Abstract This work is aimed to evaluate in vitro regeneration potential of seven commercial soybean varieties Bohemians, Cardiff, Gallec, Merlin, Moravians, Naya and Silensia (Glycine max L.) cultivated in Central Europe. Our results showed the half-seeds could be effectively used as an explant source for all tested cultivars. The regeneration was initiated on the media containing growth regulators 1.67 mg.l-1 BAP and 0.25 mg.l-1 GA3. Within the first five days culture, green chlorophyll-containing explants were observed with frequency from 18.3% to 55.9%. Two weeks later, the explants responded by production of calli with the efficiency up to 83.0%. First shoots appeared after 2–3 weeks of subculture on the media. The soybean regeneration showed to be genotype-dependent with variable efficiencies from 5.7% (cv. Naya) to 37.7% (cv. Gallec). The cultivars Cardiff, Merlin and Gallec appear to be the most promising candidates for further biotechnological use. Application of antioxidants such as L-cysteine, dithiothreitol and sodium thiosulfate does not have effect on the explant regeneration for the first five days.

In vitro: regeneration of Chenopodium quinoa Willd. was achieved from cotyledonary nodes explants. In this study, used 6-Benzylaminopurine (BAP) and α-Naphthalene Acetic Acid (NAA) of plant growth regulators with different concentrations individually as well as in combination and used different concentrations of sugar (sucrose) with different concentrations. For was rooting, used half strength (½MS), full-strength MS and ½ MS supplemented with 0.2 mg/l of NAA. The results mentioned, explant responding (%) to multiplication was about 73% for all BAP treatments compared with control and average numbers of shoot increased with increased BAP concentration except 5 mg/l of BAP. The highest explant responding (%) was in media supplemented BAP without NAA compared other treatments noted that the media with combination of BAP and NAA gives formation of callus in bases of the plantlets. Also, the result inducted the combinations between (BAP–NAA) was highly significantly (P≤ 0.001) and less effective on number of shoots where the highest number of shoot was 3.40 in media with 3 mg/l BAP compared other treatments. The highest of explant responding 93.33% was in media supplement with 10 g/l sucrose and (10 g/l sucrose + 3 mg/l BAP), but sucrose level for good greening and developed shoots (4 shoots) was in medium supplement with 10 g/l sucrose. The shoots rooted well on half-strength MS medium with 60% percentage of root. The rooted shoots were acclimatized and transferred to green house to follow their development.

One of the challenges related to propagation of Arenga pinnata is its lengthy period of seed dormancy. In this study, in vitro regeneration was carried out to determine the effect of hormonal treatment on the embryo explant of Arenga pinnata. Embryos were surface sterilized and cultured into different media supplemented with various hormones concentrations and combinations. Each treatment contained of Kinetin (KN) hormone (1.0, 2.0, and 3.0 mg/l) and in combination with indole-3-acetic acid (IAA) of 0.1, 0.2, 0.3 mg/l. The height of plumule and length of radical was observed and recorded. Treatment 8 (3 mg/ml KN + 0.1 mg/ml IAA) showed 59.09% in plumule height increment while treatment 4 (1 mg/ml KN + 0.3 mg/ml IAA) showed the highest radical increments with 93.62%. The knowledge gained in this study consequently helps us to better understand the role of KN and IAA in the in vitro regeneration protocol. Since in vitro method able to produce higher number of in vitro seedlings at one time, it is important to establish the in vitro regeneration protocol for this plant.

O objetivo do experimento consistiu na simulação in vitro da soja transgênica tolerante ao glyphosate, através do cultivo de explantes em meios de cultura contendo aminoácidos aromáticos. As avaliações basearam-se nos efeitos do glyphosate sobre sementes oriundas de explantes de soja (Glycine max (L.) Merr.) cv. UFV-16. Para tanto, explantes de soja foram cultivados em meios de cultura líquidos com pH em torno de 5,0. Cada explante constou de um legume completamente expandido contendo duas sementes de aproximadamente 100 mg, conectada a um segmento de caule de 45 mm de comprimento. Os tratamentos testados foram: A = glutamina (Gln); B = Gln + fenilalanina (Phe) + tirosina (Tyr) + triptofano (Trp); C = Gln + glyphosate; D = Gln + Phe + Tyr + Trp + glyphosate. O experimento foi conduzido sob irradiância de 80 mmol-2s-1 a 25oC por 204 horas. Nos tratamentos que receberam aminoácidos aromáticos e glutamina, o herbicida não afetou as massas fresca e seca das sementes, como também, não afetou seus constituintes bioquímicos (proteínas, óleo, ácidos graxos, carboidratos e clorofilas). Portanto, a suplementação exógena de aminoácidos aromáticos suprime os efeitos fitotóxicos do glyphosate sobre explantes de soja, permitindo estudos sobre o seu modo de ação e metabolismo nas sementes, uma vez que os explantes se comportaram analogamente à soja transgênica não suscetível ao herbicida.

