Tanaman budi daya merupakan tanaman yang sering diserang oleh cendawan pathogen, sehingga mengakibatkan penurunan populasi dan produksi tanaman. Pengendalian hayati dengan cendawan antagonis merupakan salah satu metode yang paling efektif dan lebih ramah lingkungan dalam menekan pertumbuhan patogen tanaman. Penelitian ini bertujuan untuk menguji aktivitas antagonis cendawan isolat lokal Riau terhadap beberapa cendawan patogen pada tanaman budi daya. Uji antagonis dilakukan secara in vitro dengan metode dual culture menggunakan lima belas cendawan isolat lokal Riau terhadap Fusarium oxysporum f.sp. lycopersici, Ganoderma philippii, G. boninense, Rigidoporus microporus dan Colletotrichum sansevieria. Hasil penelitian menunjukkan bahwa Trichoderma sp. PNE 4 memiliki aktivitas antagonis tertinggi dan isolat FER C1 serta isolat LLB07 hanya memiliki aktivitas antagonis yang tinggi dalam menekan pertumbuhan cendawan patogen. Trichoderma sp. PNE 4 mampu menghambat pertumbuhan miselium F. oxysporum sebesar 85,30%, G. Philippii (100%), G. boninense (100%), dan C. sansevieria (100%). Isolat FER C1 hanya menghambat R. Microporus (50,39%) dan isolat LLB07 menghambat G. philippii (52,20%). Trichoderma sp. PNE 4 merupakan cendawan uji yang terpilih sebagai cendawan antagonis, karena memiliki kemampuan daya hambat  >70%.Abstract Cultivated plants are often attacked by pathogenic fungi resulting in a decline of population and crop production. Biocontrol with antagonistic fungi is one of the most effective and environmentally friendly methods in suppressing the growth of plant pathogens. This study aims to examine the antagonistic activity of local isolates fungi Riau against some pathogenic fungi on cultivated plants. The antagonistic test was performed in vitro by dual culture method using fifteen local isolates fungal Riau against Fusarium oxysporum f.sp. lycopersici, Ganoderma philippii, G. boninense, Rigidoporus microporus and Colletotrichum sansevieria. The results showed that Trichoderma sp. PNE 4 isolate exhibited highest activites and  FER C1 and LLB07 isolates exhibited high activities suppressed the growth of the fungal pathogen. Trichoderma sp. PNE 4 isolate inhibited mycelial growth F. oxysporum (85.30%), G. philippii (100%), G. boninense (100%) and C. sansevieria (100%). FER C1 isolate only inhibited R. microporus (50.39%), and LLB07 isolate inhibited G. philippii (52.20%). Trichoderma sp. PNE 4 isolate is test isolates as fungal antagonistic.

<p>The Potential of Biological Agents to Inhibit Growth of Phytium sp. In Vitro. The study aimed at testing the potential of some antagonistic fungi isolated from durian seedlings media to inhibit growth of Phytium sp. Research was done at the Central Laboratory of Tropical Fruit Research Solok in July-September 2009 by using a complete randomized design with 5 treatments and 4 replications. Tests was conducted by dual culture method on PDA. The results showed that Gliocladium sp., Trichoderma sp.a, Trichoderma sp.b, Aspergilus sp., and Penicillium sp. can inhibit growth of Phytium sp., with growth inhibition of 50, 49.5, 47, 48, and 38.3% respectively. Inhibition mecanism of Gliocladium sp., and Trichoderma sp. were competition, antibiosis, lisis, and parasitism, while Penicillium sp. was antibiosis. Gliocladium sp., Trichoderma sp.a, Trichoderma sp.b, Aspergilus sp., and Penicillium sp. can be used as biological agents to control pathogenic fungi Phytium sp.</p><p> </p><p><strong>Abstrak</strong></p><p>Penelitian bertujuan untuk menguji potensi beberapa cendawan antagonis hasil isolasi dari media pembibitan durian dalam menghambat pertumbuhan Phytium sp. Penelitian dilakukan di Laboratorium Balai Penelitian Tanaman Buah Tropika Solok pada bulan Juli-September 2010. Penelitian disusun dalam rancangan acak lengkap dengan 5 perlakuan dan 4 ulangan. Pengujian daya antagonis cendawan dilakukan dengan metode dual culture yang diinokulasikan pada media PDA. Hasil penelitian menunjukkan bahwa Gliocladium sp., Trichoderma sp.a, Trichoderma sp.b., Aspergilus sp., dan Penicillium sp. dapat menghambat pertumbuhan Phytium sp. secara in vitro, dengan daya hambat masing-masing 50; 49,5; 47; 48; dan 38,3% secara berurutan. Mekanisme antagonis Gliocladium sp. dan Trichoderma sp. adalah kompetisi, antibiosis, lisis, dan parasitisme, sedangkan Penicillium sp. hanya bersifat antibiosis. Gliocladium sp., Trichoderma sp., Aspergilus sp., dan Penicillium sp. dapat digunakan sebagai agen hayati untuk mengendalikan cendawan patogen Phytium sp.</p>

