Evaluation of resistance of Vigna unguiculata L. landraces, used as green beans, to Aphis craccivora Koch was tested. Eleven landraces and standard genotypes were used in a free choice protocol: BR 17-Gurguéia, VITA 7 (susceptible), BRS Guariba and TVu 408P2 (resistant). A completely randomized blocks design was used with six replicates. Seedlings were infested with five six-day-old adult insects. The number of alive adults after 48 h and of alive nymphs after 96 h was evaluated. The results evidenced that there are high genetic diversity among the landraces, the antibiosis as the main mechanism of resistance. By means of multivariate analysis we suggest crosses among the commercial cultivar BRS Guariba and the most dissimilar varieties CCE-088, CCE-075, CCE-013 and CCE-062 to obtain superior hybrids.

Cowpea is a multiple-purpose drought-tolerant leguminous pulse crop grown in several dry tropical areas. Its domestication center is thought to be East or West Africa, where a high level of genetic diversity is apparently still found. However, detailed genetic information is lacking in many African countries, limiting the success of breeding programs. In this work, we assessed the genetic variation and gene flow in 59 Vigna unguiculata (cowpea) accessions from 10 landraces spanning across six agro-ecological zones of Mozambique, based on nuclear microsatellite markers. The results revealed the existence of high genetic diversity between the landraces, even in comparison to other world regions. Four genetic groups were found, with no specific geographic pattern, suggesting the presence of gene flow between landraces. In comparison, the two commercial varieties had lower values of genetic diversity, although still close to the ones found in local landraces. The high genetic diversity found in Mozambique sustains the importance of local genetic resources and farm protection to enhance genetic diversity in modern varieties of cowpea worldwide.

Abstract. Widyawan MB, Hasanah A, Taryono, Alam T, Sayekti RS, Pramana AAC, Wulandari RA. 2020. Multivariate analysis unravels genetic diversity and relationship between agronomic traits, protein, and dietary fiber in yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.). Biodiversitas 21: 5662-5671. Yardlong bean (Vigna unguiculata subsp. sesquipedalis Verdc.) is a vegetable legume that possesses the highest amount of protein and total dietary fiber (TDF) content among other vegetables. However, protein and TDF content diversity are found among yardlong bean genotypes. Genetic improvement to produce cultivars with high yield and improved nutritional content is required. This study aimed to analyze genetic diversity and identify the relationship between important agronomic traits from 21 yardlong bean genotypes assessed by multivariate statistical analysis. The experiment was arranged in alpha design with three replicates, each consist of six blocks. Six pod-related important agronomic traits, protein, and TDF content were observed in this study. Total pod weight, pod weight, and pod length were found to be negatively correlated with protein and TDF content based on correlation and principal component analysis (PCA). Although negatively correlated, several variables possess a large and positive direct effect on protein and TDF content, such as the number of pod, pod weight, and days to flower. D2 analysis and Tocher’s method classify genotypes used in this study into five clusters with diverse traits value. Information from this study laid a foundation for the improvement of important agronomic traits as well as nutritional content in yardlong bean.

Thirty three seed cow genotypes (Vigna unguiculata L. WALP) collected from different geographical region were assessed for their genetic divergence based on ten characters during kharif-2020 at Post Graduate Farm, MPKV, Rahuri. The genotypes were grouped into six clusters which indicating high genetic diversity among them. It indicated that genetic diversity and geographic diversity are not related. The present study revealed that days to 50% flowering contributed maximum (34.09%) for divergence followed by plant height (32.58%), number of pods per plant (10.98%) and test weight (8.71%). Based on inter-cluster distance, intra-cluster distance and per se performance the genotypes viz., CP-04, CP-10, CP-10R, PCP-1411, CP-37, CP-10R,CP-39 and Phule Sonali are suggested to utilize for future breeding programme.

Cryptosporidiumfelis is an important cause of feline and human cryptosporidiosis. However, the transmission of this pathogen between humans and cats remains controversial, partially due to a lack of genetic characterization of isolates from cats. The present study was conducted to examine the genetic diversity of C. felis in cats in China and to assess their potential zoonotic transmission. A newly developed subtyping tool based on a sequence analysis of the 60-kDa glycoprotein (gp60) gene was employed to identify the subtypes of 30 cat-derived C. felis isolates from Guangdong and Shanghai. Altogether, 20 C. felis isolates were successfully subtyped. The results of the sequence alignment showed a high genetic diversity, with 13 novel subtypes and 2 known subtypes of the XIXa subtype family being identified. The known subtypes were previously detected in humans, while some of the subtypes formed well-supported subclusters with human-derived subtypes from other countries in a phylogenetic analysis of the gp60 sequences. The results of this study confirmed the high genetic diversity of the XIXa subtype family of C. felis. The common occurrence of this subtype family in both humans and cats suggests that there could be cross-species transmission of C. felis.
