Guidelines for DNA barcoding of coralline algae, focusing on Lithophylloideae (Corallinales) from Brazil

AbstractMultiple-marker (COI-5P, UPA,psbA andrbcL-3P) and two algorithmic approaches [automatic barcode gap discovery (ABGD) and Poisson tree process (PTP)] were used for species delimitation of Lithophylloideae in Brazil. The integrative approach was mostly congruent between markers and algorithmic methods of species delimitation, suggesting the occurrence of 24 species. Based on morphology and molecular data,Amphiroa rigida,Amphiroa vanbosseae,Lithophyllum atlanticum,Lithophyllum kaiseri,Lithophyllum margaritae,Titanoderma pustulatum,Titanoderma prototypumandPaulsilvella huveorum, which were previously reported for Brazil, are confirmed in this work. Six new species are distinguished by both molecular and morphological traits, and they are provisionally named asAmphiroasp. 1,Amphiroasp. 2,Amphiroasp. 3,Lithophyllumsp. 1,Lithophyllumsp. 2 andLithophyllumsp. 3. Another 10 species are cryptic and cannot be distinguished based on traditionally used morphological traits. These includeAmphiroasp. 4,Lithophyllumsp. 4, three species that are morphologically named underAmphiroa beauvoisii, and six that share the morphology described forAmphiroa fragilissima. All four markers used were useful for species delimitation. However, a combination of practical aspects and levels of intra- and interspecific divergence values led us to propose the use ofrbcL-3P as a standard DNA barcode marker for the Corallinales.

The genus Pseudostegana (Okada, 1978) currently contains thirty-nine described species. A number of Pseudostegana were collected from the fieldwork in southwestern China from 2010 to 2017. Eleven new species were discovered and are described from southwestern China: Pseudostegana alpina Zhang & Chen, sp. nov.; Pseudostegana amnicola Zhang & Chen, sp. nov.; Pseudostegana amoena Zhang & Chen, sp. nov.; Pseudostegana mailangang Zhang & Chen, sp. nov.; Pseudostegana meiduo Zhang & Chen, sp. nov.; Pseudostegana meiji Zhang & Chen, sp. nov.; Pseudostegana mystica Zhang & Chen, sp. nov.; Pseudostegana stictiptrata Zhang & Chen, sp. nov.; Pseudostegana stigmatptera Zhang & Chen, sp. nov.; Pseudostegana ximalaya Zhang & Chen, sp. nov. and Pseudostegana zhuoma Zhang & Chen, sp. nov. A key to all Chinese Pseudostegana species based on morphological characters is provided. Two mitochondrial loci (COI and ND2) and one nuclear locus (28S rRNA) were sequenced for the Pseudostegana specimens, and Bayesian and RAxML concatenated analyses were run. Molecular species delimitation is performed using the distance-based automatic barcode gap discovery (ABGD) method. Molecular data support the morphological characteristics observed among these Chinese species and confirm the new species as being distinctly different.

The Neotropical water scavenger beetle genus Chasmogenus Sharp, 1882 is a diverse lineage with 34 described species. Here, three new species from the southwestern margin of the Guiana Shield are described using an integrative approach combining adult morphology and molecular data: Chasmogenus clinatus sp. n. (Brazil: Amazonas, Venezuela: Amazonas), C. gironae sp. n. (Venezuela: Amazonas), and C. inpa sp. n. (Brazil: Amazonas). Examination of the male genitalia is essential for confident identifications and thus unassociated females may prove extremely difficult to identify. New distributional records in Brazil for C. acuminatus Smith & Short, 2020 are provided as well as habitus images of newly described species and an updated key to the species from Venezuela, Suriname, Guyana, French Guiana and northern Brazil.

AbstractThe new species Ramalina fleigiae from Brazil is described growing on rocks in riverbeds in high altitude grasslands of southern Brazil. It grows in areas with constant water flow, sometimes almost immersed, and always in exposed habitats. Through an integrative approach, the detailed description of R. fleigiae includes morphological, anatomical, ecological, chemical and molecular data. Ribosomal DNA-based phylogenies suggest that R. fleigiae is more closely related to a species that shares its habitat preference (R. laevigata) than to the morphologically and chemically similar R. exiguella and R. gracilis. Ramalina fleigiae and R. laevigata can be distinguished by thallus morphology (irregularly flat branches in R. fleigiae vs. flat to canaliculate in R. laevigata) and pattern of chondroid tissue, as genetic distances between them are compatible with the interspecific range. It is possible that many species of Ramalina still remain hidden within the morphological or chemical variation of currently accepted species. Combining ecological, anatomical and molecular data will improve our future understanding of this genus.

Two new species of the genus Analges (Astigmata: Analgoidea) are described from two species of accentors (Passeriformes: Prunellidae): Analges himalayanus sp. nov. from the Altai Accentor P. himalayana (Blyth), and A. slovakiensis sp. nov. from the Alpine Accentor Prunella collaris (Scopoli). Both species are closely related to A. bidentatus Giebel, 1871 described from the Dunnock Prunella modularis (Linnaeus). We extended the standard morphological descriptions of feather mites by species delimitation analyses carried out on DNA barcode sequences.