Butt rot disease, caused by Thielaviopsis paradoxa (De Seynes) Hohn., is one of the major diseases in pineapple cultivation in Malaysia. The objectives of this study were to evaluate the antifungal effect of antagonist bacteria against T. paradoxa, a causal agent of butt rot disease, and to observe the mechanism of antifungal activity of tested antagonist bacteria microscopically. In this study, in vitro antifungal potential of 5 antagonist bacteria, namely B1, B2, B3, B4, and B5, were isolated from infected and non-infected soil samples and evaluated using dual culture method against T. paradoxa. The mechanisms of antifungal activities of antagonist bacteria against the pathogen were microscopically observed. All of the bacteria showed inhibitory effects against the pathogenic fungi. B1 bacteria showed the highest inhibitory potential, with 73 % inhibition, followed by B2, B3, B4, and B5, with 71, 57, 56, and 48 % of inhibition compared to control, respectively. The results also showed that B2, B3, and B4 bacteria exhibited positive inhibition towards the pathogen, with more than 50 % percentage inhibition. The development of a new product for use as a biocontrol agent, used as an additional control or used in combination with existing ones, may reduce dependency on chemical control and increase antagonistic activity efficiency.

This study aims to determine the ability of two endophytic fungi in Gondang Manis jamaican apple to inhibit the development of indigenous pathogenic fungi on these plants. The isolates of Trichoderma sp and Metarhizium sp were obtained from the exploration of endophytic fungi on the tissue of the Gondang Manis jamaican apple plant, while Fusarium sp was the result of exploration on the rhizosphere soil of the plant. Tests were carried out in vitro with a dual culture method using a completely randomized design (CRD) with FT (Fusarium - Trichoderma) and FM (Fusarium - Metarhizium) treatments, each with 5 replications. Determination of the resistance level based on PIRG and Bell Rating. Analysis using ANOVA p (˂0.05) and LSD test (p˃ 0.05). The results showed that Trichoderma sp has a high inhibiting ability (PIRG = 70%; Bell Rating = 2) against Fusarium sp, while Metarhizium sp is considered low (PIRG = 20.5%; Bell rating = 4). This shows that Trichoderma sp. Isolate has a better ability than Metarhizium sp in inhibiting the development of Fusarium sp. The mechanism of inhibition of the pathogen Fusarium sp by Trichoderma sp is to seize nutrients from pathogens   Key word : Inhibition, Fusarium sp, Metarhizium sp, Trichoderma sp.

Microorganisms for biological control are capable of producing active compounds that inhibit the development of phytopathogens, constituting a promising tool toob tain active principles that could replace synthetic pesticides. This study evaluatedtheability of severalpotentialbiocontrol microorganismsto produce active extracellular metabolites. In vitro antagonistic capability of 50 bacterial isolates from rhizospheric soils of "criolla" potato (Solanum phureja) was tested through dual culture in this plant with different plant pathogenic fungi and bacteria. Isolates that showed significantly higher antagonistic activity were fermented in liquid media and crude extracts from the supernatants had their biological activities assessed by optical density techniques. Inhibitory effecton tested pathogens was observed for concentrations between 0.5% and 1% of crude extracts. There was a correlation between the antimicrobial activity of extracts and the use of nutrient-rich media in bacteria fermentation. Using a bioguided method, a peptidic compound, active against Fusarium oxysporum, was obtained from the 7ANT04 strain (Pyrobaculum sp.). Analysis by nuclear magnetic resonance and liquid chromatography coupled to mass detector evidenced an 11-amino acid compound. Bioinformatic software using raw mass data confirmed the presence of a cyclic peptide conformed by 11 mostly non-standard amino acids.