The present experimental study was aimed to overcome the traditional methods of propagation that limit the number of propagules by in-vitro regeneration through nodal explants of Dendrocalamus hamiltonii with a comparative study of growth regulators during the shooting and rooting process. Dendrocalamus hamiltonii is distributed from the Himalayas (Nepal) to the northern part of Burma. Collection of explants was done from different selected sites of CPTs. There was the use of HgCl2 and Ca (OCl)2 as sterilizing agents in different concentrations and its effect was visualized during the sprouting stage. Culm explants were cultured in a bottle containing White media (Wm) supplemented with BA and Kinetin for sprouting and IAA, IBA, NAA for rooting. There is also the use of IAA+IBA+NAA in combined form as a supplementary solution 0.1% HgCl2 treatment for 20-minute results into77.80% aseptic buds and 72% bud -break. Among the used growth-hormones, BA with concentration 0.25mg/l and 0.50mg/l respectively were appropriate for shoot-multiplication rate, 4.01±0.3 and 4.3±0.4 were ideal observation incorporation with BA (1.00mg/l) and BA (1.50mg/l) respectively. Maximum sprouting rate14.77±3.37with application of BA (2.00mg/l) and maximum shoot length4.3±0.4 is observed at BA (1.50mg/l). The applications of rooting hormone IAA+IBA+NAA in the concentration of 1.0 mg/l results in 72.5±0.3(rooting) and 11.1±0.3 (av. No. of the root).

Foram desenvolvidos dois experimentos com objetivo de avaliar o potencial de preparados de cavalinha (Equisetum sp.) na síntese de metabólitos de defesa em cotilédones de soja (Glycinemax L.) e o efeito sobre o crescimento de Rhizoctonia solani, in vitro. O delineamento experimental utilizado para os experimentos foi inteiramente casualizado em esquema fatorial 3x5 (formas de extração x concentrações), com quatro repetições. As formas de extração foram extrato alcoólico, infusão e maceração, nas concentrações de zero; 1; 10, 20 e 40%. No primeiro experimento foi avaliada a indução de compostos de defesa vegetal em cotilédones de soja em resposta aos derivados a base de cavalinha, sendo quantificada a atividade da enzima fenilalanina amônia-liase (FAL), via espectofotometria, a fitoalexina gliceolina, e o teor de fenóis totais. No segundo experimento, in vitro, a unidade experimental foi uma placa de Petri, sendo os preparados de cavalinha incorporados ao meio BDA (Batata-dextrose e Agar) e avaliado o crescimento micelial de R. Solani. Os preparados de extrato alcoólico, infusão e maceração de cavalinha apresentaram capacidade de indução das fitoalexinas gliceolinas em cotilédones de soja, bem como, ativaram o metabolismo de compostos fenólicos. Entre os preparados, o extrato alcoólico e a maceração, se sobressaem sobre a infusão. Os preparados de extrato alcoólico, infusão e maceração de cavalinha em todas as suas concentrações inibem o crescimento do fungo R. solani, in vitro.

Abstract The in vitro regeneration potential of shoot apical tips (2 to 3 mm in length), meristems (0.3 to 0.5 mm in length), and nodal segments (4 to 7 mm long with an axillary bud) of diploid (2n = 2x = 20) and triploid (2n = 3x = 30) cytotypes of Arachis pintoi was evaluated. Explants were cultured on MS medium supplemented with different concentrations and combinations of naphthaleneacetic acid (NAA) and benzyladenine (BA). In one experiment the effect of gibberellic acid was tested. The cultures were done in liquid and solid media. Plant regeneration can be readily achieved from all explants in one step of 30 d culture on MS + 0.01 mg/L each of NAA and BA or two steps consisting of 1) shoots regeneration through culture of explants on MS + 0.01 mg/L each of NAA and BA, and 2) induction of rooting in regenerated shoots by reculture on MS + 0.01 mg/L NAA. The plantlets were successfully transferred to pots in a greenhouse.

<p>Shiny dome-like structures measuring less than 1mm in length were excised aseptically from shoot tip buds of infected of two cassava (<em>Manihot esculenta</em> Crantz) local cultivars (Boma and Mpelo Nlongi) and cultivated <em>in vitro</em> in two types of media with different combination of growth hormone: Murashige and Skoog supplemented of sucrose (20 g/l), Naphtalenacetic acid (NAA, 10 μM), Ben-zylaminopurine (BAP, 0.66 μM) as well as Gibberellic acid (GA3, 0.1 μM) with 80 mg/l of Adenine sulphate and MS-free growth regulators. After four weeks, data were scored: 29.5% responding explant with callus formation and 20.5% responding explants to shoot development in the medium with growth regulators for the cultivar Boma whereas the cultivar Mpelo-Nlongi presented 5.7% and 25.7% respectively of callus formation and shoot development. The cultivar Boma presented a tendency more pronounced for the callus formation rather than with the shoot development contrary to the cultivar Mpelo-Nlongi. In regards of this experiment, it was shown that the media composition and genotype are essential factors, which influence in vitro growth, mainly the shoot development, in the culture of meristems for cassava local accessions.</p>
