Detection and residence time of bisphosphonates in bone of horses

Bisphosphonates are potent anti-resorptive agents that have the potential to adversely affect bone healing in equine athletes, and normal bone adaption in young racehorses. A concern exists that bisphosphonate inhibition of normal bone metabolism could lead to increased bone fractures during high-intensity exercise. We found only a single report describing concentrations of tiludronate in the bone of horses, and no studies describing clodronate. Knowledge of the residence time in bone could allow for a better understanding of the long-term effects of these compounds. Our objectives were to develop a method for detection of bisphosphonates in bone and add to the limited information available regarding the disposition of these drugs in the bone of horses. Two horses received clodronate and 2 tiludronate disodium. Postmortem collection of bones and teeth occurred either 4 or 30 d post drug administration. Additionally, postmortem blood, synovial fluid, aqueous humor, and bone samples from racehorses with various histories of bisphosphonate administration were collected, and concentrations determined using the developed LC-MS/MS method. Bisphosphonates were detected in bones and teeth tested at 4 and 30 d. In a postmortem sample, clodronate was detected in bone from a horse with reported administration 18 mo prior; clodronate was not detected in other sample types collected from this horse. Bisphosphonates reside in bone for extended periods of time, which could lead to potential long-term effects, increasing the potential for bone fractures in young and/or athletic horses.

Abstract The waterways of the lower Great Lakes and St. Lawrence River, between Sarnia and the Saguenay Fiord, are made up of four limnological units. The first comprises the high discharge, rapid flow rivers, namely the St. Clair, Detroit, Niagara and St. Lawrence. Second are the four shallow, short residence time, riverine lakes, namely St. Clair, St. Francois, St. Louis and St. Pierre. Third are the two, relatively deep, long residence time, lower Great Lakes Erie and Ontario. Lastly, there is the freshwater-salt water mixing zone of the upper St. Lawrence Estuary. The rivers are essentially sources and transport systems of toxic contaminants on a grand scale. The riverine lakes provide only temporary storage or sinks even for contaminants associated with sediments because these are eventually resuspended and moved on downstream. The major sinks, where long-term effects are most evident are the two lower Great Lakes and the St. Lawrence Estuary. These sites are also where sediment associated contaminants can be permanently removed by deep burial in bottom sediments. However, even here, a proportion of the contaminant load passes on downstream and eventually out to the Gulf of St. Lawrence. The distinctive characteristics of the four limnological units are discussed in relation to sources and fate of toxic contaminants. Understanding the role of the units is critical to development of toxic chemicals control strategies and reduction in aquatic ecosystem contamination.

It is generally known that during the 100 days of genocide against the Tutsi in Rwanda in 1994, sexual violence was committed on an unprecedented scale. Many women were first raped and then killed. With a certain degree of probability, the majority of Tutsi women who survived had been raped. Limited information is available regarding the experiences of these women. However, there is enough empirical evidence provided in human rights accounts and research reports substantiating that these women were exposed to unimaginable horror, which for the majority of them had a range of devastating short and long term effects. The programme of community-based sociotherapy was implemented in 2005 in the north of Rwanda in what was previously known as Byumba province, and subsequently in 2008 in Bugesera district in the south-east, one of the epicentres of the genocide.

Case summary A 7-year-old neutered male Norwegian Forest Cat was presented with decreased appetite and activity, weight loss, fever, neutrophilia and hyperglobulinaemia. A physical examination showed painful stifle joints and enlarged popliteal lymph nodes. Blood examination showed neutrophilia, hyperglobulinaemia and increased serum amyloid A. Urinalysis, thoracic and abdominal radiographs, and abdominal ultrasonography were unremarkable. Synovial fluid from the knee joints had diminished viscosity and revealed neutrophilic inflammation on the smear. There was no evidence of infection in a microbiological culture of the synovial fluid. A diagnosis of idiopathic immune-mediated polyarthritis (IMPA) was made. Prednisolone was initiated at 2 mg/kg q24h PO and tapered with additional immunosuppressants (leflunomide, ciclosporin A and methotrexate); however, prednisolone could not be discontinued. Informed consent was obtained from the owner and mycophenolate mofetil (MMF) at a dosage of 10 mg/kg q12h PO was initiated on day 798. There were no adverse effects of MMF and prednisolone was discontinued on day 1183. Clinical signs resolved and the cat’s general condition remained stable with MMF alone at a dosage of 10 mg/kg q48h PO on day 1600. Relevance and novel information There is limited information describing feline IMPA and its treatment options other than the use of prednisolone. This is the first report of the successful treatment and long-term follow-up of feline IMPA with MMF. MMF may be a safe and effective option as an additional immunosuppressant in feline IMPA.