Biocontrol is an important aspect of disease management for plant pathogens, especially for the soil borne fungi. Trichoderma species is the most exploited biocontrol agent in recent years. The soil specific nature of Trichoderma species is a well-known fact and hence native Trichoderma isolates should be more emphasized for control of plant pathogens. Fifty soil samples from rhizosphere of various agricultural crops were collected for isolation of Trichoderma sp. Ten isolates of Trichoderma were tested in dual culture with soil borne pathogens Fusarium solani, Rhizoctonia solani and Sclerotinia sclerotiorum in an in vitro assay. All of the test isolates were found to be significant in terms of mycelial inhibition growth as compared to control. However, varying degrees of antagonism by different Trichoderma isolates were observed for above mentioned soil borne pathogens. The isolate (T363) was found to exhibit more than 80% inhibition of S. sclerotiorum while the isolate T357 was found to control F. solani by more than 80%.  For the control of R. solani, six of the tested Trichoderma isolates showed more than 80% inhibition of its radial colony growth. The Trichoderma isolates seen effective in this study need to be tested in pot and field experiments for exploiting the use and benefits of biocontrol.

Mechanism of parasitism of Trichoderma harzianum on Fusarium oxysporum on Acacia mangium seedlings. Fusarium oxysporum is one of the most important soil-borne fungi the causal agent of damping-off disease. Detailed information it needed to know how the pathogen can be inhibited by Trichoderma harzianum. The objective of this research was to investigate the inhibition mechanism of T. harzianum on F. oxysporum in vitro and in planta. Green Flourescent Protein (GFP) T. harzianum was used as biocontrol agent of F. oxysporum. An in vitro inhibition test of T. harzianum was performed using dual culture method. In the in planta inhibition tests, seedlings of A. mangium were applied with GFP T. harzianum two days before inoculation of F. oxysporum; GFP T. harzianum was simultaneously applied with F. oxysporum and GFP T. harzianum was applied two days after inoculation of F. oxysporum. The inhibition effect of T. harzianum GFP was observed at seven days incubation, indicated by attachment of T. harzianum to F. oxysporum hyphae. GFP T. harzianum hyphae covered the colonies of F. oxysporum at 12 days after incubation. The highest life percentage of A. mangium seedlings was found on the treatment of GFP T. harzianum two days before inoculation of F. oxysporum (82.22%), whereas the lowest life percentage was found on seedling applied with GFP T. harzianum two days after inoculation of F. oxysporum (64.44%).

Trichoderma species are well known for their biocontrol activity which colonize many soil and tuber-borne and foliage plant pathogens. In this study, 12 native isolates of Trichiderma spp were collected from various crop rhizosphere soil samples and characterized them phenotypically based on morphological and cultural features and genotypically based on sequence analysis of internal transcribed spacer (ITS) region-PCR amplification. The results obtained from phenotypic and genotypic observation revealed that isolates were belonged to five different species namely T. asperellum, T. harzianum, T. longibrachiatum, T. koningii and T. koningiopsis. All Trichoderma isolates produced ~600 bp amplicon and phylogenetic analysis revealed that all isolates were grouped with respective species. Further, the antagonistic potential of all the isolates was evaluated against Fusarium spp. following in vitro dual culture method. The results showed that isolates of T. harzianum exhibited maximum growth inhibition activity. The highest rate of inhibition was recorded with T. harzianum isolate TBT6 (87.1%) followed by TBT7 (82.2%), while the least inhibition was observed in T. longibrachiatum isolate TBT10 (59.7%) after 7 days of incubation. The antagonistic T. harzianum isolate TBT6 can be used for development of Trichoderma based bio-formulation and served as bio-control agent against Fusaium spp. under field conditions.

<p class="Abstract">Basal plate rot is a major disease on shallot caused by <em>Fusarium oxysporum</em>. Endophytic fungi is promising to use as antagonist agent to the pathogen. Endophyte is microbes that are living in plant cells and have an asymptomatic characteristic. Nowadays, fungal endophyte is believed to produce antimicrobial substances similar with their plant host's natural product. <em>Jatropha curcas</em> is one of the plants containing secondary metabolites that have antifungal activities. The research aimed to study the ability of endophyte from <em>Jatropha curcas</em> to inhibit the growth of <em>Fusarium oxysporum</em>. The dual culture method was used in this research and the data were analyzed by SPSS software. This antagonism test was conducted by 9 isolates endophyte and each plate consisted of 3 replicates. The result revealed endophyte fungal obtaining 9 isolates with the radial growth of 4,5 cm/2 days. Endophytes of <em>Jatropha curcas</em> L. were able to inhibit the growth of <em>Fusarium oxysporum</em> C2. The percentage of inhibition of <em>Fusarium oxysporum </em>causing of root blight diseases was controlled by up 38.27 - 74.48%. The highest percentage of inhibition is gained by B4b and the lowest of it is A2b. Our observations showed that each endophyte has a consistent linear trend. B4b still leaded as the highest strength to inhibit the growth of pathogen on the monitoring of 3, 5, and 7 days. Moreover, the ability of fungi endophyte from <em>Jatropha curcas</em> as antagonist agent to <em>Fusarium oxysporum</em> needs to be further examined by the in vivo method.</p>

