The aim of this study was to evaluate the glucogenic property of glycerol supplementation in the dairy cow’s diet. Sixty primiparous cows (control, n=30, and glycerol supplemented, n=30) were used to measure milk yield and components, blood hormone and metabolite profiles, and body condition score. Feed intake and apparent total-tract digestibility were also measured using 10 primiparous cows (control, n=5, and glycerol supplemented, n=5). Dry glycerol was top dressed at 250 g/day/cow from parturition to 21 days postpartum. Average feed intake, milk yield and components were not affected by glycerol supplementation. Apparent total–tract digestibility of organic matter and neutral detergent fibre were not influenced by dry glycerol supplementation, but lipid digestibility was greater (<em>p</em>=0.01) in cows fed glycerol. The serum concentration of glucose and insulin tended to be higher in dry glycerol-supplemented cows (<em>p</em>=0.1; <em>p</em>=0.06, respectively). While, serum concentrations of nonesterified fatty acids and <em>β</em>-hydroxybutyrate were not affected. Supplemented cows had lower body condition loss during weeks 1 to 5 after calving (<em>p=</em>0.09). The glucogenic effect of glycerol did not affect milk yield during the first 3 weeks of lactation. However, daily milk yield during the 13 weeks recording period was higher in the glycerol-supplemented cows (28.5 <em>vs</em>. 30.3 kg, <em>p&lt;</em>0.001). Percentages of cows cycling at the planned breeding date was greater (<em>p</em>=0.01) for cows fed dry glycerol. The results demonstrated that feeding dry glycerol as a glucogenic supply could be useful in saving body reserves and improving energy balance of primiparous Holstein dairy cows during the early postpartum period.

This study investigated the efficacy of intrauterine infusion of a chitosan solution (CHT) on uterine recovery in early postpartum dairy cows with or without endometritis, and their subsequent reproductive performance. In Experiment 1, cows with endometritis at 3 weeks postpartum were administered CHT (n = 5) and prostaglandin F2α (PGF2α) (n = 4). Untreated cows (n = 7) served as the control group. In Experiment 2, 18 cows with a normally recovered uterus at the fresh cow check (mean, 35 days postpartum) were assigned to the CHT (n = 10) and control (n = 8) groups, and intrauterine infusion was conducted in the CHT group. Overall, in Experiment 1, the percentage of polymorphonuclear leukocytes significantly declined in the CHT group (32.3 ± 10.2 to 5.5 ± 2.4, p < 0.05) from week 3 to week 5, but no decline occurred in the PGF2α and control groups. In Experiment 2, the CHT and control groups showed no significant differences in reproductive parameters, suggesting the absence of adverse effects of CHT on fertility. These results suggest that intrauterine infusion of CHT in the early postpartum period effectively accelerates uterine recovery from endometritis and might be a suitable replacement for PGF2α administration.

Although it has become increasingly clear that fertility in modern dairy cattle is declining in association with increased milk yields, the underlying mechanism is poorly understood. The first ovulation post partum is delayed in dairy cows undergoing selection for genetic merit for milk yield in association with lower circulating insulin concentrations. The aim of this study was to investigate whether feeding a diet to increase circulating insulin concentrations can overcome this delay in the first ovulation post partum. The experiment was a 2 x 2 factorial design (n = 10 per group) involving diet and genetic merit for milk yield. The dietary treatment started on the day of calving and lasted for 50 days. Plasma samples were collected each day and ovarian ultra-sonography was performed three times a week during the experimental feeding period. Milk yield was recorded each day, and body weight and body condition score were determined each week. Milk samples were collected three times a week from day 50 to day 105 post partum, and reproductive performance data were recorded for all the cows as part of the routine farm practice. The dietary treatment induced significant differences in plasma insulin concentrations in both high and low genetic merit cows. Although high genetic merit cows produced more milk, lost more body weight and had lower body condition scores during the experiment, no significant effect of diet was observed on these measurements. The high insulin inducing diet increased the proportion of cows ovulating within 50 days of calving and reduced the intervals from calving to first ovulation, and tended to reduce the intervals from calving to first service and to conception. These fertility parameters were also more favourable in low than in high genetic merit cows, but no interaction between diet and genetic merit was observed for any of these parameters. Genetic merit, but not diet, also affected the number of services required per conception and the conception rate. In conclusion, these results have confirmed that genetic selection for high milk yield is associated with a decrease in reproductive performance in dairy cows. More importantly, this study has demonstrated that it is possible to alleviate this problem by nutritional manipulation.
