Phylogenetic Relationships of Species of Hypselobarbus (Cypriniformes: Cyprinidae): An Enigmatic Clade Endemic to Aquatic Systems of India

Very little is known about the diversity and systematics of the genus cypriniform genus Hypselobarbus. Currently, the genusincludes at least eleven species, all endemic to freshwater systems of Peninsular India. While these species are commonlyknown in India and are frequently used as a food source, little is known about the morphological diversity within and betweenspecies and nothing is known regarding intraspecific genetic diversity or species relationships. Herein, we examine the geneticdiversity in the genus for 11 mitochondrial genes for eleven populations representing nine of the known 11 species.Hypselobarbus is resolved as monophyletic, with the inclusion of P. carnaticus, and species relatioships are very stronglysupported. Because of the unambiguous relationships strongly supported B. carnaticus is allocated to Hypselobarbus. Thisresearch and ongoing morphological and molecular work with the genus supports the existence of additional new species inpeninsular India in need of further molecular and morphological study. Genetic diversity in the genus is high; for the twospecies wherein more than one sample, and the two of each are suspected to represent undescribed taxa, these populationsexhibited greater genetic divergence than that observed between any two of the other currently recognized species,corroborating our hypothesis based on morphological evidence. Clearly the genus warrants more thorough geographicsampling and examination of morphological and molecular data/analyses to reveal the natural lineages existing in this endemic and enigmatic genus.

Parasesarma semperi (Bürger, 1893) was first described from Bohol in the Philippines and is considered to be widely distributed in Southeast Asia. Parasesarma longicristatum (Campbell, 1967) was originally described as a subspecies of P. semperi from Queensland, Australia, and later recognised as a full species. In this study, we re-examine specimens of the two species from across their entire geographic range using genetic markers, a morphometric analysis, and traditional morphological characters. Previous taxonomic species diagnoses were found to be unreliable, but morphometric principle component analyses consistently separate the two species, with the length to width ratio of the propodus of the fourth pereiopod being of particular importance. Genetic data corresponding to the mitochondrial genes COI, ND1 and 16S confirmed a close sister relationship between the two species, forming reciprocally monophyletic groups. Both species have high haplotype diversities and high intraspecific gene flow.

A new species of Cymbidium (Orchidaceae), Cymbidium xichouense, from Yunnan Province, China, is described and illustrated based on morphological evidence and molecular analyses. The new orchid is morphologically similar to C. qinbeiense, but it has several morphological features that distinguish it from C. qiubeinense and all other recognized species in Cymbidium. Phylogenetic analyses based on nuclear (ITS) and plastid DNA (matK) were conducted, and the results also supported the status of C. xichouense as a new species, which is sister to C. qiubeiense.

A molecular phylogeny of the western Palearctic weevil genus Kyklioacalles Stüben, 1999 is presented, combining two mitochondrial genes (CO1 and 16S) in a Bayesian analysis. Based on molecular data, the validity of the subspecies Kyklioacalles punctaticollis punctaticollis (Lucas, 1849) and Kyklioacalles punctaticollis meteoricus (Meyer, 1909) is discussed and the morphological differentiation of the endophalli and known distributions of both subspecies are verified. Glaberacalles subg. n. (formerly Kyklioacalles punctaticollis-group) and two new species are described, Kyklioacalles atlasicus sp.n. from Morocco and Kyklioacalles plantapilosus sp.n. from Spain. Kyklioacalles berberi (Stüben, 2005), comb. n. and Kyklioacalles olcesei (Tournier, 1873) comb. n. are transferred from Acalles Schoenherr. The molecular results further advocate a transfer of Onyxacalles pyrenaeus (Boheman, 1844) to Kyklioacalles; however this is not supported by morphological evidence. Kyklioacalles almadensis Stüben, 2004 syn. n. (Spain) is synonymized with Kyklioacalles bupleuri Stüben, 2004 (Tunisia). A catalogue of all 40 (sub-)species of Kyklioacalles is given and a key of the species of the subgenus Glaberacalles is presented.

Abstract. Azimishad F, Sheidai M, Talebi SM, Noormohammadi Z. 2019. Species relationship and genetic diversity in some Iranian Lamium L. species using ISSR markers. Biodiversitas 20: 1963-1972. Lamium is a widespread and taxonomically complex genus of Lamiaceae which comprises of 16-38 species. This genus is represented in Iran by nine species. In the present study, we used morphological and molecular (ISSR, Cp DNA, and nrITS) data to evaluate species relationships, genetic diversity and population genetic structure of the genus. 27 morphological characteristics, including 13 qualitative and 14 quantitative, and ten ISSR markers were used for morphological and genetical evaluation of 73 accessions from eight taxa. In general, species relationships obtained from morphological and molecular data were largely congruent. In the morphological study, characteristics like the life form, leaf shape, absence/existence of bracts and shape of corolla, were distinctive traits and we did not encounter intermediate forms. Our findings indicated a very high efficiency of the ISSR markers in the identification and delimitation of Lamium species. These results confirmed the placement of L. galeobdolon in the genus Lamium and segregation of L. purpureum and L. garganicum in section Lamium. AMOVA analysis revealed that the species of this genus are genetically differentiated. Nm analysis showed very low value of gene flow among the studied species and mantel test indicated isolation by distance occurred among them.

