Komoditas budidaya Cherax semakin meningkat karena memiliki ketahanan tubuh tinggi, mudah dibudidayakan, dapat dikonsumsi dan dijadikan sebagai krustacea hias. Penelitian ini bertujuan untuk mengetahui pengaruh pakan berbeda terhadap pertumbuhan panjang dan bobot tubuh serta kelangsungan hidup lobster air tawar (Cherax quadricarinatus). Wadah penelitian menggunakan akuarium berukuran 60x40x40 cm yang dilengkapi peralatan aerasi. Metode penelitian menggunakan rancangan acak lengkap (RAL) dengan 3 perlakuan yaitu C1: Pakan Pellet, C2: Pakan Kelapa dan C3 : Pellet+Kelapa dan masing-masing 3 kali ulangan. Penelitian tersebut memberikan pengaruh terhadap pertumbuhan bobot dan panjang tubuh namun tidak berpengaruh terhadap kelangsungan hidup lobster air tawar. Pertumbuhan bobot tertinggi diperoleh pada perlakuan C3 yaitu sebesar 4.400 gram/ekor, panjang tertinggi pada perlakuan C2 yaitu sebesar 1.220 cm/ekor dan kelangsungan hidup tertinggi yaitu pada perlakuan C2 dan C3 yaitu masing-masing 86,67%.

Natural populations of crayfish are in depression in Russia and local species are not cultivated. In this situation, experimental cultivation of allochtonous australian crayfish Cherax quadricarinatus is conducted. This species is distinguished by high reproductive abilities and good consumer properties. It has domesticated in Russia spontaneously and produced 9–10 generations in Astrakhan Region. Certain natural selection in the process of domestication provides adaptive ability of this species to local environments and its capabil­ity to reproduce a viable progeny, so there is no doubts in good prospects of its cultivation in industrial conditions.

Carbonic anhydrase (CA), Na+/K+-ATPase (NKA) and Vacuolar-type H+-ATPase (HAT) play vital roles in osmoregulation and pH balance in decapod crustaceans. As variable pH levels have a significant impact on the physiology of crustaceans, it is crucial to understand the mechanisms by which an animal maintains its internal pH. We examined expression patterns of cytoplasmic (CAc) and membrane-associated form (CAg) of CA, NKA α subunit and HAT subunit a in gills of freshwater crayfish, Cherax quadricarinatus, at three pH levels – 6.2, 7.2 (control) and 8.2 – over 24 h. Expression levels of CAc were significantly increased at low pH and decreased at high pH conditions 24 h after transfer. Expression increased at low pH after 12 h, and reached its maximum level by 24 h. CAg showed a significant increase in expression at 6 h after transfer at low pH. Expression of NKA significantly increased at 6 h after transfer to pH 6.2 and remained elevated for up to 24 h. Expression for HAT and NKA showed similar patterns, where expression significantly increased 6 h after transfer to low pH and remained significantly elevated throughout the experiment. Overall, CAc, CAg, NKA and HAT gene expression is induced at low pH conditions in freshwater crayfish.

To explore the pathogenic mechanism of white spot syndrome virus (WSSV) in crayfish (Cherax quadricarinatus), we analysed activities of the three immune-related enzymes PO, SOD and LSZ in haemolymph tissue ofC. quadricarinatusbefore and after infection, and simultaneously studied the ultrastructural pathology. The results show that WSSV infection affects activities of the three enzymes. After 6-24 h of WSSV infection, the activities of PO, SOD and LSZ increased, but decreased significantly during longer infection times. The enzyme activities in WSSV-infected crayfish were significantly lower than those in controls at 72 h, except for LSZ (). Interestingly, the activities of PO, SOD and LSZ in the group treated with immune-polysaccharides before challenge with WSSV were higher than in the directly infected group, and the immune protection rate reached 51.9%, suggesting that the polysaccharides could improve enzyme activities and enhance antiviral defences of the organism. Ultrastructural pathological changes showed damaged haemolymph tissue, deformed golgiosomes, fuzzy damage in the mitochondrial structures, and nuclear membrane deformation and fracture. High levels of heterochromatin appeared in the nucleus; organoid and chromatin dissolved in dying blood cells, cytoplasm appeared oedematous and cells dissolved. WSSV particles were visible in blood cell nuclei of infected crayfish.

AbstractThe differentiation of the male reproductive system, the copulatory structures, "red patch" and onset of sexual maturity were studied in Cherax quadricarinatus. For this characterization 353 males were observed, dissected and processed for histological analysis. Three developmental stages were identified. At stage I the testes are two parallel cords with a few lobes or acinii, connected medially by a short thin bridge (X-shape) and the vasa deferentia are straight transparent cords. In the second stage, more lobulated testes present an H-shape and the vas deferens begins to differentiate into three distinct regions: proximal, medial and distal. In the last stage testes are two parallel cords tending to fuse along their entire length and become a single structure. From this stage, the development of the "red patch" occurs and spermatophore formation is observed in the vas deferens giving it white colour. These primary and secondary sexual characters could indicate the onset of sexual maturity. The morphology of the copulatory structure is lobular at stage I and it differentiates into an elongated structure during stage II. This study shows that in C. quadricarinatus the differentiation of primary and secondary sexual characters of males starts simultaneously and very early in development (0.09-0.10g achieved nearly 1 month old) and that acquisition of sexual maturity is also an early event (6g achieved nearly 3 month old). The compared features within Astacidea and Decapoda and the possible applications in the analysis of phylogenetic relationships between different taxa are discussed.
