molecular (D2-D3 28S-rRNA) data. Aporcelinus zapotitlanensis sp. n. is characterised by its 1.19-1.43 mm long body, lip region offset by a deep constriction and 16.5-18.5 μm broad, odontostyle 19.5-22 long, neck 325-368 μm long, pharyngeal expansion 150-201 μm long or 46-55% of the total neck length, uterus 130-167 μm long and tripartite, V = 54-56, tail conical with finely rounded tip (30-37 μm, c = 34-43, c′ = 1.2-1.4) lacking a distinct dorsal concavity, spicules 48-51 μm long, and 11-12 spaced ventromedian supplements with one or two of them located within the range of the spicules. Molecular analyses confirm the monophyly of Aporcelinus and places the new species within a clade shared with A. infundibulicaudatus and A. amazonicus.

Summary A new dimorphic species of Deladenus isolated from Sirex californicus from Washington, USA, is described as D. beddingi n. sp. Evolutionary relationships of the new species with other Deladenus species were assessed using multilocus sequencing. Phylogenetic relationships derived from analyses of mtCO1 and ITS showed D. beddingi n. sp. to be genetically distinct from other North American Deladenus parasitising Sirex. Molecular analyses indicated that D. beddingi n. sp. is a member of the D. siricidicola species complex, which also includes undescribed native Deladenus from Sirex cyaneus and S. nitidus, and D. siricidicola from S. noctilio. Mycophagous adults were characterised by the position of the excretory pore, which was located 32 (22-52) and 48 (38-69) μm anterior to the hemizonid in mycophagous females and males, respectively. Typologically, the new species is most similar to D. siricidicola, D. proximus and D. nitobei, but can be distinguished from these species by several morphometric traits, including the value of ratios a, b, c of the mycophagous females and males, ratio b of the infective females, and the morphology of the tail of the mycophagous females, which is narrow and gradually tapering. This novel nematode species feeds on the fungus Amylostereum chailletii during its mycophagous phase. Experimental results showed very little reproduction by D. beddingi n. sp. when feeding on A. areolatum compared to robust reproduction when feeding on A. chailletii.

Summary A new species of Aporcelaimellus, collected in a watermelon field in Nigeria, is described, including its morphological and molecular (D2-D3 28S-rDNA, 18r-DNA) characterisation. Aporcelaimellus nigeriensis sp. n. is distinguishable by its 2.76-3.55 mm length, very coarse ventral body pores, lip region offset by deep constriction and 24-27 μm broad odontostyle 30-36 μm long at its dorsal and 28-31 μm at its ventral side, neck 648-779 μm long, pharyngeal expansion occupying 54-60% of total neck length, uterus 300-473 μm or 2.1-3.2 body diam. long and tripartite, V = 49-54, tail short and convex conoid (27-41 μm, c = 72-115, c′ = 0.5-0.7), spicules 108-137 μm long, and 9-10 spaced ventromedian supplements with hiatus. LSU analysis revealed a close relationship of A. nigeriensis sp. n. with other Aporcelaimellus species and questioned, once more, the monophyly of Aporcelaimidae. SSU phylogenetic tree was not able to resolve the relationship between the new species and other closely related species.

Ciliates infected 0.25% of adult Aedes sp. mosquitoes collected in Guelph, Ontario. Morphological observations of live and stained specimens indicated that these ciliates belonged to the Tetrahymena pyriformis sibling species complex. This study provides the first well-documented case of insects being infected by a species in the T. pyriformis complex. Mating experiments demonstrated that these "mosquito" ciliates were reproductively isolated from previously described biological species in the complex, and are designated a new species, Tetrahymena empidokyrea n.sp. Phylogenetic analysis using SSrDNA sequences suggested that T. empidokyrea n.sp. is either basal to all species in the genus Tetrahymena or basal to one of the two main lineages in that genus.

AbstractPerlesta placida (Hagen), long regarded as a widespread variable species, is a complex of at least 12 species which differ in pigmentation, egg morphology and male and female genital structures. Species removed from the synonymy of P. placida (= Chloroperla virginica Banks, 1898; = P. virginica immaculata Klapálek, 1921) include P. cinctipes (Banks, 1905), P. decipiens (Walsh, 1862) (= Perla brunnipennis Walsh, 1862, syn. n.; = Isoperla texana Banks, 1914, syn. n.; = P. costalis Klapálek, 1921, syn. n.) and P. nitida Banks, 1948. New species described include P. adena, P. baumanni, P. bolukta, P. browni, P. lagoi, P. nelsoni and P. shubuta. Lectotypes are designated for most older species, including P. frisoni Banks, 1948, which is also valid, and all are redescribed from types.