AbstractContinuous seed propagation in Turkey has given rise to a great number of seedling walnut trees which represents valuable walnut genetic resources. The number of native walnut trees is estimated to be over 5 million in Turkey and they possess large phenotypic variability in yield, nut and kernel characteristics, late bud breaking, late flowering, winter hardiness and tolerance to diseases. Progress in walnut breeding requires the exploitation of genetic variation among cultivars and landraces. In this study, we used 32 local diverse walnut genotypes obtained from seeds and 2 standard cultivars (‘Sebin’ and ‘Bilecik’). This study implemented 21 previously used simple sequence repeats (SSR) markers to determine genetic diversity. The analysis revealed 135 alleles with an average of 6.43 alleles per locus. Genetic similarity ranged from 0.23 (for samples KW22 and KW29) to 0.87 (for samples KW27 and KW28). The highest number of alleles per locus was obtained from WGA276 locus (11 alleles), followed by WGA054 (9 alleles), WGA202 and WGA321 (8 alleles) while the lowest number was detected in WGA027. According to the morphological and molecular data, the genotypes differed from each other and the cvs. Sebin and Bilecik. The majority of the genotypes had higher fruit weight and some of the genotypes had higher kernel ratio than cvs. Sebin and Bilecik implying the importance of registering genotypes as cultivars. This research provides information on the genetic relationship of walnut genotypes and cultivars and emphasises the importance of protection and utilisation of seed-propagated walnut genetic resources.

Eight minor and neglected cultivars of Vitis vinifera L. were characterised according to their ampelographic and agronomic traits and discriminated by molecular analysis with SSR markers. These results are the first complete ampelographic description of these minor and neglected cultivars from the Mount Etna region. The results of this study reveal high morphological diversity of ancient grapevines growing in this region. SSR markers enabled us to discriminate the cultivars and revealed the genetic divergence between them and several autochthonous cultivars. Our efforts could contribute to a better knowledge of grape biodiversity based on morphological and molecular data and could be useful for the development of a reliable germplasm conservation strategy.

A new species, Camellia psilocarpa (Theaceae), from Guangdong Province, China, is described and illustrated. It is morphologically most similar to C. magnocarpa, from which it differs by having ovate-oblong leaves with apex caudate or long acuminate, bracteoles and sepals 10-12, grayish white velutinous outside, and especially smaller fruit. The morphological evidence shows that the new species is a member of Camellia sect. Camellia. Additionally, a molecular and phylogenetic analysis using two chloroplast DNA markers also gave support to its taxonomic position.

AbstractTaxonomy and phylogeny of termites of the genus Reticulitermes in central and eastern Mediterranean lands are poorly understood, partly due to insufficient sampling. This study aims to contribute to the knowledge of east Mediterranean termites by presenting morphological and molecular data on Reticulitermes from Cyprus. Samples from 15 colonies were collected throughout the island. Qualitative and quantitative morphological characters were analyzed for alate and soldier castes. Partial sequences of the mitochondrial genes COII and 16S were used to evaluate genetic diversity of Cypriot colonies and to determine their phylogenetic relationships with taxa from central and eastern Mediterranean areas.Cypriot alates have some morphological features in common with the Israeli R. clypeatus: an enlarged postclypeus and a wide unpigmented margin of the eye. They are larger than R. clypeatus but smaller than western European species, such as R. banyulensis, R. lucifugus corsicus and R. grassei. For Cypriot soldiers, two size groups were identified, possibly in relation with the age of their mother colonies.Phylogenetic analysis shows that, contrary to what might be expected, the samples with the highest affinity with Cypriot samples are not those from the nearby mainland (south Turkey, Israel), but from north-eastern Greece. Comprehensive sampling in the nearby mainland is lacking, so the possibility that populations exist it that region with an affinity towards Cypriot Reticulitermes sp. cannot be ruled out. Together with samples from the Halkidiki peninsula, north-eastern Greece, northern Turkey and Crete, Cypriot Reticulitermes form a well-supported north-eastern Mediterranean clade.

Abstract Polyploidization is generally considered a major evolutionary force that can alter the genetic diversity, morphology, physiology and ecology of plants. One striking example is the polyploid Urtica dioica complex, in which diploid taxa are often found in remote and partly relictual geographical ranges, in contrast to tetraploid individuals, which have an unknown evolutionary history and occur in a variety of synanthropic habitats. We used a set of 279 plants, evenly representing the geographical and morphological variation of U. dioica s.l. in Europe and Southwest Asia, and employed multivariate and geometric morphometrics and Hyb-Seq sequencing to estimate the extent of differentiation of diploid taxa and the ubiquitous tetraploid cytotype. Diploid subspecies form more-or-less separate clusters in morphological analyses, but our molecular evaluation did not reveal any structure. Moreover, tetraploids coalesced with diploids in both morphological and molecular analyses. This disparity between morphological and molecular data might be driven by (1) local adaptation of the diploid cytotype that is mirrored in specific phenotypes, (2) only recent genetic diversification of the group and (3) homoploid and heteroploid hybridization events.

Sesame (Sesamum indicum L.), is the most ancient and important oilseed crop for its good quality edible oil. Much variability among genotypes exists in sesame which helps the breeder to choose good parents in hybridization. In the present study, 30 genotypes were studied for 12 morphological traits. Correlation analysis exhibited that seed yield per plant was positive and significantly correlated with capsules per plant and 1000-seed weight. Apart from morphological data, initial genotyping was conducted for 32 microsatellites but finally only polymorphic primers were selected for genetic diversity study. Estimated polymorphism information content (PIC) value ranged from 0.07 to 0.87 and SSR 24 showed to be highest at 0.87. The dendrogram was constructed using the software NTSYS Pc Ver. 2.20 based on morphological and molecular data. Geographical diversity did not corroborate with genotypic diversity as the cluster composition revealed that genotypes belonging to different geographical origin grouped in the same cluster.