Antraknosa yang disebabkan oleh Colletotrichum gloeosporioides merupakan penyakit pascapanen penting pada buahbuahan. Penelitian ini bertujuan untuk mengetahui efektifitas kitosan dan Trichoderma sp. dalam menghambat pertumbuhan koloni jamur C. gloeosporioides secara in vitro dan mengetahui efektifitas kitosan dan Trichoderma sp. terhadap intensitas penyakit antraknosa yang disebabkan oleh C. gloeosporioides pada buah pisang cavendish. Penelitian dilakukan di Laboratorium Penyakit Tumbuhan, Fakultas Pertanian, Universitas Lampung dari bulan Mei 2013 sampai dengan September 2013. Percobaan ini disusun dalam rancangan acak lengkap (RAL) yang terdiri atas lima perlakuan dan lima ulangan, yaitu kontrol (P0), kitosan (P1), Trichoderma sp. (P2), kombinasi kitosan dan Trichoderma sp. (P3) dan fungisida mankozeb (P4). C. gloeosporioides diperoleh dari isolasi buah pisang yang bergejala antraknosa. Pengujian secara in vitro, perlakuan kitosan konsentrasi 0,75% dicampurkan ke dalam media potato sukrose agar, perlakuan Trichoderma sp. (dual culture method), perlakuan kombinasi kitosan dan Trichoderma sp. dengan mencampurkan kitosan pada media dan dikombinasikan dengan Trichoderma sp. (dual culture method) serta perlakuan fungisida mankozeb konsentrasi 0,1% dicampurkan ke dalam media. Metode tersebut juga digunakan untuk pengujian secara in vivo pada buah pisang. Hasil pengujian secara in vitro, perlakuan kitosan, Trichoderma sp. dan kombinasi kitosan dan Trichoderma sp. efektif menghambat pertumbuhan koloni jamur C. gloeosporioides. Pada uji in vivo, masing-masing perlakuan tidak efektif menghambat intensitas penyakit antraknosa.

Antifungal activity of total saponins originated from roots of <i>Medicago hybrida</i> (Pourret) Trautv. were evaluated <i>in vitro</i> against six pathogenic fungi and eight individual major saponin glycosides were tested against one of the most susceptible fungi. The total saponins showed fungitoxic effect at all investigated concentrations (0.01%, 0.05% and 0.1%) but their potency was different for individual fungi. The highest saponin concentration (0.1%) was the most effective and the inhibition of <i>Fusarium oxysporum</i> f. sp. <i>callistephi</i>, <i>Botrytis cinerea</i>, <i>Botrytis tulipae</i>, <i>Phoma narcissi</i>, <i>Fusarium oxysporum</i> f. sp. <i>narcissi</i> was 84.4%, 69.9%, 68.6%, 57.2%, 55.0%, respectively. While <i>Fusarium oxysporum</i> Schlecht., a pathogen of <i>Muscari armeniacum</i>, was inhibited by 9.5% only. Eight major saponin glycosides isolated from the total saponins of <i>M. hybrida</i> roots were tested against the mycelium growth of <i>Botrytis tulipae</i>. The mycelium growth of the pathogen was greatly inhibited by hederagenin 3-O-<i>β</i>-D-glucopyranoside and medicagenic acid 3-O-<i>β</i>-D-glucopyranoside. Medicagenic acid 3-O-<i>β</i>-D-glucuronopyranosyl-28-O-<i>β</i>-D-glucopyranoside and oleanolic acid 3-O-[<i>β</i>-D-glucuronopyranosyl(1→2)-<i>α</i>-L-galactopyranosyl]-28-O-<i>β</i>-D-glucopyranoside showed low fungitoxic activity. Medicagenic acid 3-O-a-D-glucopyranosyl- 28-O-β-D-glucopyranoside, hederagenin 3-O-[α-L- hamnopyranosyl(1→2)-β-D-glucopyranosyl(1→2)-β-D-glucopyranosyl]- 28-O-α-D-glucopyranoside and hederagenin 3-O-<i>β</i>-D-glucuronopyranosyl-28-O-<i>β</i>-D- lucopyranoside did not limit or only slightly inhibited growth of the tested pathogen. While 2<i>β</i>, 3<i>β</i>-dihydroxyolean-12 ene-23-al-28-oic acid 3-O-<i>β</i>-D-glucuronopyranosyl-28-O-<i>β</i>-D-glucopyranoside slightly stimulated mycelium growth of <i>B. tulipae</i>.