The life cycle of Parasitengona includes major morphological changes precluding an instar association based only on the morphology. This makes rearing and/or molecular data necessary to associate the heteromorphic instars. Most of the described species are known from either post larval instars or larva. Following a previous study on Palearctic Erythraeidae, in the present study the instar association was made through an integrative approach including rearing trials and molecular analysis of the cytochrome oxidase I (COI) gene with the Bayesian Generalized Mixed Yule Coalescent (bGMYC) algorithm for species delimitation. Two new cave dwelling Erythraeidae (Trombidiformes: Parasitengona) species are described Lasioerythraeus jessicae sp. nov. and Leptus sidorchukae sp. nov. including all active instars. Additionally, a complete description of the previously unknown adults of Charletonia rocciai Treat & Flechtmann, 1979 is provided with notes on the larva and deutonymph. We also demonstrate experimentally that Ch. rocciai larvae are not attached to the same individual host during the entire feeding stage. We discuss the presence of troglomorphisms in Le. sidorchukae sp. nov.; and the distribution of the species.

Caiophora is a taxonomically difficult, nearly exclusively Andean genus of the largely South American family Loasaceae subfam. Loasoideae. Elevational distribution and flower morphology argue for a relatively basal position of loasoid Caiophora in the genus. Caiophora has not been revised since 1900, and details of their morphology, distribution and species delimitation are incompletely understood. The Caiophora pterosperma-group clearly belongs to Caiophora based on habit, fruit morphology, karyology and molecular data, but is florally similar to members of the closely allied genera Loasa and Scyphanthus. The Peruvian members of the Caiophora pterosperma-group are here revised. Three species are recognized: C. pterosperma, endemic to the departments of Junín and Pasco (including C. smithii, C. serropetala and C. pavonii), C. stenocarpa from the departments Cuzco and Huancavelica, and the new species Caiophora dederichiorum, endemic to the department of Ancash. All three species occur at low elevations for the genus (down to 2200 m a.s.l.) and in seasonally dry habitats, unlike most representatives of the genus (usually found in mesic habitats at higher elevations). A key to the Peruvian members of the group, diagnoses, drawings and photographs are provided for all species recognized.

We diagnose and describe a new freshwater mussel species of the genus Inversidens, I. rentianensissp. nov. from Jiangxi Province, China based on morphological characters and molecular data. This paper includes a morphological description and photograph of the holotype, and partial sequences of mitochondrial COI as DNA barcode data.

Abstract Lactifluus (Pers.) Roussel is an ectomycorrhizal genus that was recently recognized to be distinct from the genus Lactarius. To date, 216 Lactifluus species have been reported worldwide. Misidentification of Lactifluus species is common because of intraspecific morphological variation, cryptic diversity, and the limited number of taxonomic keys available. Molecular data are indispensable for species delimitation; a multilocus phylogenetic analysis showed that most Asian Lactifluus species are not conspecific with morphologically similar species present on other continents. In particular, Korea has misused European and North American Lactifluus names. In this study, we evaluated the taxonomy of Lactifluus in Korea using both morphological and multilocus molecular (ITS, nrLSU, rpb1, and rpb2) data. We examined 199 Lactifluus specimens collected between 1980 and 2016, and a total of 24 species across the four Lactifluus subgenera were identified. All Korean species are distinct and clearly separated from European and North American species. Five taxa corresponded to previously described species from Asia and the remaining 19 taxa are confirmed as new species. Herein, we provide keys to the Korean Lactifluus species within their subgenera, molecular phylogenies, a summary of diversity, and detailed description of the new species.

A large scale semi-quantitative biodiversity assessment was conducted in white oak woodlands in areas included in the Spanish Network of National Parks, as part of a project aimed at revealing biogeographic patterns and identify biodiversity drivers. The semi-quantitative COBRA sampling protocol was conducted in sixteen 1-ha plots across six national parks using a nested design. All adult specimens were identified to species level based on morphology. Uncertain delimitations and identifications due to either limited information of diagnostic characters or conflicting taxonomy were further investigated using DNA barcode information.We identified 376 species belonging to 190 genera in 39 families, from the 8,521 adults found amongst the 20,539 collected specimens. Faunistic results include the discovery of 7 new species to the Iberian Peninsula, 3 new species to Spain and 11 putative new species to science. As largely expected by environmental features, the southern parks showed a higher proportion of Iberian and Mediterranean species than the northern parks, where the Palearctic elements were largely dominant. The analysis of approximately 3,200 DNA barcodes generated in the present study, corroborated and provided finer resolution to the morphologically based delimitation and identification of specimens in some taxonomically challenging families. Specifically, molecular data confirmed putative new species with diagnosable morphology, identified overlooked lineages that may constitute new species, confirmed assignment of specimens of unknown sexes to species and identified cases of misidentifications and phenotypic polymorphisms.