Limited information is available on the long-term effects of providing care for adults when caregiving begins in childhood in the United States. The current study provided an examination of the effects of youthful caregiving on the mental health of these persons when adults, and provided a description of their early family relations. Twenty-four individuals, 21 to 58 years old, were given brief phone interviews with semistructured questions about their early caregiving experiences, and then they completed questionnaires on their early caregiving experiences, mental health, and early parent–child relations. To be included, respondents must have provided primary caregiving assistance (i.e., bathing, feeding, etc.) for a parent or adult relative when the caregiver was under 21 years old. Results showed that the sample reported more positive mental health than negative mental health, though 42% had high depressive scores on the total CES-D. Individuals who reported fathers as too protective reported less current positive mental health. Early caregiving is not associated with poor mental health in adulthood for many young caregivers. However, some individuals do appear at risk of depression in adulthood.

Manual material handling follows in nearly all working surroundings, although labors in agriculture fields, construction sites, restaurants, and hotels area are likely to be working with heavy jobs in terms of weight, currencies, Manual handling of the heavy load may turn in to a growing disorder due to the steady or increasing drop of the musculoskeletal system due to constant material lifting / handling events. It may also reason to acute trauma like bone fractures or skin cuts from accidents. One must begin to identify the correct process of methods to deal with each material to solve the problem. Adherence to standard procedures for long-term resolution of the long-term effects caused by incorrect manual material handling, is addressed through specially designed workouts after or before work and, ultimately, also automated technology Or involved in providing assistance through manual tools that assist the user in dealing with content. The proposed task is to conduct studies to identify process issues and future results and to provide solutions to the industry by means of analysis and following standard procedures. This paper mainly deals with issues directly or indirectly in many aspects and discusses solutions implemented to deal with manual content in various industry. The study concluded with a novel automation solution for rapid analysis of effects due to manual analysis that could take years to identify.

The developing mammalian fetus is thought to be highly sensitive to ionizing radiation. However, dose, dose-rate relationships are not well established, especially the long term effects of protracted, low-dose exposure. A previous report (1) has indicated that bred beagle bitches exposed to daily doses of 5 to 35 R 60Co gamma rays throughout gestation can produce viable, seemingly normal offspring. Puppies irradiated in utero are distinguishable from controls only by their smaller size, dental abnormalities, and, in adulthood, by their inability to bear young.We report here our preliminary microscopic evaluation of ovarian pathology in young pups continuously irradiated throughout gestation at daily (22 h/day) dose rates of either 0.4, 1.0, 2.5, or 5.0 R/day of gamma rays from an attenuated 60Co source. Pups from non-irradiated bitches served as controls. Experimental animals were evaluated clinically and hematologically (control + 5.0 R/day pups) at regular intervals.

Previous short term toxicological studies of one to two weeks duration have demonstrated that MDL 19,660 (5-(4-chlorophenyl)-2,4-dihydro-2,4-dimethyl-3Hl, 2,4-triazole-3-thione), an antidepressant drug, causes a dose-related thrombocytopenia in dogs. Platelet counts started to decline after two days of dosing with 30 mg/kg/day and continued to decrease to their lowest levels by 5-7 days. The loss in platelets was primarily of the small discoid subpopulation. In vitro studies have also indicated that MDL 19,660: does not spontaneously aggregate canine platelets and has moderate antiaggregating properties by inhibiting ADP-induced aggregation. The objectives of the present investigation of MDL 19,660 were to evaluate ultrastructurally long term effects on platelet internal architecture and changes in subpopulations of platelets and megakaryocytes.Nine male and nine female beagle dogs were divided equally into three groups and were administered orally 0, 15, or 30 mg/kg/day of MDL 19,660 for three months. Compared to a control platelet range of 353,000- 452,000/μl, a doserelated thrombocytopenia reached a maximum severity of an average of 135,000/μl for the 15 mg/kg/day dogs after two weeks and 81,000/μl for the 30 mg/kg/day dogs after one week.
